Responsive nanoplatforms: Versatile design strategies for efficient cancer theranostics

Abstract:In recent years, with close cooperation between different disciplines, more and more smart "all-in-one" theranostic nanoplatforms with stimulus-responsive capability have been designed for biomedical applications, attracting much attention around the world. In this context, the stimuli-responsive components play a decisive role in the design of theranostic nanoagents with in vivo spatiotemporal mode. In this review, we outline some typical design strategies of responsive nanoplatforms for efficient cancer therapy from both material and biological perspectives. From the materials point of view, researchers have designed the smart nanoplatforms by functionalized modifications or doping strategy combined with adjustable or variable skeleton structure to achieve promising biomedical applications. From the biological point of view, the constructed nanoplatforms targeting the different physiological factors that characterize specific diseases are hopeful to address the challenges posed by the pathological environment in a specifical and efficient way, in order to achieve an individualized therapy. Finally, we prospect the present limitations and future development of responsive smart biomaterials.

Abstract:Abstract Until now, enormous smart materials have been engineered with endogenous stimulators such as pH, reactive oxygen species, glutathione, hypoxia and enzyme, or exogenous stimulators such as temperature, light, ultrasound, radiation, and magnetic field in drug delivery. As footstone of stimuli‐responsive nanocarriers, endogenous/exogenous responsive smart materials possess many properties, such as responding ability to specific triggers, controlled drug release, long blood circulation, increased tumor accumulation, “ON‐OFF” switch activities, enhanced diagnostic accuracy, and therapeutic efficacy. Smart materials have attracted considerable attention because they provide likelihood strategy for individualized and comprehensive therapy. In this review, significant research achievements of smart materials responsive to different triggers including their synthesis and formulation mechanism, responsive mechanism, applications, multiple functions are summarized and discussed separately. We primarily focus on the studies in the past few years (2017‐2020). The current situation and remaining challenges of stimuli‐sensitive materials‐based nanocarriers for clinical translation are discussed rationally at the end. It is hope that this timely and overall review would provide some helpful information for researchers in this field.

Abstract:Cancer therapy is unsatisfactory as it typically has serious side effects, because normal cells in healthy organs are destroyed along with the tumor. Thus, researchers have tried to develop effective therapies with minimal side effects. One such method is to use nanotechnology to carry the drugs or therapeutic agents to the tumor region by secure encapsulation without leakage. Once the nanomedicine enters the target tumor site, it can release therapeutic agents in an effective manner. Accordingly, various nanomedicines have been developed to enhance the efficiency of cancer therapy and minimize the systematic toxicity. Here, we provide an overview and discuss the different types of responsive nanomedicines including physically, chemically, biologically, dual, and multi-responsive nanomedicines, for the in situ release of cargos in recent years. We propose critical considerations that must be considered for the design of excellent stimuli-nanomedicine. Furthermore, the possible directions for the development of successful stimuli-responsive (smart) nanomedicine are highlighted. With the development of responsive nanomedicines, precise and personalized nanomedicine will be realized with great promise in the future. Abstract:Conventional drugs used for cancer chemotherapy have severe toxic side effects and show individually varied therapeutic responses. The convergence of nanotechnology, biology, material science and pharmacy offers a perspective strategy for cancer chemotherapy. Nanoparticles loaded with anti-cancer drug have been designed to overcome the limitations associated with conventional drugs, several nanomedicines have been approved by FDA and shown good performances in clinical practice. However, the therapeutic efficacies cannot be enhanced. Taking this into account, stimuli responsive nanoparticles present the ability to enhance therapeutic efficacy and reduce side effects. In this review, we systematically summarized the recent progresses of controlled anti-cancer drug release systems based on nanoparticles with different stimuli response including pH, temperature, light, redox and others. If the achievements of the past can be extrapolated into the future, it is highly likely that responsive nanoparticles with a wide array of desirable properties can be eventually developed for safe and efficient cancer therapy.

Abstract:CONSPECTUS: Cancer is a disease characterized by uncontrolled growth and division of abnormal cells, which could be effectively suppressed by various therapeutic agents in clinics. With the increasing knowledge of cancer, the rapid development of different therapeutic agents, such as small molecule cytotoxic drugs, nucleic acids, proteins/peptides, immunomodulators, and so forth, has expanded the modalities of cancer therapy. However, their clinical application is still hampered by the low tumor specificity when applied in vivo. The irrational distribution in healthy tissues may cause severe side effects and reduce the therapeutic effect. Undoubtedly, achieving the on-demand release of therapeutic agents at the tumor site is critical for effective anticancer efficacies with reduced side effects. Benefiting from the development of nanotechnology, nanomaterials are widely exploited to significantly boost the specificity of drugs to the tumor tissue and simultaneously decrease the toxic effects on the normal tissues, leading to improved therapeutic outcomes. Nevertheless, conventional nanomaterials still have multiple drawbacks, such as poor tumor targeting and unavoidable leakage of therapeutic agents. In this regard, the self-adaptive nanomaterials, which respond to the signal changes emitted from the tumor site, might realize the spatiotemporally and quantitatively specific release of drugs. Self-adaptive nanomaterials exhibited capabilities of self-regulation and self-feedback, whose properties, such as charge, size, and shape, underwent an on-demand transformation in response to specific stimuli. Compared to the conventional ones, self-adaptive nanomaterials successfully decrease the frequency of drug release doses within normal tissues and maintain drug concentrations in the tumor cells for a more extended period, thus promoting rational This Account highlights recent developments in the field of self-adaptive nanomaterials used for rational drug delivery in cancer therapy, primarily based on progress made by our group. We discuss self-adaptive nanomaterials responding to endogenous and exogenous stimuli to trigger the release of therapeutic agents in the therapeutic targets of cancer. Chemical approaches used in the construction of self-adaptive nanomaterials and representative examples of recently developed nanomaterials are reviewed, and multistimuli-responsive nanomaterials, addressing the shortcomings of single stimulus-responsive nanomaterials, are also discussed. In addition, we provide a summary of ongoing clinical trials involving self-adaptive nanomaterials in cancer therapy. Finally, we provide an overview of the issues encountered during the transition of these materials from research settings to clinical use. The exploration and optimization of nanomaterial chemical design is an ongoing process. It is hoped that the insights provided in this Account will be a valuable reference for creating the next generation of these promising self-adaptive nanomaterials.

Abstract:This is a provisional file, not the final typeset article Rational design for advanced nanostructures or is a promising solution for improving 33 the therapeutic efficacy. Nanostructures significant advantages they may overcome long-term 34 issues such as poor drug solubility, drug metabolism, systemic half-life, and undesirable side effects 35 (Yu et al., 2023). However, it still remained the challenge how to enhance the permeability and 36 retention in the tumor stroma. During the recent decades, researchers developed numerous kinds of 37 nanostructures with different size, shapes, or even charge distributions to enhance the accumulation of 38 therapeutic agents in tumor sites (Sun et al., 2017). Not only that, strategies like surface modification 39 with specific ligands, camouflage with cell membrane, charge or size reversal triggered by tumor-40 microenvironment (TME), etc. were also employed to improve their clinical potential. 41 However, there is a significant need for additional validation of the biological composition and activity 42 of these biomimetic nanostructures to ensure their real properties in vivo.

Abstract:Cancer is a serious health problem which increasingly causes morbidity and mortality worldwide. It causes abnormal and uncontrolled cell division. Traditional cancer treatments include surgery, chemotherapy, radiotherapy and so on. These traditional therapies suffer from high toxicity and arouse safety concern in normal area and have difficulty in accurately targeting tumour. Recently, a variety of nanomaterials could be used for cancer diagnosis and therapy. Nanomaterials have several advantages, e.g. , high concentration in tumour via targeting design, reduced toxicity in normal area and controlled drug release after various rational designs. They can combine with many types of biomaterials in order to improve biocompatibility. In this review, we outlined the latest research on the use of bioresponsive nanomaterials for various cancer imaging modalities (magnetic resonance imaging, positron emission tomography and phototacoustic imaging) and imaging-guided therapy means (chemotherapy, radiotherapy, photothermal therapy and photodynamic therapy), followed by discussing the challenges and future perspectives of this bioresponsive nanomaterials in biomedicine.

Abstract:Cancer immunotherapy has been demonstrated as a promising therapeutic strategy in clinic owing to its unique advantages. However, although more and more immunotherapeutic agents have been approved for clinical use to activate the immune system, they also could interfere with the homeostatic role of immune system at non-target sites after systemic administration, which may be associated with fatal side effects such as lifelong autoimmune diseases. Thus, it is desirable to develop local delivery systems that could be applied at the targeted sides and engineered to locally control the pharmacokinetics of various immunotherapeutics, including small molecules, macromolecules or even cells. Advancements in biomaterials, biotechnology, nanomedicine and engineering have facilitated the development of local delivery systems for enhanced cancer immunotherapy. This review will summarize the recent advances in developing different local delivery systems and discuss how these delivery systems could be designed to regulate the release behavior of different immunotherapeutics to sustainably stimulate the systemic immune system, effectively and safely inhibiting the cancer recurrence and metastasis. Furthermore, we will discuss how biomaterials-assisted local delivery systems would contribute to the development of cancer immunotherapy, together with their challenges and potential of clinical translation.

Abstract:Cancer vaccines aimed at expanding the pool or increasing the activity of tumor-specific T cells against malignancies is an important immunotherapy modality that has been extensively pursued in the past decades. However, the clinical efficacy of cancer vaccines remains modest in comparison to other immunotherapies, such as checkpoint blockade and adoptive T cell therapy. This unsatisfactory performance is likely due to the suboptimal selection of tumor antigens for vaccine and inefficient delivery platform. Recently, vaccines designed to target cancer neoantigens have shown marked promise in both preclinical and early clinical studies. However, enormous challenges need to be overcome to develop a highly efficient and safe delivery strategy for targeting cancer vaccines to professional antigen-presenting cells and eliciting optimized immune response against cancers. To meet these challenges, biomaterials, particularly biomaterials that are designed to respond to certain environmental stimuli, termed as stimuli-responsive biomaterials, are being actively developed to precisely manipulate the trafficking and release of cancer vaccines in vivo for enhanced therapeutic efficacy and safety. In this mini review, we provide a brief overview of the recent advances in applying stimuli-responsive biomaterials in enhancing non-cellular cancer vaccines while focusing on the chemistry and material design with varied responsiveness. We also discuss the present challenges and opportunities in the field and provide a perspective for future directions.

Abstract:Spurred by advances in materials chemistry, molecular pharmaceutics and micro/nanobiotechnology, stimuli-responsive “smart” materials and systems have been studied extensively for various applications, including drug delivery, diagnosis, tissue engineering, and biomedical devices. In drug delivery, the dosage-, spatial- and/or temporal- controlled release of therapeutics significantly enhances the treatment efficacy in a precise manner. The development of stimuli-responsive materials also allows noninvasive or minimally invasive monitoring in real-time for achieving next-generation diagnostics. In addition, the smart systems with the capability of communicating and interacting with cells are highly desirable for engineering regenerative medications and biomedical devices. This theme issue focuses on the responsive materials and systems for a range of biomedical applications, with a collection of ten relevant research or review papers. For diabetes treatment, Mitragotri and coworkers1 describe a novel mucoadhesive intestinal device entrapped in a capsule with a pH-responsive enteric coating for oral delivery of insulin. This device loaded with insulin can effectively decrease blood glucose levels of diabetic rats. Gu and coworkers2 summarize a variety of stimuli-responsive delivery systems for diabetes treatment, including the pH-sensitive materials for oral delivery, the glucose-responsive delivery systems, and the on-demand delivery approaches by external ultrasound or light. For cancer therapy, Tong and Feng3 discuss general principles in polymer-drug conjugate design such as the synthetic strategies, the choice of the responsive linkers between the drug and polymer, and the in vivo delivery barriers. Liu and Liang4 review reactive oxygen species (ROS)-responsive system as well as multiple stimuli systems for enhanced delivery of therapeutics. For tissue engineering, Hammond and collaborators5 develop nanoscale polyelectrolyte complexes to deliver insulin-like growth factor-1 (IGF-1) for cartilage repair. They demonstrate IGF-1 is released by nanocomplexes within the joint space over four weeks, protecting cartilage from degradation and mitigating joint inflammation. Remote utilization of physical triggers, such as light, magnetic force and temperature to achieve spatiotemporal activation is an emerging research topic in this field. Lovell and Miranda6 discuss different mechanisms of the light-induced liposome permeabilization for controlled drug delivery, including light-induced oxidation, photocrosslinking, photoisomerization, photocleavage, and photothermal release. Xu and coworkers7 describe the development of near-infrared light-responsive liposomes for enhanced gene transfection through the photothermal effect. Gaharwar and coworkers8 review the applications of smart hydrogels based on magnetic nanoparticles and thermoresponsive polymers in therapeutic drug delivery, bioimaging, and regenerative medicine. In the remainder of this issue, Webber9 focuses on the preparation of responsive self-assembled materials for biomedical applications, including engineering therapeutics and devices for biological sensing and disease diagnostics; Cui and coworkers10 review peptide-based supramolecular nanostructures and hydrogels for biological stimuli-triggered delivery of biologics. Collectively, this issue highlights the exciting research work and key advances in leveraging responsive materials for smart and precision medications, the clinical translation of which would revolutionize health care, profoundly enhancing patients’ health and improving their quality of life. Abstract:Bio-based nanomaterials have received considerable attention in cancer therapies due to their potent characteristics such as autologous pharmaceutical, synergetic effects, biocompatibility, biodegradability and biosafety. Here, we took breast cancer (BC) for example, mainly focused on six types of bioactive substances (nucleic acid, phenolics, polysaccharides, protein and peptide, lipid and cell) and their applications in biobased nano-formulation for oncological therapies. Particularly, we have categorized the functions of these bioactive substances and exemplified the nano-systems' composite constructions, structural characterizations, versatile properties, anti-tumor mechanisms and biological performances. Ultimately, we thoroughly discussed the present challenges and future perspectives in the emerging research frontier. We believe that this review would offer inspired ideas, promising strategies and potential pathways for designing state-ofthe-bio-based-nanomaterials for cancer therapy in critical and advanced biomedical sciences. Abstract:A bioresponsive polymeric nanocarrier for drug delivery is able to alter its physical and physicochemical properties in response to a variety of biological signals and pathological changes,and can exert its therapeutic efficacy within a confined space.These nanosystems can optimize the biodistribution and subcellular location of therapeutics by exploiting the differences in biochemical properties between tumors and normal tissues.Moreover,bioresponsive polymer-based nanosystems could be rationally designed as precision therapeutic platforms by optimizing the combination of responsive elements and therapeutic components according to the patient-specific disease type and stage.In this review,recent advances in smart bioresponsive polymeric nanosystems for cancer chemotherapy and immunotherapy will be summarized.We mainly discuss three categories,including acidity-sensitive,redox-responsive,and enzyme-triggered polymeric nanosystems.The important issues regarding clinical translation such as reproducibility,manufacture,and probable toxicity,are also commented.

Abstract:Proteins are an important class of biomaterials promising a variety of applications such as drug delivery, and imaging or therapy, owing to their biodegradability, biocompatibility, as well as inherent biological activities acting as enzymes, recognizing molecules, or therapeutics by themselves. Over the few past decades, different types of proteins with desired properties have been widely explored for biomedical applications. Many therapeutic proteins have now entered clinical use. This review therefore summarizes various strategies in the engineering of biomaterials for delivery of therapeutic proteins, as well as the recent development of protein‐based biomaterials for cancer theranostics.
The Smart Dual-Stimuli Responsive Nanoparticles for Controlled Anti-Tumor Drug Release and Cancer Therapy

Abstract:Background: In recent years, the emergence of stimuli-responsive nanoparticles has made drug delivery more efficient. As an intelligent and effective targeted delivery platform, it can reduce the side effects generated during drug transportation while enhancing the treatment efficacy. The stimuli-responsive nanoparticles can respond to different stimuli at corresponding times and locations to deliver and release their drugs and associated therapeutic effects. Objective: This review aims to inform researchers on the latest advances in the application of dual-stimuli responsive nanoparticles in precise drug delivery, with special attention to their design, drug release properties, and therapeutic effects. Syntheses of nanoparticles with simultaneous or sequential responses to two or more stimuli (pH-redox, pH-light, redox-light, temperature-magnetic, pH-redox-temperature, redox-enzyme-light, etc.) and the applications of such responsivity properties for drugs control and release have become a hot topic of recent research. Methods: A database of relevant information for the production of this review was sourced, screened and analyzed from Pubmed, Web of Science, SciFinder by searching for the following keywords: “dual-stimuli responsive”, “controlled release”, “cancer therapy”, “synergistic treatment”. Results: Notably, the nanoparticles with dual-stimuli responsive function have an excellent control effect on drug delivery and release, playing a crucial part in the treatment of tumors. They can improve the encapsulation and delivery efficiency of hydrophobic chemotherapy drugs, combine chemo-photothermal therapies, apply imaging function in the diagnosis of tumors and even conduct multi-drug delivery to overcome Multi-Drug Resistance (MDR).
