Abstract:Cell therapy has achieved tremendous success in regenerative medicine in the past several decades. However, challenges such as cell loss, death and immune-rejection after transplantation still persist. Biomaterials have been designed as carriers to deliver cells to desirable region for local tissue regeneration; as barriers to protect transplanted cells from host immune attack; or as reactors to stimulate host cell recruitment, homing and differentiation. With the assistance of biomaterials, improvement in treatment efficiency has been demonstrated in numerous animal models of degenerative diseases compared with routine free cell-based therapy. Emerging clinical applications of biomaterial assisted cell therapies further highlight their great promise in regenerative therapy and even cure for complex diseases, which have been failed to realize by conventional therapeutic approaches.

Abstract:Nanomaterial-based cell sheet technology has been reported to be an effective method in regenerative medicine and tissue engineering. Here, we summarized several types of nanomaterials used to harvest cell sheets. Currently, the technology is divided into four categories according to the mechanisms: light-induced cell sheet technology, thermo-responsive cell sheet technology, magnetic-controlled cell sheet technology, and reactive oxygen species (ROS)-induced cell sheet technology. Furthermore, some studies have been conducted to show that nanomaterial-based cell sheets produce satisfying outcomes in the regeneration of bone, skeletal muscle, cardiac tissue, and tendon, as well as angiogenesis and osseointegration. Nevertheless, some shortcomings still exist, such as comprehensive preparation, unclear safety, and cell quality. Thus, future studies should aim to produce more types of nanomaterials to solve this problem.

Abstract:The dynamic roles of T cells in the immune system to recognize and destroy the infected or mutated cells render T cell therapy a prospective treatment for a variety of diseases including cancer, autoimmune diseases, and allograft rejection. However, the clinical applications of T cell therapy remain unsatisfactory due to the tedious manufacturing process, off-target cytotoxicity, poor cell persistence, and associated adverse effects. To this end, various biomaterials have been introduced to enhance T cell therapy by facilitating proliferation, enhancing local enrichment, prolonging retention, and alleviating side effects. This review highlights the design strategies of biomaterials developed for T cell expansion, enrichment, and delivery as well as their corresponding therapeutic effects. The prospects of biomaterials for enhancing T cell immunotherapy are also discussed in this review.

Abstract:Stem-cell-based therapies have the potential to provide novel solutions for the treatment of a variety of diseases, but the main obstacles to such therapies lie in the uncontrolled differentiation and functional engraftment of implanted tissues. The physicochemical microenvironment controls the self-renewal and differentiation of stem cells, and the key step in mimicking the stem cell microenvironment is to construct a more physiologically relevant 3D culture system. Material-based 3D assemblies of stem cells facilitate the cellular interactions that promote morphogenesis and tissue organization in a similar manner to that which occurs during embryogenesis. Both natural and artificial materials can be used to create 3D scaffolds, and synthetic organic and inorganic porous materials are the two main kinds of artificial materials. Nanotechnology provides new opportunities to design novel advanced materials with special physicochemical properties for 3D stem cell culture and transplantation. Herein, the advances and advantages of 3D scaffold materials, especially with respect to stem-cell-based therapies, are first outlined. Second, the stem cell biology in 3D scaffold materials is reviewed. Third, the progress and basic principles of developing 3D scaffold materials for clinical applications in tissue engineering and regenerative medicine are reviewed.

Abstract:Biotherapy mainly refers to the intervention and the treatment of major diseases with biotechnologies or bio-drugs, which include gene therapy, immunotherapy (vaccines and antibodies), bone marrow transplantation and stem-cell therapy. In recent years, numerous biomaterials have emerged and were utilized in the field of biotherapy due to their biocompatibility and biodegradability. Generally, biomaterials can be classified into natural or synthetic polymers according to their source, both of which have attracted much attention. Notably, biomaterials-based non-viral gene delivery vectors in gene therapy are undergoing rapid development with the emergence of surface-modified or functionalized materials. In immunotherapy, biomaterials appear to be attractive means for enhancing the delivery efficacy and the potency of vaccines. Additionally, hydrogels and scaffolds are ideal candidates in stem-cell therapy and tissue engineering. In this review, we present an introduction of biomaterials used in above biotherapy, including gene therapy, immunotherapy, stem-cell therapy and tissue engineering. We also highlighted the biomaterials which have already entered the clinical evaluation.
The role of biomaterials in stem cell-based regenerative medicine

Abstract:Stem cell therapy is a promising alternative approach to the treatment of a number of incurable degenerative diseases. However, low cell retention and survival after transplantation limit the therapeutic efficacy of stem cells for clinical translational applications. The utilization of biomaterials has been progressively successful in controlling the fate of transplanted cells by imitating the cellular microenvironment for optimal tissue repair and regeneration. This review mainly focuses on the engineered microenvironments with synthetic biomaterials in modification of stem cell behaviors. Moreover, the possible advancements in translational therapy by using biomaterials with stem cells are prospected and the challenges of the current restriction in clinical applications are highlighted.

Abstract:Stem cell-based regenerative therapies, which harness the self-renewal and differentiation properties of stem cells, have been in the spotlight due to their widespread applications in treating degenerative, aging, and other, generally intractable diseases. Therapeutically effective hematopoietic stem cells, mesenchymal stem cells, embryonic stem cells, and induced pluripotent stem cells have been used in numerous basic and translational studies with exciting results. However, pre-/post-transplantation issues of poor cell survival and retention, uncontrolled differentiation, and insufficient numbers of cells engrafted into host tissues are the major challenges in stem cell-based regenerative therapies. Engineered biomaterials have adjustable biochemical and biophysical properties that significantly affect cell behaviors, such as cell engraftment, survival, migration, and differentiation outcomes, thereby enhancing the engraftment of implanted stem cells and guiding tissue regeneration. Therefore, the combination of stem cell biology with bioengineered materials is a promising strategy to improve the therapeutic outcomes of stem cell-based regenerative therapy. In this review, we summarize the advances in the modulation of behaviors of stem cells via engineered biomaterials. We then present different approaches to harnessing bioengineered materials to enhance the transplantation of stem cells. Finally, we will provide future directions in regenerative therapy using stem cells.

Abstract:Cell therapy, the treatment of diseases using living cells, offers a promising clinical approach to treating refractory diseases. The global market for cell therapy is growing rapidly, and there is an increasing demand for automated methods that can produce large quantities of high quality therapeutic cells. Biomaterials can be used during cell production to establish a biomimetic microenvironment that promotes cell adhesion and proliferation while maintaining target cell genotype and phenotype. Here we review recent progress and emerging techniques in biomaterial-assisted cell production. The increasing use of auxiliary biomaterials and automated production methods provides an opportunity to improve quality control and increase production efficiency using standardized GMP-compliant procedures.

Abstract:Cell transplantation holds immense potential for reversing diseases that are currently incurable and for regenerating tissues. However, poor cell survival, cell aggregation and lack of cell integration into the host tissue constitute major challenges for the clinical translation of cell transplantation approaches. Biomaterials can influence cell behaviour in vitro and in vivo. The mechanical and biochemical properties of biomaterials can be tailored to affect cell survival, differentiation and migration. Therefore, the integration of advanced material design with stem cell biology may hold the key to improving the efficacy of cell transplantation. In this Review, we discuss biomaterial design strategies for their potential to influence the fate of transplanted cells and to manipulate the host microenvironment. We examine how biomaterial properties can be modulated to improve transplanted cell survival, differentiation and cell engraftment and how the host tissue can be manipulated for cell transplantation by inducing plasticity and vascularization. Finally, we emphasize the importance of the host immune cells for tissue repair and cell transplantation and discuss strategies to tune the immune response through modulating the mechanical properties, architecture, chemistry and functionalization of biomaterials.

Abstract:Stem cells have been utilized as 'living drugs' in clinics for decades. Their self-renewal, differentiation, and immunomodulating properties provide potential solutions for a variety of malignant diseases and disorders. However, the pathological environment may diminish the therapeutic functions and survival of the transplanted stem cells, causing failure in clinical translation. To overcome these challenges, researchers have developed biomaterial-based strategies that facilitatein vivotracking, functional engineering, and protective delivery of stem cells, paving the way for next-generation stem cell therapies. In this perspective, we briefly overview different types of stem cells and the major clinical challenges and summarize recent progress of biomaterials applied to boost stem cell therapies.

Abstract:Introduction: Organ failure and tissue loss are challenging health issues due to widespread injury, the lack of organs for transplantation and limitations of conventional artificial implants. The field of tissue engineering aims to provide alternative living substitutes that restore, maintain or improve tissue function. Areas covered: In this paper, a wide range of porous scaffolds are reviewed, with an emphasis on phase-separation techniques that generate advantageous nanofibrous 3D scaffolds for stem cell-based tissue engineering applications. In addition, methods for presentation and delivery of bioactive molecules to mimic the properties of stem cell niches are summarized. Recent progress in using these bioinstructive scaffolds to support stem cell differentiation and tissue regeneration is also presented. Expert opinion: Stem cells have great clinical potential because of their capability to differentiate into multiple cell types. Biomaterials have served as artificial extracellular environments to regulate stem cell behavior. Biomaterials with various physical, mechanical and chemical properties can be designed to control stem cell development for regeneration. Conclusion: The research at the interface of stem cell biology and biomaterials has made and will continue to make exciting advances in tissue engineering.

Abstract:Biomaterials that exhibit bioinspired behavior enable a range of novel biomedical applications. Cell-friendly biomaterials incorporating crucial physicochemical cues can instruct and dictate cell behavior both in vitro and in vivo. Despite substantial advances, engineering a complex cell regulatory system that incorporates mechanical and chemical signals targeting specific intracellular events has remained far from a reality. Recently, biological studies have begun to decipher the dynamic interplay between stem cells and their microenvironments. Moreover, the myriad recently developed advanced material biotechnologies, such as 3D bioprinting, protein engineering, release technology, high-throughput screening, and microfluidics, have enabled the engineering of sophisticated biomaterial systems that increasingly closely mimic the spatial and temporal characteristics of the native cell milieu. Thus, biomaterials engineered in the laboratory not only provide a fertile environment for fostering stem cells but also exhibit properties that elicit favorable crosstalk between the structural components, the cells and the host tissue in a precise physiological manner, enabling balanced matrix generation and degradation for successful tissue regeneration to be achieved. This progress report discusses these advanced biotechnologies for the design, fabrication, and functionalization of biomaterials, highlighting their merits over traditional methodologies for mimicking the structural and mechanical properties of their natural in vivo counterparts.

Abstract:Biotherapy has recently become a hotspot research topic with encouraging prospects in various fields due to a wide range of treatments applications, as demonstrated in preclinical and clinical studies. However, the broad applications of biotherapy have been limited by critical challenges, including the lack of safe and efficient delivery systems and serious side effects. Due to the unique potentials of biomaterials, such as good biocompatibility and bioactive properties, biomaterial-assisted biotherapy has been demonstrated to be an attractive strategy. The biomaterial-based delivery systems possess sufficient packaging capacity and versatile functions, enabling a sustained and localized release of drugs at the target sites. Furthermore, the biomaterials can provide a niche with specific extracellular conditions for the proliferation, differentiation, attachment, and migration of stem cells, leading to tissue regeneration. In this review, the state-of-the-art studies on the applications of biomaterials in biotherapy, including drug delivery, vaccine development, gene therapy, and stem cell therapy, have been summarized. The challenges and an outlook of biomaterial-assisted biotherapies have also been discussed.

Abstract:Literature in the field of stem cell therapy indicates that, when stem cells in a state of single-cell suspension are injected systemically, they show poor in vivo survival, while such cells show robust cell survival and regeneration activity when transplanted in the state of being attached on a biomaterial surface. Although an attachment-deprived state induces anoikis, when cell-surface engineering technology was adopted for stem cells in a single-cell suspension state, cell survival and regenerative activity dramatically improved. The biochemical signal coming from ECM (extracellular matrix) molecules activates the cell survival signal transduction pathway and prevents anoikis. According to the target disease, various therapeutic cells can be engineered to improve their survival and regenerative activity, and there are several types of biomaterials available for cell-surface engineering. In this review, biomaterial types and application strategies for cell-surface engineering are presented along with their expected efficacy.

Abstract:Biomaterials have gained immense attention for various diseases due to their unique advantages over chemical drugs regarding cost, ease of production, toxicity, and therapeutic efficacy. However, these materials need an inert delivery system that locally delivers cells and minimally elicits foreign body response (FBR). In this aspect, various immunomodulatory biomaterials, including silk, collagen, fibrin, hyaluronic acid, and alginate‐based biomaterials, have great potential due to large natural sources, ease of extraction, low cost, chemical inertness, facile surface functionalization, long‐term polymeric stability, high biocompatibility, and fine tunability of biomaterials sizes and shapes. In this review article, the recent advancements in the biomedical applications of immunomodulatory biomaterials in different diseases, including diabetes, cancer, cardiovascular disease, bone regeneration, etc, are highlighted. Moreover, the FBR on immunomodulatory biomaterials is discussed and the current and future strategies for the potential mitigation of considerable immune responses toward immunomodulatory biomaterials and devices are highlighted. In this review article, the recent advancements in the biomedical applications of immunomodulatory biomaterials in different diseases, including diabetes, cancer, cardiovascular disease, bone regeneration, are highlighted.

Abstract:The precise regulation of cell fate by biomaterials is one of the core focuses in tissue engineering and regenerative therapeutics. Cell fate is controlled by the niche signaling cues, including growth factors/morphogens, direct cell-cell contact, and extracellular matrix components. Here, we discuss how to bio-mimic cell-cell interaction by tailoring the interface of biomaterials to design effective scaffolds. Cadherins, as a key cell membrane protein family mediating the intercellular adherens junction, have attracted extensive attention in the functional modification of biomaterials. Therefore, cadherin-based biomaterials, their ability to modulate stem cell fate, and their applications in regenerative medicine are reviewed. Furthermore, we provide an outlook for exploiting the bio-mimic biomaterials in advanced cell therapeutics.
