Abstract:How to achieve the interfacing between macro world and microchip is still one of the important topics in the research of microfluidic chips. In this paper, the recent progress in high-throughput microfluidic sample introduction techniques is described, mainly based on the work achieved in the authors' group. Various high-throughput microfluidic sample introduction systems in three modes including reservoir, flow-through cell and sampling probe based on microchips or capillaries are described. The developing prospects of high-throughput sample introduction techniques are also forecasted.

Abstract:A microfluidic system for introducing nanolitre sample in inductively coupled plasma mass spectrometry using electrokinetic flow combined with hydrodynamic flow was developed, which consisted of a microfluidic chip, a syringe pump, a high voltage power supply, a homemade microflow nebulizer and a heated single pass spray chamber. In the microfluidic chip, the sample in the reservoir S automatically flew across the intersection P-1 to the reservoir SR under hydrodynamic pressure. Two porous polymer plugs C-1 and C-2 were created on the microchip to isolate electrokinetic flow from the pressure-driven flow. Once a high voltage between S and buffer reservoir B was applied, the sample was driven through the porous polymer plug C-1 into the metering channel by electrokinetic force. Simply changing the sampling time can freely optimize the sampling volume down to 0.18 nL, which was injected into the nebulizer by the pressure-driven makeup solution. The nanolitre sample introducing system has the advantages of low sample consumption, satisfactory precision, low sampling dead volume (0.38 nL) and low sampling dead time (0.42 s). The proposed system was successfully applied to determine platinum content in a serum sample of a cancer patient on cisplatin chemotherapy. The sample consumption of 1.8 nL was about 10(5) times less than that with the conventional sampling system. The sample throughput up to 112 h(-1) could be achieved with the detection limit of 64 ng L-1 for Pt-195. The content of Pt in the sample was 8.4 +/- 0.3 mu g L-1, agreeing well with that (8.5 +/- 0.3 mu g L-1) determined by the conventional sampling system.

Abstract:The development of efficient sample introduction and pretreatment systems for microfluidic chip-based analytical systems is important for their application to real-life samples. In this work, world-to-chip interfacing was achieved by a novel flow-through sampling reservoir featuring a guided overflow design. The flow-through reservoir was fabricated on a 30 x 60 x 3 mm planar glass chip of crossed-channel design used for capillary electrophoresis separations. The 20-microL sample reservoir was produced from a section of plastic pipet tip and fixed at one end of the sampling channel. Sample change was performed by pumping 80-microL samples sandwiched between air segments at approximately 0.48 mL/min flow rate through the flow-through reservoir, introduced from an access hole on the bottom side of the chip. A filter paper collar wrapped tightly around the reservoir guided the overflowing sample solution into a plastic trough surrounding the reservoir and then to waste. The performance of the system was demonstrated in the separation and determination of FITC-labeled arginine, glycine, phenylalanine, and glutamic acid with LIF detection, by continuously introducing a train of different samples through the system without electrical interruption. Employing a separation channel of 4 cm (2-cm effective separation length) and 1.4-kV separation voltage, maximum throughputs of 80/h were achieved with <4.1% carryover and precisions ranging from 1.5% for arginine to 2.6% RSD (n = 11) for glycine. The sampling system was tested in the continuous monitoring of the derivatizing process of amino acids by FITC over a period of 4 h, involving 166 analytical cycles. An outstanding overall precision of 4.8% RSD (n = 166) was achieved for the fluorescein internal standard.

Abstract:In this work, a simple, robust, and automated microfluidic chip-based FIA system with gravity-driven flows and liquid-core waveguide (LCW) spectrometric detection was developed. The high-throughput sample introduction system was composed of a capillary sampling probe and an array of horizontally positioned microsample vials with a slot fabricated on the bottom of each vial. FI sample loading and injection were performed by linearly moving the array of vials filled alternately with 50-microL samples and carrier, allowing the probe inlet to enter the solutions in the vials through the slots sequentially and the sample and carrier solution to be introduced into the chip driven by gravity. The performance of the system was demonstrated using the complexation of o-phenanthroline with Fe(II) as a model reaction. A 20-mm-long Teflon AF 2400 capillary (50-microm i.d., 375-microm o.d.) was connected to the chip to function as a LCW detection flow cell with a cell volume of 40 nL and effective path length of 1.7 cm. Linear absorbance response was obtained in the range of 1.0-100 microM Fe(II) (r2=0.9967), and a good reproducibility of 0.6% RSD (n=18) was achieved. The sensitivity was comparable with that obtained using conventional FIA systems, which typically consume 10,000-fold more sample. The highest sampling throughput of 1000 h-1 was obtained by using injection times of 0.08 and 3.4 s for sample and carrier solution, respectively, with a sample consumption of only 0.6 nL for each cycle.

Abstract:An automated nanoliter sample introduction system was combined to a liquid-core waveguide (LCW)-based microfluidic CE system for high-throughput analysis of DNA fragments. The main component of the sample introduction system was a motor-driven plate, on which a circular array of bottom-slotted vials containing sample/buffer solutions was placed. A 7cm-long LCW capillary served as both the sample probe and separation channel. The inlet terminal of the capillary could pass through the slots of the vials for electrokinetic sample introduction, and the capillary outlet was immersed in the solution of a reservoir, behind which a PMT facing directly to the outlet was positioned. A diode laser was used as excitation source for LCW LIF detection. Performance of the system was demonstrated through the separation of DNA fragments. Baseline separation was achieved for all 11 fragments of (Phi X174-HaeIII digest DNA with a throughput of 33/h. Theoretical plate number for 603 bp fragment was 7.3 X 106/M, corresponding to a plate height 0.14 mu m. The detection limitation for 603 bp fragment was 0.4 ng/mu L with a precision of 2.2% RSD for the peak height. Automated sample changing and introduction were achieved with only 0.3 nL gross sample consumption for each cycle.

Abstract:A newly designed capillary electrophoresis (CE) microchip with a simple and efficient sample introduction interface is described. The sample introduction is carried out directly on the separation channel through a sharp inlet tip placed in the sample vial, without an injection cross, complex microchannel layouts or hardware modification. Alternate placement of the inlet tip in vials containing the sample and buffer solutions permits a volume defined electrokinetic sample introduction. Such fast and simple sample introduction leads to highly reproducible signals with no observable carry over between different analyte concentrations. The performance of the system was demonstrated in flow-injection and CE measurements of nitroaromatic explosives and for on-chip enzymatic assays of glucose in the presence of ascorbic acid. Employing an 8 cm long separation channel and a separation voltage of 4000 V it offers high-throughput flow-injection assays of 100 samples h(-1) with a relative standard deviation of 3.7% for TNT ( n = 100). Factors influencing the analytical performance of the new microchip have been characterized and optimized. Such ability to continuously introduce discrete samples into micrometer channels indicates great promise for high-speed microchip analysis.

Abstract:Sample preparation turns out to be one of the important procedures in complex sample analysis by affecting the accuracy, selectivity, and sensitivity of analytical results. However, the majority of the conventional sample preparation techniques still suffer from time‐consuming and labor‐intensive operations. These shortcomings can be addressed by reforming the sample preparation process in a microfluidic manner. Inheriting the advantages of rapid, high efficiency, low consumption, and easy integration, microfluidic sample preparation techniques receive increasing attention, including microfluidic phases separation, microfluidic field‐assisted extraction, microfluidic membrane separation, and microfluidic chemical conversion. This review overviews the progress of microfluidic sample preparation techniques in the last 3 years based on more than 100 references, we highlight the implementation of typical sample preparation methods in the formats of microfluidics. Furthermore, the challenges and outlooks of the application of microfluidic sample preparation techniques are discussed.

Abstract:A sequential injection micro-sample introduction system was coupled to a microfluidic-chip based capillary electrophoresis system through a split–flow sampling interface integrated on the micro-chip. The microfluidic system measured 20×70×3mm in dimension, and was produced using a non-lithographic approach with components readily available in the analytical laboratory. In the H-configuration channel design the horizontal separation channel was a 75μm I.D.×60mm quartz capillary, with two vertical side arms produced from plastic tubing. The conduits were embedded in silicon elastomer with a planar glass base. Sequential introduction of a series of samples with about 2.5% carryover was achieved at 48h−1 throughput with samples containing a mixture of fluorescein isothiocyanate (FITC)-labeled amino acids using SI sample volumes of 3.3μl and carrier flow-rate of 2.0mlmin−1. Baseline separation was achieved for FITC-labeled arginine, phenylalanine, glycine and FITC (laser induced fluorescence detection) in sodium tetraborate buffer (pH 9.2) within 8–80s, at separation lengths of 25–35mm and electrical field strengths of 250–1500Vcm−1, with plate heights in the 0.7–3μm range.

Abstract:An automated microfluidic sequential injection analysis system that efficiently manipulates sample and reagent solutions in the nanoliter range in approximately 10 s per analytical cycle is described. The system consisted of a 6-cm-long, typically 75-microm i.d., fused-silica capillary (which functioned as a sampling probe and reactor as well as a flow-through detection cell), a horizontally oriented waste reservoir that provided liquid level differences for inducing gravity-driven flows, an autosampling device holding samples and reagents with horizontally fixed slotted microvials, and a laser-induced fluorescence detection system. Sample and reagent zones were sequentially introduced via gravity-driven flow by scanning the capillary tip (functioning as the sampling probe) through the vial slots, while vials containing sample, reagent, and carrier were sequentially rotated to the probe by programmed movement of the vial holders. Sequentially injected nanoliter zones were rapidly mixed by convection and diffusion within the carrier flow, demonstrating a behavior that conformed well to the Taylor dispersion model, and zone penetration effects were characterized and optimized under Taylor's dispersion theory guidelines. For the determination of fluorescein, a high throughput of 400 h(-1) was achieved, rapidly producing calibration curves (five points) within 45 s. Owing to its adaptability to the Taylor's dispersion model, the system was used also for measuring diffusion coefficients of fluorescent species. Potentials for using the system in enzyme inhibition assays were demonstrated by a reaction involving the conversion of fluorescein digalactoside to fluorescent hydrolysates via beta-galactosidase and the inhibition of beta-galactosidase by diethylenetriaminepentaacetic acid.
Fabrication of a Monolithic Sampling Probe System for Automated and Continuous Sample Introduction in Microchip‐based CE

Abstract:A fabrication process for producing monolithic sampling probes on glass chips, with tip diameters of a few hundred micrometers was developed, using simple tools including a glass cutter and a bench drill. Microfluidic chips with probes fabricated by this approach were coupled to a linearly moving slotted-vial array sample presentation system for performing continuous sample introduction in the chip-based CE system. On-chip horizontal tubular reservoirs containing working electrolyte and waste were used to maintain a stable hydrostatic pressure in the chip channels during prolonged working periods. The performance of the system was demonstrated in the separation of FITC-labeled amino acids with LIF detection, by continuously introducing a train of different samples without interruption. Throughputs of 30-60/h were achieved with <1.0% carry-over and reproducibilities in peak height of 3.6, 3.3, and 3.5% RSD for arginine, FITC, and phenylalanine, respectively (n = 11). Continuous analysis of a mixture of FITC-labeled amino acids for 2 h, involving 60 analytical cycles, yielded an RSD of 7.5 and 6.8% for arginine and FITC (n = 60), respectively. An extremely low sample consumption of 30 nL for each analysis was obtained. Separation efficiencies in plate numbers were in the range of 0.8-2x10(5)/m. In addition to the application in sample introduction, the sample/reagent introduction system was also used to produce working electrolyte gradients during a CE separation to improve the separation efficiency. Comparing with isocratic electrophoresis separation, gradient CE demonstrated better separation efficiencies for a mixture of FITC-labeled amino acids.

Abstract:An automated and continuous sample introduction system for microfluidic chip-based capillary electrophoresis (CE) was developed in this work. An efficient world-to-chip interface for chip-based CE separation was produced by horizontally connecting a Z-shaped fused silica capillary sampling probe to the sample loading channel of a crossed-channel chip. The sample presentation system was composed of an array of bottom-slotted sample vials filled alternately with samples and working electrolyte, horizontally positioned on a programmable linearly moving platform. On moving the array from one vial to the next, and scanning the probe, which was fixed with a platinum electrode on its tip, through the slots of the vials, a series of samples, each followed by a flow of working electrolyte was continuously introduced electrokinetically from the off-chip vials into the sample loading channel of the chip. The performance of the system was demonstrated in the separation and determination of FITC-labeled arginine and phenylalanine with LIF detection, by continuously introducing a train of different samples. Employing 4.5 kV sampling voltage (1000 V cm(-1) field strength) for 30 s and 1.8 kV separation voltage (400 V cm(-1) field strength) for 70 s, throughputs of 36 h(-1) were achieved with <1.0% carryover and 4.6, 3.2 and 4.0% RSD for arginine, FITC and phenylalanine, respectively (n = 11). Net sample consumption was only 240 nL for each sample.

Abstract:Microfabrication techniques used for the production of MEMS (micro electro-mechanical systems) have been successfully used to produce highly efficient microfluidic capillary electrophoresis chip systems. A limitation of this approach are the difficulties associated with the creation of the micrometer-sized structures in glass or other substrates, which currently involve specialized and expensive lithographic and etching processes. A further limitation is that hitherto most microfluidic chips are not designed for continuous introduction of a series of different samples, which limits the overall throughput of such systems. This article reviews the development of a microfluidic system for rapid CE separations, produced at a low cost of less than a dollar each, using equipment and materials readily available in the ordinary laboratory. Applications of the system, after coupling to flow-injection and/or sequential-injection sample introduction, for the determination of FITC-labeled amino acids by laser-induced fluorescence, trace metals by chemiluminescence, carbohydrates by amperometry, and inorganic and organic anions by indirect UV absorbance are exemplified to illustrate the performance and versatility of the microfluidic system.
Fabrication of Micro/Nanofluidic Chip on Glass Substrate and Its Application for Electrokinetic Concentration of Proteins

Abstract:A simple and cost-effective double-lithography and double wet-etching method (abbriciated as double-etching technique) was developed for fabricating the full glass chips with hybrid micro- and nanochannels. The microchannels were firstly constructed on a glass substrate by standard UV photolithography and wet etching technique. After washing away the rest photoresist and spin coating of a new layer of photoresist on the microchannel-structured glass substrate, nanochannels with a depth less than 100 nm were prepared on the same substrate by second lithography and wet etching. Finally, the channel-structured glass substrate was sealed to a cover glass plate using room temperature bonding technique. Without clean room facilities and expensive facilities, fluidic chips with hybrid micro- and nanochannel network were fabricated with high yields. The analytical features of those fluidic chips were demonstrated by the successful concentration of fluorescein isothiocyanate(FITC)-labeled human serum albumin(HSA) via electrokinetic ion trapping. Enrichment factor of more than 200 was achieved for 0.5 mg/mL of FITC-HSA solutions within 30 s.

Abstract:A novel microfluidic chip analyzer was developed, in which the capillary electrophoresis (CE) separation with laser induced fluorescence (LIF) detection, simplified optics involving the use of optical fibers, capable of non-interrupted continuous sample introduction using a sequential injection (SI) system were used. Abstract:A facile method is presented for achieving comprehensive two-dimensional droplet manipulations in closed microstructures consisting of microwell arrays and a straight microchannel. In this method, picoliter/nanoliter droplets with controllable sizes and numbers are sampled from nanoliter samples/reagents with almost 100% efficiency. Droplet motions are precisely controlled in the ±X-direction and ±Y-direction by managing hydrostatic pressure and magnetic repulsion, respectively. As a demonstration, a fluorescein-labeled droplet and a deionized droplet are successively generated and trapped in adjacent microwells. Then their positions are quickly exchanged without cross-contamination and fusion is implemented on-demand. After operations, hydrophobic ferrofluid can be completely replaced by mineral oil and droplets still remain in microwells safely. A typical fluorescence intensity-based assay is demonstrated: droplet arrays containing copper ion are diluted disproportionately first and then detected by addition of droplet arrays containing Calcein. With the ability of comprehensive two-dimensional droplet manipulations, this method could be used in various miniaturized biochemical analyses including requirements of multistep procedures and in situ monitoring.

Abstract:We describe a simple miniature injection device that can be used for introduction of nanoliter sample volumes in microfluidic systems. The hybrid microstructure consists of two hydraulically connected parts, a pulse micropump, and a multilevel cross-flow injector. Sample injection is accomplished by creating a transient pressure pulse in the sample line by means of the solenoid-based micropump. The sample line is aligned at right angles to the main carrier flow line. The two flow channels are located in two different parallel planes. The cross section of the two channels is defined by a self-sealing aperture in an elastomer. During the pressure pulse, the sample is introduced through this aperture directly into the main flow stream. Fast impulse-based injection causes rapid mixing of the injected sample with the main flow stream. This permits simple single-line manifold micro flow injection (MFI) systems. The deformation/relaxation of the elastomer is fast and repeatable; as such, rapid serial actuations essentially result in a larger injected sample volume without significantly affecting the peak shape. In the present form, 2-40-nL samples are easily injected by single injection, and the injected volume can be chosen by system parameters. The injection repeatability as observed by a photometric detector is better than 1.2% (n = 100).
