Numerical Simulation of Micro Flow-Through Pcr Chip

Abstract:The miniaturization of clinical analysis devices using microfabrication has greatly impacted the medical and biological fields. The present work describes a two-dimensional model to numerically analyze the thermal cycling of the micro flow-through PCR chip, which differs from conventional PCR instruments and the micro chamber PCR chip. In the micro flow-through PCR chip, sample and reagent (PCR mixture) continuously flows through a microchannel in the chip with three different temperature regions (denaturation, annealing, and extension) to realize the nucleic acid amplification. Two parameters, U and D, are used to describe the temperature nonuniformity and the deviation. The numerical model was used to study the effects of the microchip's geometries, materials, heater temperatures for the three regions, sample and reagent flow rate, and the thermal boundary conditions of the microchip on the thermal cycling of the micro flow-through PCR chip. The results show that the silicon-glass–bonded micro PCR chip provides the best design and the heater temperatures in the three regions should be optimized in advance. The results also illustrate the applicability of a silicon-glass–bonded micro flow-through PCR chip with only the denaturation region heated.
