Abstract:Hexamethyldisilazane-mediated reaction of quinazolin-4(3H)-ones with primary amines led to facile formation of 4-aminoquinazolines through tandem silylation and substitution in a single pot. Excellent yields of the products (83-97%) and environmental friendliness (avoiding the use of chlorination reagents) are the advantages of this method.

Abstract:A simple, environmentally friendly, one-pot method for the synthesis of 4-aminoquinazolines using microwave irradiation has been developed. Structures of derivatives 3, 4, and 5 were confirmed by single-crystal X-ray diffraction. The in vitro cytotoxicity of each compound was investigated using an MTT assay with A549 and HepG2 cell lines to calculate half-maximal inhibitory concentrations.

Abstract:Quinazolin-4 （3H）-ones were conveniently synthesized from anthranilie acids and amidines via cyclization reaction, which could be chloridized with POCl3 to obtain 4-chloroquinazoline derivatives. The structures of the products were characterized by 1HNMR and MS.

Abstract:An efficient direct amination of quinazolin-4(3H)-ones has been developed. Treatment of quinazolin-4(3H)-ones with HCCP, DIPEA, and N-contained nucleophiles in acetonitrile could be able to form the corresponding 4-aminoquinazoline derivatives. Under the optimal reaction conditions, the amination products were achieved in good yields.

Abstract:A novel and catalyst-free one-pot method for the preparation of quinazoline compounds has been reported.Of the prepared 15 quinazoline compounds,three are new compounds.Their yields are among 37％～95％.The effects of electron effect and steric hindrance on the reaction were discussed.A plausible novel mechanism concerning repeating nucleophilic addition and elimination to give quinazoline has been proposed.By this mechanism,the reaction could be applied to acid anhydride instead of N,N-dimethylformamide (DMF).
