Abstract:A Rh(III)-catalyzed one-pot reaction of benzamides, ketones, and hydrazines for facile access to isoindolinones is reported. In this method, various ketones are transformed into donor-donor diazo compounds, which sequentially engage in insertion with benzamides under Rh(III) catalysis to generate N-substituted quaternary isoindolinones.

Abstract:A cobalt and copper catalyzed reaction protocol has been developed to achieve the oxidative C-H/N-H annulation of benzamides containing an 8-aminoquinoline moiety as the directing group with ketones. Structurally diverse isoindolin-1-ones were furnished by the reaction of various substituent benzamides with ketones.

Abstract:A Rh(III)-catalyzed reaction of N-methoxybenzamides and 4-diazoisochroman-3-imines is described. This method offers a rapid entry to 3-amino-4-arylisoquinolinone architectures from readily available starting materials under mild reaction conditions. A one-pot cascade C-H bond activation/formal [4 + 2] cycloaddition mechanism is proposed. The synthesized 3-amino-4-arylisoquinolinones can be conveniently converted into dibenzo[c,f][1,8]naphthyridines.

Abstract:An efficient and convenient Rh(III)-catalyzed double aryl C(sp2)-H bond and N-H activation and annulation reaction is reported for the synthesis of indolo[2,1-a]isoquinolines in the presence of the Cu(OAc)2 oxidant under heating conditions. Distinct from previous works with other arylamine donors, one molecule of 1,3-diarylurea can serve as a precursor of two molecules of arylamine in the reaction with diaryl-substituted alkynes.

Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:Synthesis of isoquinolines based on efficient C-C and C-N bond formation through Rh(III)-catalyzed C-H activation and subsequent intramolecular cyclization is reported. Diazodiesters serving as a C-2 source in the newly formed heterocycles are first demonstrated. Additionally, the Rh(III)-catalyzed direct C-H activation/cyclization of benzimidates with diazoketoesters is also described.

Abstract:Rhodium-catalyzed C H activation/annulation reactions of ketenimines with N-methox-ybenzamides are reported. The outcome of reactions is dependent on the structure of ketenimines. The beta-alkyl-substituted ketenimines furnish 3-iminoisoquinolin-1(2H)-ones in a formal [4 + 2] annulation manner, while the beta-ester substituted ketenimines afford 3-aminoisoindolin-l-ones in a formal [4 + 1]. annulation manner. The synthesized [4 + 2] products undergo an intramolecular Cu-catalyzed C N coupling to be converted to benzo[4,5]imidazo[1,2-b]isoquinolin-11-ones, which can, be directly prepared from ketenimines and N-methoxybenzamides coupling sequence. by a one-pot Rh-catalyzed annulation/Cia-catalyzed C-N coupling sequence.

Abstract:Rh(III)-catalyzed C-H activation/annulation of benzimidates with 4-diazoisochroman-3-imines furnished 8-alkoxy-5H-isochromeno[3,4-c]isoquinolines in moderate to excellent yields with a broad range of substrate scope. The reaction was carried out under mild reaction conditions and could be scaled up with practical usage. Similar reaction between benzimidates and 4-diazoisoquinolin-3-ones provided 1-alkoxy-4-arylisoquinolin-3-ols in excellent yields. Moreover, the synthesized products could be conveniently transformed to the corresponding heterocycles with a 1,8-naphthyridinone or isochromenopyridinone core, which are privileged structures in medicinal chemistry.

Abstract:A Rh(III)-catalyzed C-H bond activation for the synthesis of fused 2H-isoindole scaffolds from oxadiazolones with diazo compounds was developed. The reaction proceeded through C-H activation of oxadiazolones/[4 + 1] annulation, intramolecular cyclization, and an unusual acyl migration cascade to afford target scaffolds with good yields. These 2H-isoindole derivatives could be further transformed into intriguing drug privileged scaffolds.

Abstract:Comprehensive Summary A series of 3‐acyl‐substituted isoindolinone derivatives were synthesized in one‐pot manner via the reaction of o‐bromobenzaldehydes, isocyanides, and carboxylic acids in the presence of palladium catalyst and base. The reaction employing easily available starting materials features simple operation and high efficiency. The mechanistic study showed that the reaction might undergo 1) Pd‐catalyzed [3+2] cyclization of o‐bromobenzaldehyde with isocyanide and the re‐insertion of another molecule of isocyanide, 2) addition of carboxylic acid to in situ formed ketenimine followed by a rearrangement relay to give 3,3‐diacyl‐substituted isoindolinone derivative.
