Abstract:Asymmetric expansion: A catalytic asymmetric ring-expansion reaction of the title compounds occurs in the presence of a Sc(OTf)(3) catalyst bearing an N,N'-dioxide-based ligand. Highly functionalized 2-quinolone derivatives containing a chiral C4-quaternary stereocenter were obtained in high yields and high levels of selectivity under mild reaction conditions (see scheme; Tf=trifluoromethanesulfonyl).

Abstract:A general method was developed for the synthesis of 1-difluoroalkyl isoquinolines via the palladium-catalyzed radical cascade difluoroalkylation-cyclization of vinyl isocyanides with bromodifluoroacetic derivatives. The difluoroalkylated cyclization products were readily converted to various other valuable gem-difluoro-containing compounds.

Abstract:A catalyst-free chemoselective trimerization reaction of readily available isocyanides is described. This domino reaction provides facile access to a wide range of 2-(indol-2-yl)-quinolines and 2-(indol-2-yl)-pyridines in moderate to excellent yields. A "head to head" heterodimerization of two isocyanides is proposed as the key step of this reaction.

Abstract:A synthetic strategy for multi-substituted isoquinoline derivatives has been developed using visible light-promoted vinyl isocyanide insertion with diaryliodonium salts at room temperature. Abstract:A mechanistically new strategy has been described for the simple, practical, and environmentally friendly preparation of 6-(trifluoromethyl)phenanthridine derivatives using ionic isocyanide insertion from biphenyl isocyanide derivatives and Umemoto's reagent. These reactions were promoted only by inorganic base in good-to-excellent chemical yields without any external stoichiometric oxidants and radical initiators.

Abstract:Condensation of alpha,beta-unsaturated trifluoromethyl ketones with anilines in trichloroacetic acid (TCA) afforded 2-trifluoromethyl quinolines in moderate to excellent yields, in which a reversal of the regiochemistry of the standard Skraup-Doebner-Von Miller quinoline synthesis was achieved. Our endeavors will lead to a better understanding of the controlling elements behind the structural motif.

Abstract:Herein, a general strategy for the remote-site-selective cascade addition/cyclization of unactivated C(sp3)-H bonds in free alcohols and sulfonamides to build isoquinolinonedione skeletons is developed. The site selectivity occurs predominantly via a 1,5-hydrogen atom transfer (HAT) process, triggered by heteroatom-centred radicals generated directly under silver catalysis. A broad substrate scope and excellent regio-/chemo-selective control are demonstrated in this method.

Abstract:A regiospecific synthesis of ortho-trifluoromethvlated and ortho-(fluoro)alkylated pyridine derivatives has been developed. This strategy is enabled by visible light-promoted vinyl isocyanide insertions with Umemoto's reagent and electron-deficient bromides at room temperature. The methodology presented here provides an access to highly functionalized ortho-(fluoro)alkylated pyridine derivatives regiospecifically under mild conditions with good yields. The proposed mechanism was supported by TEMPO trapping experiments. Stern-Volmer analysis and light off/on and time profile experiments.
