Abstract:Self-assembled peptide-based nanomaterials have exhibited wide application potential in the fields of materials science, nanodevices, biomedicine, tissue engineering, biosensors, energy storage, environmental science, and others. Due to their porous structure, strong mechanical stability, high biocompatibility, and easy functionalization, three-dimensional self-assembled peptide hydrogels revealed promising potential in bio-related applications. To present the advances in this interesting topic, we present a review on the synthesis and functionalization of peptide hydrogels, as well as their applications in drug delivery, antibacterial materials, cell culture, biomineralization, bone tissue engineering, and biosensors. Specifically, we focus on the fabrication methods of peptide hydrogels through physical, chemical, and biological stimulations. In addition, the functional design of peptide hydrogels by incorporation with polymers, DNA, protein, nanoparticles, and carbon materials is introduced and discussed in detail. It is expected that this work will be helpful not only for the design and synthesis of various peptide-based nanostructures and nanomaterials, but also for the structural and functional tailoring of peptide-based nanomaterials to meet specific demands.

Abstract:Hydrogel materials are of great value in medical field due to their similarity to human soft tissue.With silk fibroin as raw materials,organic solvent induction method can be employed to shorten the gelling time of silk fibroin to several hours.Silk fibroin/collagen composite hydrogel prepared with carbodiimide (EDC) as activation cross-linking agent has a storage modulus of up to 10 kPa.In vivo experiments indicate that silk fibroin hydrogel created by enzymatic crosslinking has good biocompatibility and biodegradability.We first explain the gelling mechanism of silk fibroin hydrogels,and then systematically introduce the research progress on preparation and modification methods of various silk fibroin hydrogels.Besides,potential applications of silk fibroin hydrogels as novel biomaterial in clinical applications are analyzed.Basic theoretical and technological issues to be solved for clinical applications of silk fibroin hydrogels are also discussed.

Abstract:Protein hydrogels with three-dimensional network structure can be synthesized through both covalent and noncovalent cross-linking of protein monomers and assemblies, which have shown wide applications in various fields due to their low toxicity, higher biocompatibility, better bioactivity, and better flexibility compared to traditional inorganic and polymeric hydrogels. In this review, we present recent advances (2016 to current) in the synthesis, structural and functional regulation, as well as healthcare applications of protein hydrogels. We provide detailed information on the synthesis of protein hydrogels via chemical, physical, and biological stimulations, and then introduce their unique self-healing, stimuli-responsive, swelling, mechanical, biological, and injectable properties. Furthermore, we analyze the strategies that can be utilized to regulate the structures and functions of protein hydrogels through tailoring the coiled-coil, porous, and periodic structure, as well as adding various functional materials into protein hydrogels. Finally, the healthcare applications of protein-based hydrogels for wearable devices, biosensors, drug/gene delivery, disease treatment, tissue engineering, cancer therapy, and bioimaging are demonstrated and discussed. It is the first effort to demonstrate excellent contributions of protein hydrogels to healthcare fields, which will be valuable for the readers to design and synthesize functional bio-hydrogels rationally and further to develop novel hydrogels for advanced applications.

Abstract:Over the past few decades, polymeric hydrogels have gained significant progress in various fields. As the most biochemically and structurally functional macromolecules in biological organisms, proteins have structural and mechanical properties similar to that of natural extracellular matrices (ECM). By combination of polymers with protein, protein-polymer hydrogel can be constructed as biomaterials with diverse properties and possess a wide range of applications. In this review, we summarized the progress of research on protein-polymer hydrogels and introduced the common fabrication methods. The properties of the protein-polymer hydrogels were briefly described, including biocompatibility, adhesion properties, responsiveness, mechanical properties, swelling properties, and degradability. In addition, we also reviewed the applications of protein-polymer hydrogels in tissue engineering, drug delivery and encapsulation, wearable sensors, adsorption, and other applications. Finally, we summarize the research on protein-polymer hydrogels and highlight the current challenges and future prospects of the field, which may provide a meaningful reference for the field of protein-polymer hydrogels.

Abstract:In recent years, supramolecular hydrogels have made groundbreaking research progress in biomedical fields such as drug delivery, biosensing, imaging analysis, and tissue engineering. Peptides, with their unique characteristics of facile preparation, low immunogenicity and easy biodegradability, are commonly used as building blocks of supramolecular hydrogels. Peptide-based supramolecular hydrogels loaded with drugs, prepared via physical means or covalent crosslinking, exhibit unique three-dimensional network structures and strong water retention capacities. These properties enhance drug bioavailability and reduce side effects, enabling drug accumulation and responsive release at disease sites, significantly improving the therapeutic efficacy. Here, we review recent advancements in peptide-based supramolecular hydrogels and their biotherapeutic applications, including chemotherapy, photothermal therapy, photodynamic therapy, immunotherapy, gene therapy, antibacterial and anti-inflammatory treatments, and other biological applications. This review aims to provide new inspiration for the development of biomaterials in the therapeutic field and provide more personalized options for disease treatment. Additionally, challenges and limitations in this field are briefly discussed.

Abstract:Polyethylene glycol (PEG) is hydrophilic, biocompatible, and electrically neutral in an aqueous solution due to its unique internal structure. PEG is widely used to prepare hydrogels for applications in drug delivery and tissue engineering. In addition, PEG can be used as a functionalization agent of biomaterials to prolong the presence of drug systems in vivo in biomedical applications. The functionalization of PEG hydrogels using functional peptides gives PEG hydrogels the advantages of better biocompatibility, higher loading efficiency, more adjustable lifetime, and lower cost, which have exhibited promising applications in biomedical fields. In this review, recent advances in the functionalization of PEG hydrogels with peptides for biomedical applications are presented. For this aim, first, the basic properties of PEG hydrogels are described by introducing various preparation methods, and then, the functional regulation of PEG hydrogels is demonstrated with different motif-designed peptides to achieve specific properties. Finally, biomedical applications of peptide-engineered PEG hydrogels in controlled drug delivery, tissue engineering, medical diagnostics, bioimaging, and biosensing are described in detail. This work provides a systematic literature review on this promising topic and facilitates readers in the design of composite materials and functional tailoring of PEG hydrogels with biomolecules.

Abstract:Hydrogels are a class of soft and wet materials that typically consist of a three-dimensional polymer network that swells yet does not dissolve in water. There are extensive research and development of hydrogel materials for diverse applications over the past two decades. More recently, hydrogel films with controlled thickness have been fabricated based on crosslinked natural or synthetic polymer networks. These films not only show the chemical/physical properties of bulk gels, such as biocompatibility and stimuli responsiveness, but also exhibit faster response speed and high flexibility/adaptivity, allowing for new functions and applications. In this minireview, we summarize the recent advances in hydrogel films with focuses on two aspects: (i) methods for facile fabrication of hydrogel films, and (ii) versatile applications of hydrogel films in biomedical and emerging fields. Hydrogel films with controllable thickness, fast response, good compliance, and tunable mechanical properties are ideal materials as artificial muscles and wound dressing, and to construct soft actuators, flexible electronics, and so forth. Current challenges and future perspectives of hydrogel films are also given at the end of this minireview.

Abstract:The hierarchical formation of self-assembling peptide-based hydrogels (SAPHs) starts from peptide to nanofibers, following with the entanglement into hydrogels with nanofibrous network. Such characteristic structure and extraordinary biocompatibility, and the peptide components endow the SAPHs with diverse applications in biotechnological field. Therefore, the thorough comprehension of SAPHs is significant to broadening their application. In this review, fabrication, properties, and biological applications of the SAPHs are introduced, and the factors influencing the synthesis process as well as the properties of the SAPHs products are also systematically explained. Meanwhile, we conclude the problems to be solved and provide our perspective to the future development of SAPHs in the biotechnology.

Abstract:Dipeptide self-assembled hydrogels have potential biomedical applications because of their great biocompatibility, bioactivity, and tunable physicochemical properties, which can be modulated in the molecular level by design of amino acid sequences. Herein, a series of dipeptides (Fmoc-FL, -YL, -LL, and -YA) are designed to form shear-thinning hydrogels with self-healing and tunable mechanical properties by adjusting the synergetic effect of hydrophobic interactions (π-π stacking and hydrophobic effect) and hydrogen bonds of peptides through substitution of amino acid residues. The enhancement of hydrophobic interactions is a primary factor to promote mechanical rigidity of hydrogels, and strong hydrogen-bonding interactions between molecules contribute to the instantaneous self-healing property, which is supported by experimental studies (FTIR, CD, SEM, AFM, and rheology) and molecular dynamics simulations. The injectable dipeptide hydrogels were certified as an ideal endoscopic submucosal dissection filler to make operation convenient and secure in mice and living mini-pig's experiments with a longer duration time, higher stiffness, and lower inflammatory response than commercial clinical fillers.

Abstract:Self-assembled peptide-based hydrogels have shown great potential in bio-related applications due to their porous structure, strong mechanical stability, high biocompatibility, and easy functionalization. Herein, the structure and characteristics of hydrogels and the mechanism of action of several regular secondary structures during gelation are investigated. The factors influencing the formation of peptide hydrogels, especially the pH responsiveness and salt ion induction are analyzed and summarized. Finally, the biomedical applications of peptide hydrogels, such as bone tissue engineering, cell culture, antigen presentation, antibacterial materials, and drug delivery are reviewed.

Abstract:Peptide-based hydrogels have gained considerable attention as a compelling platform for various biomedical applications in recent years. Their attractiveness stems from their ability to seamlessly integrate diverse properties, such as biocompatibility, biodegradability, easily adjustable hydrophilicity/hydrophobicity, and other functionalities. However, a significant drawback is that most of the functional self-assembling peptides cannot form robust hydrogels suitable for biological applications. In this study, we present the synthesis of novel peptide-PEG conjugates and explore their comprehensive hydrogel properties. The hydrogel comprises double networks, with the first network formed through the self-assembly of peptides to create a β-sheet secondary structure. The second network is established through covalent bond formation via N-hydroxysuccinimide chemistry between peptides and a 4-arm PEG to form a covalently linked network. Importantly, our findings reveal that this hydrogel formation method can be applied to other peptides containing lysine-rich sequences. Upon encapsulation of the hydrogel with antimicrobial peptides, the hydrogel retained high bacterial killing efficiency while showing minimum cytotoxicity toward mammalian cells. We hope that this method opens new avenues for the development of a novel class of peptide-polymer hydrogel materials with enhanced performance in biomedical contexts, particularly in reducing the potential for infection in applications of tissue regeneration and drug delivery.

Abstract:The design and application of biomimetic hydrogels have become an important and integral part of modern tissue engineering and regenerative medicine. Many of these hydrogels are prepared from synthetic macromers (e.g., poly(ethylene glycol) or PEG) as they provide high degrees of tunability for matrix crosslinking, degradation, and modification. For a hydrogel to be considered biomimetic, it has to recapitulate key features that are found in the native extracellular matrix, such as the appropriate matrix mechanics and permeability, the ability to sequester and deliver drugs, proteins, and or nucleic acids, as well as the ability to provide receptor-mediated cell-matrix interactions and protease-mediated matrix cleavage. A variety of chemistries have been employed to impart these biomimetic features into hydrogel crosslinking. These chemistries, such as radical-mediated polymerizations, enzyme-mediated crosslinking, bio-orthogonal click reactions, and supramolecular assembly, may be different in their crosslinking mechanisms but are required to be efficient for gel crosslinking and ligand bioconjugation under aqueous reaction conditions. The prepared biomimetic hydrogels should display a diverse array of functionalities and should also be cytocompatible for in vitro cell culture and/or in situ cell encapsulation. The focus of this article is to review recent progress in the crosslinking chemistries of biomimetic hydrogels with a special emphasis on hydrogels crosslinked from poly(ethylene glycol)-based macromers.

Abstract:The injury of both central and peripheral nervous systems can result in neurological disorders and severe nervous diseases, which has been one of the challenges in the medical field. The use of peptide-based hydrogels for nerve repair and regeneration (NRR) provides a promising way for treating these problems, but the effects of the functions of peptide hydrogels on the NRR efficiency have been not understood clearly. In this review, we present recent advances in the material design, matrix fabrication, functional tailoring, and NRR applications of three types of peptide-based hydrogels, including pure peptide hydrogels, other component-functionalized peptide hydrogels, and peptide-modified polymer hydrogels. The case studies on the utilization of various peptide-based hydrogels for NRR are introduced and analyzed, in which the effects and mechanisms of the functions of hydrogels on NRR are illustrated specifically. In addition, the fabrication of medical NRR scaffolds and devices for pre-clinical application is demonstrated. Finally, we provide potential directions on the development of this promising topic. This comprehensive review could be valuable for readers to know the design and synthesis strategies of bioactive peptide hydrogels, as well as their functional tailoring, in order to promote their practical applications in tissue engineering, biomedical engineering, and materials science.

Abstract:Hydrogels are widely used in the field of biomedicine due to their high water content and strong drug loading capacity. Different types and quantities of amino acids are combined to form a wide variety of peptides. At the same time, the self-assembled peptide hydrogel is formed from the peptide into nanofibers, and the nanofibers are further wound to form a three-dimensional structure of the hydrogel. This special structure and the composition of the peptide give the hydrogel special characteristics and properties. Polypeptide-based self-assembled hydrogels have a variety of applications in the biomedical field. Therefore, it is of far-reaching significance to deeply understand polypeptide-based self-assembled hydrogels and explore their applications. This paper introduces the structure, preparation, properties, and biological applications of self-assembling peptide hydrogels. At the same time, the future development of peptide self-assembled gels is prospected.

Abstract:Microporous hydrogels have been utilized in an unprecedented manner in the last few decades, combining materials science, biology, and medicine. Their microporous structure makes them suitable for wide applications, especially as cell carriers in tissue engineering and regenerative medicine. Microporous hydrogel scaffolds provide spatial and platform support for cell growth and proliferation, which can promote cell growth, migration, and differentiation, influencing tissue repair and regeneration. This review gives an overview of recent developments in the fabrication techniques and applications of microporous hydrogels. The fabrication of microporous hydrogels can be classified into two distinct categories: fabrication of non-injectable microporous hydrogels including freeze-drying microporous method, two-phase sacrificial strategy, 3D biofabrication technology, etc., and fabrication of injectable microporous hydrogels mainly including microgel assembly. Then, the biomedical applications of microporous hydrogels in cell carriers for tissue engineering, including but not limited to bone regeneration, nerve regeneration, vascular regeneration, and muscle regeneration are emphasized. Additionally, the ongoing and foreseeable applications and current limitations of microporous hydrogels in biomedical engineering are illustrated. Through stimulating innovative ideas, the present review paves new avenues for expanding the application of microporous hydrogels in tissue engineering.

Abstract:<p>Nucleic acid‐based hydrogels that integrate intrinsic biological properties of nucleic acids and mechanical behavior of their advanced assemblies are appealing bioanalysis and biomedical studies for the development of new‐generation smart biomaterials. It is inseparable from development and incorporation of novel structural and functional units. This review highlights different functional units of nucleic acids, polymers, and novel nanomaterials in the order of structures, properties, and functions, and their assembly strategies for the fabrication of nucleic acid‐based hydrogels. Also, recent advances in the design of multifunctional and stimuli‐responsive nucleic acid‐based hydrogels in bioanalysis and biomedical science are discussed, focusing on the applications of customized hydrogels for emerging directions, including 3D cell cultivation and 3D bioprinting. Finally, the key challenge and future perspectives are outlined.</p>
