Abstract:An efficient and regioselective method to multisubstituted quinoline and quinazoline derivatives with diaryliodonium salts catalyzed by Cu(OTf)(2) has been presented. The reaction proceeded via cascade annulation in the mode of [2+2+2] form. This novel strategy enables use of readily available materials and bears great flexibility on the substitution groups.

Abstract:An efficient and rapid synthesis of multiply substituted quinolines is described. This method is enabled by a three-component cascade annulation of readily available aryl diazonium salts, nitriles, and alkynes. This reaction is catalyst- and additive-free. Various aryl diazonium salts, nitriles, and alkynes can participate in this transformation, and the yields are up to 83%.

Abstract:CuOTf-catalyzed substituent-controlled cascade [2 + 2 + 2] and [4 + 2] annulation reactions among heterocumulenes, alkynes, and diaryliodonium salts were achieved. Various quinoline derivatives could be obtained in good yields with excellent selectivity. This methodology provided a novel pathway to activate heterocumulenes via a unique highly reactive cationic intermediate. The reaction process was well elucidated by density functional theory calculations.

Abstract:An efficient route to N(4)-substituted 2,4-diaminoquinazolines has been developed by employing tandem condensation of cyanoimidate-amine and reductive cyclization in iron-HCl system. This method is tolerant of a following intramolecular N-alkylation and produces two fused heterocycles in a one-pot procedure. This protocol is a facile two-step synthesis of tricyclic quinazolines, which is effected by potent cyanoimidation and tandem reductive cyclization from 2-nitrobenzaldehydes. Moreover, the forming process of tricyclic quinazolines has been investigated from the ring-opening/ring-closing cascade point of view. It is found that the preparation of tricyclic quinazolinones in good yields relies on the selective hydrolysis of tricyclic quinazolines in base or acid system.
