Abstract:This is a provisional file, not the final typeset article Rational design for advanced nanostructures or is a promising solution for improving 33 the therapeutic efficacy. Nanostructures significant advantages they may overcome long-term 34 issues such as poor drug solubility, drug metabolism, systemic half-life, and undesirable side effects 35 (Yu et al., 2023). However, it still remained the challenge how to enhance the permeability and 36 retention in the tumor stroma. During the recent decades, researchers developed numerous kinds of 37 nanostructures with different size, shapes, or even charge distributions to enhance the accumulation of 38 therapeutic agents in tumor sites (Sun et al., 2017). Not only that, strategies like surface modification 39 with specific ligands, camouflage with cell membrane, charge or size reversal triggered by tumor-40 microenvironment (TME), etc. were also employed to improve their clinical potential. 41 However, there is a significant need for additional validation of the biological composition and activity 42 of these biomimetic nanostructures to ensure their real properties in vivo.
