Abstract:Rh(III)-catalyzed C(sp(3)) - H bond aminocarbonylation of 8-methylquinolines and isocyanates has been realized under mild conditions. This approach is applicable to different aryl and alkyl isocyanates, leading to the synthesis of various alpha-quinolinyl amide compounds in moderate to excellent yields. A plausible mechanism for this transformation is proposed according to the experimental results obtained.

Abstract:An efficient RhIII-catalyzed ortho-C-H bond activation for the synthesis of substituted isoquinolines and heterocycle-fused pyridines in aqueous medium has been developed. This method involves the in situ generation of ketimines from ketones and ammonium acetate and subsequent oxidative C-H bond activation/annulation of ketimines with alkynes to form the C-C/C-N bonds spontaneously. Various aryl ketones and internal alkynes were smoothly transformed into the desired heterocycles in moderate to excellent yields. The reaction exploits molecular oxygen as the sole oxidant by producing water as the only byproduct. Moreover, easily available NH4OAc was utilized as the nitrogen source, which makes this protocol practical and environmentally friendly.

Abstract:One-pot syntheses of 1-benzyl-, 1-alkoxyl-, and 1-alkylamino- isoquinolines through automatic directing group (DGauto)-assisted, rhodium(III)-catalyzed aryl C-H activation and annulation with internal alkynes were developed. The reactions affording 1-benzylisoquinolines involve a cascade oximation of diarylacetylenes with hydroxylamine, forming aryl benzyl ketone oxime, and oxime-assisted rhodium(III)-catalyzed aryl C-H activation and followed annulation with another molecule of diarylacetylene in a one-pot manner. The formation of 1-alkoxyl/amino isoquinolines includes the addition of nucleophilic alcohols or amines to aryl nitriles, imine-assisted rhodium-catalyzed aryl C-H activation, and subsequent alkyne annulation.

Abstract:Isoquinolines have been synthesized from the redox-neutral dehydrative C-N and C-C cross-coupling between oximines and alkynes using a catalytic amount of (pentamethylcyclopentadiene)rhodium dichloride dimer {[RhCp*Cl-2](2)} and cesium acetate (CsOAc), a process that involves ortho C-H activation of oximines and subsequent functionalization with alkynes. This redox-neutral catalytic isoquinoline synthesis operates under mild conditions, and is insensitive to moisture or air. A broad scope of coupling partners has been established, and a likely mechanism has been suggested.

Abstract:Rh(III)-catalyzed C-H activation/annulation of benzimidates with 4-diazoisochroman-3-imines furnished 8-alkoxy-5H-isochromeno[3,4-c]isoquinolines in moderate to excellent yields with a broad range of substrate scope. The reaction was carried out under mild reaction conditions and could be scaled up with practical usage. Similar reaction between benzimidates and 4-diazoisoquinolin-3-ones provided 1-alkoxy-4-arylisoquinolin-3-ols in excellent yields. Moreover, the synthesized products could be conveniently transformed to the corresponding heterocycles with a 1,8-naphthyridinone or isochromenopyridinone core, which are privileged structures in medicinal chemistry.

Abstract:A synthetic method of isoquinolines from aryl ketone O-acyloxime derivatives and internal alkynes has been developed using [Cp*RhCl(2)](2)-NaOAc as the potential catalyst system. The present transformation is carried out by a redox-neutral sequence of C-H vinylation via ortho-rhodation and C-N bond formation of the putative vinyl rhodium intermediate on the oxime nitrogen, where the N-O bond of oxime derivatives could work as an internal oxidant to maintain the catalytic cycle.

Abstract:An Rh(III)-catalyzed one-pot synthesis of 5H-isochromeno[3,4-c]isoquinolines from readily available 4-diazoisochroman-3-imines and (2-formylphenyl)boronic acids is reported. The cascade annulation involves a Rh(III)-catalyzed cross-coupling and an intramolecular nucleophilic addition-elimination process. A series of biologically important 5H-isochromeno[3,4-c]isoquinolines were obtained in good to excellent yields. The method can be extended to synthesize 7H-isochromeno[3,4-b]thieno[3,2-d]pyridines and 7H-isochromeno[3,4-b]thieno[2,3-d]pyridines from the corresponding heteroaryl boronic acids.

Abstract:The development of an efficient strategy for facile access to quinoline-based bis-heterocycles holds paramount importance in medicinal chemistry. Here in, we describe a unified approach for accessing 8-(indol-3-yl)methyl-quinolines by integrating Cp*Rh(III)-catalyzed C(sp3)–H bond activation of 8-methylquinolines followed by nucleophilic cyclization with o-ethynylaniline derivatives. Remarkably, methoxybiaryl ynones under similar catalytic conditions delivered quinoline tethered spiro[5.5]enone scaffolds via a dearomative 6-endo-dig C-cyclization. Moreover, leveraging this method for C8(sp2)−H bond activation of quinoline-N-oxide furnished biologically relevant oxindolyl-quinolines. This reaction proceeds via C(sp2)–H bond activation, regioselective alkyne insertion, oxygen-atom-transfer (OAT) and intramolecular nucleophilic cyclization in a cascade manner. One C−C, one C−N and one C=O bonds were created with concomitant formation of a quaternary center.
