Abstract:This review summarizes programmable microfluidics, an advanced method for precise fluid control in microfluidic technology through microchannel design or liquid properties, referring to microvalves, micropumps, digital microfluidics, multiplexers, micromixers, slip-, and block-based configurations. Different microvalve types, including electrokinetic, hydraulic/pneumatic, pinch, phase-change and check valves, cater to diverse experimental needs. Programmable micropumps, such as passive and active micropumps, play a crucial role in achieving precise fluid control and automation. Due to their small size and high integration, microvalves and micropumps are widely used in medical devices and biological analysis. In addition, this review provides an in-depth exploration of the applications of digital microfluidics, multiplexed microfluidics, and mixer-based microfluidics in the manipulation of liquid movement, mixing, and splitting. These methodologies leverage the physical properties of liquids, such as capillary forces and dielectric forces, to achieve precise control over fluid dynamics. SlipChip technology, which branches into rotational SlipChip and translational SlipChip, controls fluid through sliding motion of the microchannel. On the other hand, innovative designs in microfluidic systems pursue better modularity, reconfigurability and ease of assembly. Different assembly strategies, from one-dimensional assembly blocks and two-dimensional Lego®-style blocks to three-dimensional reconfigurable modules, aim to enhance flexibility and accessibility. These technologies enhance user-friendliness and accessibility by offering integrated control systems, making them potentially usable outside of specialized technical labs. Microfluidic programmable strategies for channels and flow hold promising applications in biomedical research, chemical analysis and drug screening, providing theoretical and practical guidance for broader utilization in scientific research and practical applications.

Abstract:In this article, we show a novel approach to implementing valving action in a dynamically evolving capillary filling process, by exploiting alternating current electrothermal (ACET) mechanism. The surfaces of the top and bottom walls of the capillary are asymmetrically patched with two different wettabilities. Our observations reveal that by exploiting an intricate interplay of the electrothermal effects and interfacial tension modulation, the acceleration-deceleration motion of the interface and interface pinning at specific locations may be delicately tuned. This, in turn, modulates the flow speed and residence time of the interface at designated locations, with a simple tuning of the frequency of the electrical signal. The present study, thus, unveils a novel micro valving mechanism achieved via effective manoeuvring of the interfacial electrochemistry where active and passive control mechanisms can be utilized simultaneously to achieve control over the interfacial motion of the two-phase system. This may bear far-reaching consequence in optimizing several applications in the bio-microfluidic domain, including devising novel miniaturized platforms for locally investigating reaction chemistries.
Thermally-actuated microfluidic membrane valve for point-of-care applications

Abstract:Abstract Microfluidics has enabled low volume biochemistry reactions to be carried out at the point-of-care. A key component in microfluidics is the microfluidic valve. Microfluidic valves are not only useful for directing flow at intersections but also allow mixtures/dilutions to be tuned real-time and even provide peristaltic pumping capabilities. In the transition from chip-in-a-lab to lab-on-a-chip, it is essential to ensure that microfluidic valves are designed to require less peripheral equipment and that they are transportable. In this paper, a thermally-actuated microfluidic valve is presented. The valve itself is fabricated with off-the-shelf components without the need for sophisticated cleanroom techniques. It is shown that multiple valves can be controlled and operated via a power supply and an Arduino microcontroller; an important step towards transportable microfluidic devices capable of carrying out analytical assays at the point-of-care. It is been calculated that a single actuator costs less than $1, this highlights the potential of the presented valve for scaling out. The valve operation is demonstrated by adjusting the ratio of a water/dye mixture in a continuous flow microfluidic chip with Y-junction channel geometry. The power required to operate one microfluidic valve has been characterised both theoretically and experimentally. Cyclical operation of the valve has been demonstrated for 65 h with 585 actuations. The presented valve is capable of actuating rectangular microfluidic channels of 500 μm × 50 μm with an expected temperature increase of up to 5 °C. The fastest actuation times achieved were 2 s for valve closing (heating) and 9 s for valve opening (cooling).

Advanced Capillary Soft Valves for Flow Control in Self-Driven Microfluidics

Abstract:Self-driven microfluidic devices enable fully autonomous handling of very small volumes of liquid samples and reagents. However, many applications require an active control mechanism to trigger self-driven flow in microchannels. Here, we report on capillary soft valves (CSVs), which enable stopping a liquid filling front at a precise location inside a microchannel and can resume flow of liquid upon simple actuation. The working mechanism of a CSV is based on a barrier of capillary pressure induced by an abruptly expanding microchannel. We discuss the influence of wetting conditions on the performance of a CSV and the effect of elevated temperatures on a CSV in its closed state. We introduce design features such as pillars and cavities, as well as fabrication techniques for rounded microchannels, which all may broaden the applicability and robustness of CSVs in microfluidic devices. Finally, we present CSV having multiple inlet channels. CSVs further diversify the toolbox of microfluidic functionalities and yet are simple to implement, fabricate and actuate.

An embedded microfluidic valve for dynamic control of cellular communication

Abstract:The communication between different cell populations is an important aspect of many natural phenomena that can be studied with microfluidics. Using microfluidic valves, these complex interactions can be studied with a higher level of control by placing a valve between physically separated populations. However, most current valve designs do not display the properties necessary for this type of system, such as providing variable flow rate when embedded inside a microfluidic device. While some valves have been shown to have such tunable behavior, they have not been used for dynamic, real-time outputs. We present an electric solenoid valve that can be fabricated completely outside of a cleanroom and placed into any microfluidic device to offer control of dynamic fluid flow rates and profiles. After characterizing the behavior of this valve under controlled test conditions, we developed a regression model to determine the required input electrical signal to provide the solenoid the ability to create a desired flow profile. With this model, we demonstrated that the valve could be controlled to replicate a desired, time-varying pattern for the interface position of a co-laminar fluid stream. Our approach can be performed by other investigators with their microfluidic devices to produce predictable, dynamic fluidic behavior. In addition to modulating fluid flows, this work will be impactful for controlling cellular communication between distinct populations or even chemical reactions occurring in microfluidic channels.
Microfluidic Time-Delay Valve Mechanism on Paper-Based Devices for Automated Competitive ELISA

Abstract:Paper-based technologies have been drawing increasing attentions in the biosensor field due to their economical, ecofriendly, and easy-to-fabricate features. In this paper, we present a time-delay valve mechanism to automate a series of procedures for conducting competitive enzyme-linked immunosorbent assay (ELISA) on a paper-based device. The mechanism employs a controllable time-delay valve, which has surfactants to dissolve the hydrophobic barriers, in a fluid pathway. The valves can regulate the liquid and sequentially deliver the sample flow for automating ELISA procedures in microchannels. Competitive ELISA is achieved in a single step once the sample, or small molecule pesticide (e.g., Imidacloprid), is applied onto the paper-based device with a comparable sensitivity to plate-based competitive ELISA. The results further demonstrate the appositeness of using paper-based devices with the valve designs for on-the-go ELISA detection in agriculture and biomedical applications.

Abstract:Microfluidic channels with embedded ion permselective medium under the application of electric current are commonly used for electrokinetic processes as on-chip ion concentration polarization (ICP) and bioparticle preconcentration to enhance biosensing. Herein, we demonstrate the ability to dynamically control the electrically driven ion transport by integrating individually addressable microvalves. The microvalves are located along a main microchannel that is uniformly coated with a thin layer of an ion-exchange membrane (IEM). The interplay of ionic transport between the solution within the microchannel and the thin IEM, under an applied electric current, can be locally tuned by the deformation of the microvalve. This tunability provides a robust and simple means of implementing new functionalities into lab-on-a-chip devices, e.g., dynamic control over multiple ICP layers and their associated preconcentrated molecule plugs, multiplex sensing, suppression of biofouling as well as plug dispersion, while maintaining the well-known application of microvalves as steric filtration.

Abstract:The combination of atomic force microscopy (AFM) with nanofluidics, also referred to as FluidFM, has facilitated new applications in scanning ion conductance microscopy, direct force measurements, lithography, or controlled nanoparticle deposition. An essential element of this new type of AFMs is its cantilever, which bears an internal micro-channel with a defined aperture at the end. Here, we present a new approach for in-situ characterization of the internal micro-channels, which is non-destructive and based on electrochemical methods. It allows for probing the internal environment of a micro-channeled cantilever and the corresponding aperture, respectively. Acquiring the streaming current in the microchannel allows to determine not only the state of the aperture over a wide range of ionic strengths but also the surface chemistry of the cantilever's internal channel. The high practical applicability of this method is demonstrated by detecting the aspiration of polymeric, inorganic and hydrogel particles with diameters ranging from several mu m down to 300 nm. By verifying in-situ the state of the aperture, i.e. open versus closed, electrophysiological or nano-deposition experiments will be significantly facilitated. Moreover, our approach is of high significance for direct force measurements by the FluidFM-technique and sub-micron colloidal probes.

Abstract:Fluid manipulation plays an important role in biomedical applications such as biochemical assays, medical diagnostics, and drug development. Programmable fluidic manipulation at the microscale is highly desired in both fundamental and practical aspects. In this paper, we summarize some of the latest studies that achieve programmable fluidic manipulation through intricate capillaric circuits design, construction of biomimetic metasurface, and responsive surface wettability control. We highlight the working principle of each system and concisely discuss their design criterion, technical improvements, and implications for future study. We envision that with multidisciplinary efforts, microfluidics would continue to bring vast opportunities to biomedical fields and make contributions to human health.
Wirelessly powered and remotely controlled valve-array for highly multiplexed analytical assay automation on a centrifugal microfluidic platform

Abstract:In this paper we present a wirelessly powered array of 128 centrifugo-pneumatic valves that can be thermally actuated on demand during spinning. The valves can either be triggered by a predefined protocol, wireless signal transmission via Bluetooth, or in response to a sensor monitoring a parameter like the temperature, or homogeneity of the dispersion. Upon activation of a resistive heater, a low-melting membrane (Parafilm™) is removed to vent an entrapped gas pocket, thus letting the incoming liquid wet an intermediate dissolvable film and thereby open the valve. The proposed system allows up to 12 heaters to be activated in parallel, with a response time below 3 s, potentially resulting in 128 actuated valves in under 30 s. We demonstrate, with three examples of common and standard procedures, how the proposed technology could become a powerful tool for implementing diagnostic assays on Lab-on-a-Disc. First, we implement wireless actuation of 64 valves during rotation in a freely programmable sequence, or upon user input in real time. Then, we show a closed-loop centrifugal flow control sequence for which the state of mixing of reagents, evaluated from stroboscopically recorded images, triggers the opening of the valves. In our last experiment, valving and closed-loop control are used to facilitate centrifugal processing of whole blood.
Controlling flow in microfluidic channels with a manually actuated pin valve

Abstract:There is a need for a simple method to control fluid flow within microfluidic channels. To meet this need, a simple push pin with a polydimethylsiloxane (PDMS) tip has been integrated into microfluidic networks to be placed within the microchannel to obstruct flow. This new valve design can attain on/off control of fluid flow without an external power source using readily-available, low-cost materials. The valve consists of a 14 gauge (1.6 mm) one inch piece of metal tubing with a PDMS pad at the tip to achieve a fluidic seal when pressed against a microfluidic channel’s substrate. The metal tubing or pin is then either manually pushed down to block or pulled up to allow fluid flow. The valve was validated using a pressure transducer and fluorescent dye to determine the breakthrough pressure the valve can withstand over multiple cycles. In the first cycle, the median value for pressure withstood by the valve was 8.8 psi with a range of 17.5–2.7 psi. The pressure the valves were able to withstand during each successive trial was lower suggesting they may be most valuable as a method to control the initial introduction of fluids into a microfluidic device. These valves can achieve flow regulation within microfluidic devices, have a small dead volume, and are simple to fabricate and use, making this technique widely suitable for a range of applications.

Energy-saving, High-Pressure Resistant Mini Valve Based on a Bistable Electromagnetic Actuator

Abstract:As core components of microfluidic and wearable pneumatic electronic systems, mini valves are attracting a growing intellectual interest. However, most existing mini valves exhibit either excessive energy consumption or limited flow rate and holding pressure. To address these issues, this study proposes an energy-saving mini valve with large flow rate and high holding pressure. The implementation of electromagnetic bistable structure allows the valve to remain open or closed without energy consumption. In contrast to the small and intricate flow channels seen in conventional mini valves, the straight-through flow channel design with reduced flow resistance improves its flow rate properties. And the pressure resistance characteristics are enhanced by the use of permanent magnetic attraction for sealing. Furthermore, the valve is driven by a single coil to switch between two steady states, resulting in a more compact structure. We have developed valve prototypes with diameters of 10 mm and 6 mm. Performance evaluation tests have shown that it sustains a holding pressure as high as 100 kPa and a flow rate of 1.5 L/min (@ 3 kPa), while only expending an energy of 0.37 J during switch transitions. Given these attributes, this valve demonstrates significant potential for integration within wearable pneumatic electronic systems.

Abstract:This article presents a simple approach for preparation of thermoswitchable capillary force valves on full PDMS microfluidic chips. The highly hydrophobic PDMS surface was tailored to hydrophilic or hydrophilicity/hydrophobicity thermoswitchable by surface grafting of poly (acrylic acid) (PAA) or Poly (N-isopropylacrylamide) (PNIPAAm) via benzophenone initiated surface photopolymerization techniques. A thermoswitcable capillary force valve with a hydrophobicity/hydrophilicity switchable PNIPAAm patch in otherwise hydrophilic PAA grafted PDMS channel was designed, and a two-step region-selective grafting method was established to prepare the valve inside bonded PDMS microchannels. With a micro Peltier heating/cooling device used to regulate the valve temperature, aqueous solution was allowed to pass the valve at room temperature while the steam could be stopped at the edge of the PNIPAAm patch that was at 40°C. The capability of the developed thermoswitchable capillary force valve for manipulation of aqueous flow was demonstrated on full PDMS chips with a T-shaped micro channel network.

Abstract:Trigger valves are fundamental features in capillary-driven microfluidic systems that stop fluid at an abrupt geometric expansion and release fluid when there is flow in an orthogonal channel connected to the valve. The concept was originally demonstrated in closed-channel capillary circuits. We show here that trigger valves can be successfully implemented in open channels. We also show that a series of open-channel trigger valves can be placed alongside or opposite a main channel resulting in a layered capillary flow. We developed a closed form model for the dynamics of the flow at trigger valves based on the concept of average friction length and successfully validated the model against experiments. For the main channel, we discuss layered flow behavior in the light of the Taylor-Aris dispersion theory and in the channel turns by considering Dean theory of mixing. This work has potential applications in autonomous microfluidics systems for biosensing, at-home or point-of-care sample preparation devices, hydrogel patterning for 3D cell culture and organ-on-a-chip models.

Abstract:A compact, rapid, and portable off-chip pneumatic control valve is significant for the miniaturization and integration of external pneumatic systems for microfluidic chips. In this work, an off-chip microvalve with a high-speed electromagnetic switch actuator and a polydimethylsiloxane (PDMS) material valve body has been designed to be easily encapsulated, simulated using MATLAB/Simulink software, and tested in a micromixer. Multi-physical coupling mathematical models are developed based on the elastic deformation force of the valve membrane, the driving force of the valve core, and the fluid force in the microchannel. Two single microvalves are used to form a three-way microvalve, which can control the air pressure in a pneumatic microchannel on the microfluidic chip. The relationship between the flow–duty cycle, the flow–pressure difference of the single electromagnetic microvalve, and the load pressure of the three-way microvalve is simulated and analyzed. Sample mixing performance controlled by the proposed off-chip three-way microvalve was tested to evaluate the pneumatic control capability, and the results show that the undertaking can fully satisfy the needs of a pneumatic microfluidic chip for most applications.

Microvalve-based devices for flow-free gradient generators

Abstract:Experiments with gradients of soluble bioactive species have significantly advanced with microfluidic developments that enable cell observation and stringent control of environmental conditions. While some methodologies rely on flow to establish gradients, other opt for flow-free conditions, which is particularly beneficial for studying non-adherent and/or shear-sensitive cells. In flow-free devices, bioactive species diffuse either through resistive microchannels in microchannel-based devices, a porous membrane in membrane-based devices, or a hydrogel in gel-based devices. However, despite significant advancements over traditional methods such as Boyden chambers, these technologies have not widely disseminated in biological laboratories, arguably due to entrenched practices and the intricate skills required for conducting microfluidic assays. Here, we integrated Quake-type pneumatic microvalves in place of microgrooves, membranes, or gels, and developed devices with precise control over residual flow, establishment initial gradient, and long-term stability of gradients. The Microvalve-based approach enables the generation of the automatization of delicate microfluidic manipulations, which paves the way for routine applications of controlled and tunable flow-free gradients in academic laboratories and biomedical units.

Abstract:This paper presents a novel method to fabricate a normally open micro pneumatic valve in rigid poly(methyl methacrylate) (PMMA) chips. The control and fluid substrates of the valve were prepared with PMMA sheets via hot embossing. After both control and fluid substrates were silanized with 3-aminopropyltriethoxysilane and treated with glow discharge plasma, each of them was irreversibly bonded to a plasma-treated PDMS membrane, forming a control half or a fluid half. Relying on the native adhesive force of the PDMS membrane, the control half and fluid half were then reversibly sealed to form a full microchip with the four-layer structure of PMMA-PDMA center dot center dot center dot PDMS-PMMA. The irreversible bonding between rigid PMMA substrate and elastic PDMS membrane in combination with the reversible sealing between the control half and fluid half not only eases the difficulties in chip bonding, but also allows the control half and fluid half to be replaced individually in routine analyses. The performance of the developed micro pneumatic valves was characterized, and potential applications of the valves in droplet generation and micro flow injection analysis were demonstrated.
A virtual valve for smooth contamination-free flow switching

Abstract:We present a channel geometry that allows for clean switching between different inlets of a microchip without any contamination of the inlets or the downstream flow. We drive this virtual valve with a pneumatic pressure setup that minimizes disturbance of the downstream flow during the switching procedure by simultaneous variation of the pressures applied to the different inlets. We assess the efficiency of the setup by spectroscopic measurement of downstream dye concentrations, and demonstrate its practical utility by sequentially constructing multiple layers of alginate hydrogel. The method is potentially useful for a whole series of further applications, such as changing perfusion liquids for cell culture and cell analysis, metering, chemical-reaction initiation and multi-sample chromatography, to name a few.
