Abstract:Substituted phenacyl bromides react with isoquinoline to form the corresponding quaternary salts which, when heated in ammonium acetate and acetic acid in the presence of Cu(II)O, undergo regioselective cyclisation to give 2-arylimidazo[2,1-a]isoquinolines uniquely.

Abstract:A concise approach for the synthesis of quinoxalin-2-one derivatives through substituted o-phenylenediamines and alpha-ketoester with high regioselectivity is developed. Interestingly, the two regioisomers can be selectively obtained and the regioselectivity can be regulated by acid and base. The typical regioisomer ratio is up to 15:1 in acidic conditions and it can be easily reversed as 1:4 in basic conditions.

Abstract:A one-pot procedure has been developed to prepare the derivatives of 2-arylimidazo(2, 1-a)-isoquinolines (2a similar to 2b), 2-arylimidazo(1, 2-a) quinolines (4a similar to 4b) and 2-arylimidazo(1, 2-a) phthalazines (6a similar to 6c) in moderate yields (51% similar to 62%) by the reaction of isoquinolinium salts, quinolinium salts or phdialazinium salts with ammonium acetate in the presence of tetrakispyridine cobalt(II) dichromate (TPCD).

Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:A solvent controlled regioselective metal-free synthesis of iodo-substituted N-heterocycles has been developed. This protocol undergoes a cascade iodination/cyclization/oxidation/aromatization pathway to afford multi-halogenated quinolines from readily available propargylamines under mild conditions.

Abstract:Here, a general copper-catalyzed radical cascade carbocyclization reaction with 2-arylbenzoimidazoles and a Togni reagent was realized. Structurally diverse CF3-containing tetracyclic core benzimidazo[2,1-a]isoquinoline-6(5H)-ones were obtained in moderate to good yields. The wide substrate scope, good functional group tolerance, and ease of scale-up of this method are expected to promote its potential applications in pharmacy and biotechnology.

Abstract:Herein, a general strategy for the remote-site-selective cascade addition/cyclization of unactivated C(sp3)-H bonds in free alcohols and sulfonamides to build isoquinolinonedione skeletons is developed. The site selectivity occurs predominantly via a 1,5-hydrogen atom transfer (HAT) process, triggered by heteroatom-centred radicals generated directly under silver catalysis. A broad substrate scope and excellent regio-/chemo-selective control are demonstrated in this method.

Abstract:A series of 15-arylisoquinolino[2′,1′:1,2]imidazo[4,5-f][1,10]phenanthrolines was synthesized by a copper(I)-catalyzed reaction of 2-(2-bromophenyl)-1H-imidazo[4,5-f][1,10] phenanthrolines and terminal alkyne in the presence of Cs2CO3. It was proved to be an efficient method for the synthesis of hexacyclic compounds bearing three kinds of heterocycles which were isoquinoline, imidazole and phenanthroline.

Abstract:The 1-aryl tetrahydroisoquinolines (1-aryl THIQs) are omnipresent in biologically active molecules. Here we report on the direct asymmetric synthesis of these valuable compounds via the reaction of 3,4-dihydroisoquinolinium tetraarylborates. Abstract:A copper-catalyzed sequential arylation and intramolecular annulation of 2-(2-bromophenyl)-2,3-dihydroquinazolin-4(1H)-ones with alkyl amidines has been developed for the synthesis of 6-alkyl 11bHquinazolino[ 3,4-a] quinazolin-13(12H)-ones (3) in moderate to good yields. Abstract:A novel and regioselective Ni(I) catalyzed C-C and C-N cascade coupling reactions has been developed. The cascade furnishes atom-economic access to 40 3-aryl-1-aminoisoquinolines. The regioselectivity of C(sp(3))-cyano group over C(sp(2))-cyano group was revealed and supported by mechanism studies as well as the preliminary density functional theory (DFT) calculations.

Abstract:Cyclopenta[b]quinolines and cyclohexa[b]quinolines were prepared via the reactions of α-diazo ketones with N-(2-cyclopropylidenemethylphenyl)phosphanimines and N-(2-cyclobutylidenemethylphenyl) phosphanimine, respectively. The reaction proceeds in a cascade involving ketenimine formation, 6 π-electron ring closure, and 1,3-alkyl shift. A similar approach was developed for the synthesis of dihydropyrrolo-[2,3-b]quinolines from N-(2-cyclopropylidenemethylphenyl)phosphanimines and isocyanates.

Abstract:An efficient approach to access functionalized dihydro-[1,3]oxazino[4,3-a] isoindole and tetrahydroisoquinoline skeletons has been developed through the addition-cyclization process of ynamides 8 with N-acyliminium ions generated from N,O-acetals 6,7. The reactions were conducted under the catalysis of Cu(OTf)(2), and a number of functionalized dihydro-[1,3]oxazino[4,3-a] isoindoles 9a-9y and tetrahydroisoquinolines 10a-10g, 11a-11p were generated in 48-98% yields. When chiral ynamides 8n-8u were used, optically pure products 11a-11p could be obtained with good to excellent yields and diastereoselectivities. (C) 2021 Elsevier Ltd. All rights reserved.

Abstract:Rh(III)-catalyzed C-H activation/annulation of benzimidates with 4-diazoisochroman-3-imines furnished 8-alkoxy-5H-isochromeno[3,4-c]isoquinolines in moderate to excellent yields with a broad range of substrate scope. The reaction was carried out under mild reaction conditions and could be scaled up with practical usage. Similar reaction between benzimidates and 4-diazoisoquinolin-3-ones provided 1-alkoxy-4-arylisoquinolin-3-ols in excellent yields. Moreover, the synthesized products could be conveniently transformed to the corresponding heterocycles with a 1,8-naphthyridinone or isochromenopyridinone core, which are privileged structures in medicinal chemistry.

Abstract:An iridium-catalyzed trifluoroacetic acid-promoted asymmetric cascade allylation/Pictet–Spengler cyclization reaction of azomethine ylides with aromatic allylic alcohols is reported. This protocol provides a facile and scalable method for the construction of 1,3,4-trisubstituted tetrahydroisoquinolines containing two stereogenic centers in good yields (up to 96%) with generally excellent diastereo- and enantioselectivities (up to >20:1 dr and >99% ee).
