Abstract:This is a provisional file, not the final typeset article Rational design for advanced nanostructures or is a promising solution for improving 33 the therapeutic efficacy. Nanostructures significant advantages they may overcome long-term 34 issues such as poor drug solubility, drug metabolism, systemic half-life, and undesirable side effects 35 (Yu et al., 2023). However, it still remained the challenge how to enhance the permeability and 36 retention in the tumor stroma. During the recent decades, researchers developed numerous kinds of 37 nanostructures with different size, shapes, or even charge distributions to enhance the accumulation of 38 therapeutic agents in tumor sites (Sun et al., 2017). Not only that, strategies like surface modification 39 with specific ligands, camouflage with cell membrane, charge or size reversal triggered by tumor-40 microenvironment (TME), etc. were also employed to improve their clinical potential. 41 However, there is a significant need for additional validation of the biological composition and activity 42 of these biomimetic nanostructures to ensure their real properties in vivo.

Abstract:Nanomaterials, that is, materials made up of individual units between 1 and 100 nanometers, have lately involved a lot of attention since they offer a lot of potential in many fields, including pharmacy and biomedicine, owed to their exceptional physicochemical properties arising from their high surface area and nanoscale size. Smart engineering of nanostructures through appropriate surface or bulk functionalization endows them with multifunctional capabilities, opening up new possibilities in the biomedical field such as biosensing, drug delivery, imaging, medical implants, cancer treatment and tissue engineering. This article highlights up‐to‐date research in nanomaterials functionalization for biomedical applications. A summary of the different types of nanomaterials and the surface functionalization strategies is provided. Besides, the use of nanomaterials in diagnostic imaging, drug/gene delivery, regenerative medicine, cancer treatment and medical implants is reviewed. Finally, conclusions and future perspectives are provided.

Abstract:Nanoscale materials have been explored extensively as agents for therapeutic and diagnostic (i.e. theranostic) applications. Research efforts have shifted from exploring new materials in vitro to designing materials that function in more relevant animal disease models, thereby increasing potential for clinical translation. Current interests include non-invasive imaging of diseases, biomarkers and targeted delivery of therapeutic drugs. Here, we discuss some general design considerations of advanced theranostic materials and challenges of their use, from both diagnostic and therapeutic perspectives. Common classes of nanoscale biomaterials, including magnetic nanoparticles, quantum dots, upconversion nanoparticles, mesoporous silica nanoparticles, carbon-based nanoparticles and organic dye-based nanoparticles, have demonstrated potential for both diagnosis and therapy. Variations such as size control and surface modifications can modulate biocompatibility and interactions with target tissues. The needs for improved disease detection and enhanced chemotherapeutic treatments, together with realistic considerations for clinically translatable nanomaterials will be key driving factors for theranostic agent research in the near future.

Abstract:Cardiovascular diseases as a primary driver of global morbidity and mortality. Despite the array of therapeutic avenues in clinical practice, predominantly pharmaceutical and surgical interventions, they often fall short of fully addressing the clinical exigencies of cardiovascular patients. In recent years, nanocarriers have shown great potential in the treatment and diagnose of cardiovascular diseases. They can enhance drug targeting and bioavailability while reducing side effects. Additionally, by improving imaging and detection technologies, they enhance early diagnosis and disease monitoring capabilities. These advancements in technology offer new solutions for precision medicine in cardiovascular diseases, advancing treatment efficacy and disease management. Crafted from biomaterials, metals, or their amalgamations, these nanocarriers approximate the dimensions of biologically active molecules like proteins and DNA. Cardiovascular nanomedicine, in its infancy, has only recently burgeoned. Yet, with continual refinement in nanocarrier architecture, drug delivery mechanisms, and therapeutic outcomes, the potential of nanomedical technologies in clinical contexts becomes increasingly evident. This review aims to consolidate the strides made in nanocarrier research concerning the treatment and diagnose of cardiovascular diseases.

Abstract:In the past decades, various nanomaterials with unique properties have been explored for bioapplications. Meanwhile, aptamers, generated from the systematic evolution of ligands by exponential enrichment technology, are becoming an indispensable element in the design of functional nanomaterials because of their small size, high stability, and convenient modification, especially endowing nanomaterials with recognition capability to specific targets. Therefore, the incorporation of aptamers into nanomaterials offers an unprecedented opportunity in the research fields of diagnostics and therapeutics. Here, we focus on recent advances in aptamer-embedded nanomaterials for bioapplications. First, we briefly introduce the properties of nanomaterials that can be functionalized with aptamers. Then, the applications of aptamer-embedded nanomaterials in cellular analysis, imaging, targeted drug delivery, gene editing, and cancer diagnosis/therapy are discussed. Finally, we provide some perspectives on the challenges and opportunities that have arisen from this promising area.

Abstract:Functional nanomaterials have been widely used in biomedical fields due to their good biocompatibility, excellent physicochemical properties, easy surface modification, and easy regulation of size and morphology. Functional nanomaterials for magnetic resonance imaging (MRI) can target specific sites in vivo and more easily detect disease-related specific biomarkers at the molecular and cellular levels than traditional contrast agents, achieving a broad application prospect in MRI. This review focuses on the basic principles of MRI, the classification, synthesis and surface modification methods of contrast agents, and their clinical applications to provide guidance for designing novel contrast agents and optimizing the contrast effect. Furthermore, the latest biomedical advances of functional nanomaterials in medical diagnosis and disease detection, disease treatment, the combination of diagnosis and treatment (theranostics), multi-model imaging and nanozyme are also summarized and discussed. Finally, the bright application prospects of functional nanomaterials in biomedicine are emphasized and the urgent need to achieve significant breakthroughs in the industrial transformation and the clinical translation is proposed. Abstract:Background: In recent years, nanotechnology-based biomedical platforms aid characteristic dimensions in the field of biomedicine as compared to the conventional strategies. This nanotechnological intervention carries widespread biomedical avenues in the diagnosis and treatment of an array of dreadful diseases. Objective: Silver nanoparticles (AgNPs) have the inherent property of immense biological potential such as antimicrobial and anticancer activity. AgNPs also show tremendous optical and electrical properties suggesting its candidature as promising bio-sensing agent thereby promoting environmental and biological management. The use of AgNPs as contrast agents could be exploited for enhanced bio-imaging of cellular and sub-cellular organelles giving insight into the mechanism of action of the administered drugs. More importantly, functionalized AgNPs have been driven a new wave of innovation in developing targeted drug delivery and theranostics. In this article, we summarize the current status and progress of functionalized AgNPs-based nanoplatform for biomedical applications with an emphasis on sensing, molecular imaging, and theranostics. Main Text: The present review focuses on the importance of functionalized AgNPs towards enhanced biosensing, bio-imaging and theranostics applications as compared to the conventional modalities. It also clarifies the importance of functionalization to increase the efficacy of bio-sensing, bio-imaging and theranostic modalities. Future Perspectives: The present review not only emphasizes the biomedical importance of functionalized AgNPs but also scrutinizes its widespread avenues in the near future for the transformation into clinical translation.
