Abstract:<p>Injectable therapeutics enabled by engineered biomaterials are becoming increasingly popular, transforming traditional clinical practice to become a minimally invasive and regenerative regime. Compared to preformed biomaterials, injectable biomaterials allow for more precise implantation into deeply enclosed anatomical locations and for the repair of irregularly shaped lesions, demonstrating great translational potential. Continuously emerging clinical needs and advances in materials science have driven an evolution in injectable biomaterials from structural fillers to multifunctional platforms. Integrating disparate functions to design injectable biomaterials for clinical translation remains a considerable challenge, as does the selection of the appropriate design considerations for specific applications. This article aims to review the design and fabrication considerations of injectable biomaterials in the context of medical translation, the engineering strategies used for new materials to meet the growing demands in regenerative and intelligent medicine, and the progress in their development for selected clinical applications. Specifically, three exemplary areas, injectable bone cements, hydrogels, and electronics, all of which demonstrate significant promise in terms of translation and commercialization, are reviewed in detail. In addition, their translational status and future challenges are discussed. It is also envisioned that the mutual collaboration between researchers, clinicians, entrepreneurs, engineers, and patients will inspire and catalyze the innovation and translation of injectable biomaterials.</p>

Abstract:Biodegradable polymers have been widely used and have greatly promoted the development of biomedical fields because of their biocompatibility and biodegradability. The development of biotechnology and medical technology has set higher requirements for biomedical materials. Novel biodegradable polymers with specific properties are in great demand. Biodegradable polymers can be classified as natural or synthetic polymers according to the source. Synthetic biodegradable polymers have found more versatile and diverse biomedical applications owing to their tailorable designs or modifications. This review presents a comprehensive introduction to various types of synthetic biodegradable polymers with reactive groups and bioactive groups, and further describes their structure, preparation procedures and properties. The focus is on advances in the past decade in functionalization and responsive strategies of biodegradable polymers and their biomedical applications. The possible future developments of the materials are also discussed.

Abstract:Biomedical polymers have been extensively developed for promising applications in a lot of biomedical fields, such as therapeutic medicine delivery, disease detection and diagnosis, biosensing, regenerative medicine, and disease treatment. In this review, we summarize the most recent advances in the synthesis and application of biomedical polymers, and discuss the comprehensive understanding of their property-function relationship for corresponding biomedical applications. In particular, a few burgeoning bioactive polymers, such as peptide/biomembrane/microorganism/cell-based biomedical polymers, are also introduced and highlighted as the emerging biomaterials for cancer precision therapy. Furthermore, the foreseeable challenges and outlook of the development of more efficient, healthier and safer biomedical polymers are discussed. We wish this systemic and comprehensive review on highlighting frontier progress of biomedical polymers could inspire and promote new breakthrough in fundamental research and clinical translation.

Abstract:This is a provisional file, not the final typeset article Rational design for advanced nanostructures or is a promising solution for improving 33 the therapeutic efficacy. Nanostructures significant advantages they may overcome long-term 34 issues such as poor drug solubility, drug metabolism, systemic half-life, and undesirable side effects 35 (Yu et al., 2023). However, it still remained the challenge how to enhance the permeability and 36 retention in the tumor stroma. During the recent decades, researchers developed numerous kinds of 37 nanostructures with different size, shapes, or even charge distributions to enhance the accumulation of 38 therapeutic agents in tumor sites (Sun et al., 2017). Not only that, strategies like surface modification 39 with specific ligands, camouflage with cell membrane, charge or size reversal triggered by tumor-40 microenvironment (TME), etc. were also employed to improve their clinical potential. 41 However, there is a significant need for additional validation of the biological composition and activity 42 of these biomimetic nanostructures to ensure their real properties in vivo.
