Abstract:Rh(III)-catalyzed C(sp(3)) - H bond aminocarbonylation of 8-methylquinolines and isocyanates has been realized under mild conditions. This approach is applicable to different aryl and alkyl isocyanates, leading to the synthesis of various alpha-quinolinyl amide compounds in moderate to excellent yields. A plausible mechanism for this transformation is proposed according to the experimental results obtained.

Abstract:A synthetic method of isoquinolines from aryl ketone O-acyloxime derivatives and internal alkynes has been developed using [Cp*RhCl(2)](2)-NaOAc as the potential catalyst system. The present transformation is carried out by a redox-neutral sequence of C-H vinylation via ortho-rhodation and C-N bond formation of the putative vinyl rhodium intermediate on the oxime nitrogen, where the N-O bond of oxime derivatives could work as an internal oxidant to maintain the catalytic cycle.

Abstract:One-pot syntheses of 1-benzyl-, 1-alkoxyl-, and 1-alkylamino- isoquinolines through automatic directing group (DGauto)-assisted, rhodium(III)-catalyzed aryl C-H activation and annulation with internal alkynes were developed. The reactions affording 1-benzylisoquinolines involve a cascade oximation of diarylacetylenes with hydroxylamine, forming aryl benzyl ketone oxime, and oxime-assisted rhodium(III)-catalyzed aryl C-H activation and followed annulation with another molecule of diarylacetylene in a one-pot manner. The formation of 1-alkoxyl/amino isoquinolines includes the addition of nucleophilic alcohols or amines to aryl nitriles, imine-assisted rhodium-catalyzed aryl C-H activation, and subsequent alkyne annulation.

Abstract:A Rh(III)-catalyzed oxidative annulation of pyridin-2(1H)-ones with alkynes via double C–H activation to produce highly functionalized 4H-quinolizin-4-ones is disclosed. This reaction features easily available starting materials, simple manipulation, a relatively wide substrate scope, and good functional group tolerance. The application of this protocol is demonstrated by the synthesis of a known fluorescent quinolizino[3,4,5,6-ija]quinolinium salt.

Abstract:A protocol realized by rhodium‐catalyzed C−H functionalization of O‐pivaloyl oximes with acryloylsilane was developed for synthesis of highly functionalized 3‐acylsilane‐substituted isoquinolines. Abstract:Rhodium (Rh)-catalysed [4+2] annulation of aromatic oximes with common terminal alkenes for the synthesis of 3,4-dihydroisoquinolines is presented. Through the cooperation of a Rh(III) catalyst and a catalytic amount of K2HPO4 base, the reaction enables the formation of two new bonds, a C(sp(2))-C(sp(3)) bond and a C(sp(3))-N bond, in a single reaction via functionalization of both the C-H and N-O bonds and provides a practical method to produce 3,4-dihydroisoquinolines with exquisite selectivity and excellent compatibility of functional groups.

Abstract:A rhodium-catalyzed coupling-cyclization reaction of alkenyldiazoacetates with o-alkenyl arylisocyanides has been developed. In this reaction, the highly reactive alkenylketenimine intermediates generated by coupling reaction of alkenyldiazoacetates with o-alkenyl arylisocyanides undergo intramolecular [4 + 2] cycloaddition and provide a novel, highly efficient method for the one-step synthesis of carbazoles by formation of three new bonds and two rings from readily available acyclic starting materials.

Abstract:An efficient Rh2(II,II)-catalyzed reaction has been developed under mild conditions. This synthetic method proceeds through iodination/oxidation of readily available isoquinolinium iodide salts under aerobic conditions with good to excellent yields. 4-Iodoisoquinolin-1(2H)-ones are important building blocks for biologically and medicinally important compounds. The developed methodology was applied to the gram-scale synthesis of a key intermediate in the synthesis of the CRTH2 antagonist CRA-680.

Abstract:A rhodium(III)-catalyzed one-pot three-component reaction of N-methoxybenzamide, α-diazoester, and alkyne was developed, providing an alternative way to synthesize isoquinoline derivatives. Abstract:Isoquinolines have been synthesized from the redox-neutral dehydrative C-N and C-C cross-coupling between oximines and alkynes using a catalytic amount of (pentamethylcyclopentadiene)rhodium dichloride dimer {[RhCp*Cl-2](2)} and cesium acetate (CsOAc), a process that involves ortho C-H activation of oximines and subsequent functionalization with alkynes. This redox-neutral catalytic isoquinoline synthesis operates under mild conditions, and is insensitive to moisture or air. A broad scope of coupling partners has been established, and a likely mechanism has been suggested.

Abstract:An efficient RhIII-catalyzed ortho-C-H bond activation for the synthesis of substituted isoquinolines and heterocycle-fused pyridines in aqueous medium has been developed. This method involves the in situ generation of ketimines from ketones and ammonium acetate and subsequent oxidative C-H bond activation/annulation of ketimines with alkynes to form the C-C/C-N bonds spontaneously. Various aryl ketones and internal alkynes were smoothly transformed into the desired heterocycles in moderate to excellent yields. The reaction exploits molecular oxygen as the sole oxidant by producing water as the only byproduct. Moreover, easily available NH4OAc was utilized as the nitrogen source, which makes this protocol practical and environmentally friendly.

Abstract:Rh(III)-catalyzed C-H activation/annulation of benzimidates with 4-diazoisochroman-3-imines furnished 8-alkoxy-5H-isochromeno[3,4-c]isoquinolines in moderate to excellent yields with a broad range of substrate scope. The reaction was carried out under mild reaction conditions and could be scaled up with practical usage. Similar reaction between benzimidates and 4-diazoisoquinolin-3-ones provided 1-alkoxy-4-arylisoquinolin-3-ols in excellent yields. Moreover, the synthesized products could be conveniently transformed to the corresponding heterocycles with a 1,8-naphthyridinone or isochromenopyridinone core, which are privileged structures in medicinal chemistry.

Abstract:A β‐hydroxy elimination instead of common oxidization of carbonyl groups in secondary propargylic alcohols were successfully developed to form 2‐benzyl substituted isoquinoline N‐oxides by a Rh‐catalyzed C‐H activation and annulation cascade, in which moderate to excellent yields (up to 92%) could be obtained under mild reaction conditions, along with good regioselectivity, broad generality and applicability.
