Abstract:Supramolecular peptide hydrogels have many important applications in biomedicine, including drug delivery applications for the sustained release of therapeutic molecules. Targeted and selective drug administration is often preferential to systemic drug delivery, as it can allow reduced doses and can avoid the toxicity and side-effects caused by off-target binding. New discoveries are continually being reported in this rapidly developing field. In this review, we report the latest developments in supramolecular peptide-based hydrogels for drug delivery, focusing primarily on discoveries that have been reported in the last four years (2018–present). We address clinical points, such as peptide self-assembly and drug release, mechanical properties in drug delivery, peptide functionalization, bioadhesive properties and drug delivery enhancement strategies, drug release profiles, and different hydrogel matrices for anticancer drug loading and release.

Abstract:Supramolecular peptide hydrogel (SPH) is a class of biomaterials self‐assembled from peptide‐based gelators through non‐covalent interactions. Among many of its biomedical applications, the potential of SPH in cancer therapy has been vastly explored in the past decade, taking advantage of its good biocompatibility, multifunctionality, and injectability. SPHs can exert localized cancer therapy and induce systemic anticancer immunity to prevent tumor recurrence, depending on the design of SPH. This review first gives a brief introduction to SPH and then outlines the major types of peptide‐based gelators that have been developed so far. The methodologies to tune the physicochemical properties and biological activities are summarized. The recent advances of SPH in cancer therapy as carriers, prodrugs, or drugs are highlighted. Finally, the clinical translation potential and main challenges in this field are also discussed.

Abstract:Self-assembled peptide-based nanomaterials have exhibited wide application potential in the fields of materials science, nanodevices, biomedicine, tissue engineering, biosensors, energy storage, environmental science, and others. Due to their porous structure, strong mechanical stability, high biocompatibility, and easy functionalization, three-dimensional self-assembled peptide hydrogels revealed promising potential in bio-related applications. To present the advances in this interesting topic, we present a review on the synthesis and functionalization of peptide hydrogels, as well as their applications in drug delivery, antibacterial materials, cell culture, biomineralization, bone tissue engineering, and biosensors. Specifically, we focus on the fabrication methods of peptide hydrogels through physical, chemical, and biological stimulations. In addition, the functional design of peptide hydrogels by incorporation with polymers, DNA, protein, nanoparticles, and carbon materials is introduced and discussed in detail. It is expected that this work will be helpful not only for the design and synthesis of various peptide-based nanostructures and nanomaterials, but also for the structural and functional tailoring of peptide-based nanomaterials to meet specific demands.

Abstract:Peptides are a class of active substances composed of a variety of amino acids with special physiological functions. The rational design of peptide sequences at the molecular level enables their folding into diverse secondary structures. This property has garnered significant attention in the biomedical sphere owing to their favorable biocompatibility, adaptable mechanical traits, and exceptional loading capabilities. Concurrently with advancements in modern medicine, the diagnosis and treatment of tumors have increasingly embraced targeted and personalized approaches. This review explores recent applications of self-assembled peptides derived from natural amino acids in chemical therapy, immunotherapy, and other adjunctive treatments. We highlighted the utilization of peptide hydrogels as delivery systems for chemotherapeutic drugs and other bioactive molecules and then discussed the challenges and prospects for their future application.

The aim of this review will be to present an updated report on the advancement of peptide-based hydrogels research activity in recent years in the field of anticancer drug delivery, antimicrobial and wound healing materials, 3D bioprinting and tissue engineering, and vaccines. Additionally, the biosensing applications of this key group of hydrogels will be discussed mainly focusing the attention on cancer detection.

Abstract:Although considered promising materials for assisting organ regeneration, few hydrogels meet the stringent requirements of clinical translation on the preparation, application, mechanical property, bioadhesion, and biocompatibility of the hydrogels. Herein, we describe a facile supramolecular approach for preparing gelatin hydrogels with a wide array of desirable properties. Briefly, we first prepare a supramolecular gelatin macromer via the efficient host-guest complexation between the aromatic residues of gelatin and free diffusing photo-crosslinkable acrylated β-cyclodextrin (β-CD) monomers. The subsequent crosslinking of the macromers produces highly resilient supramolecular gelatin hydrogels that are solely crosslinked by the weak host-guest interactions between the gelatinous aromatic residues and β-cyclodextrin (β-CD). The obtained hydrogels are capable of sustaining excessive compressive and tensile strain, and they are capable of quick self healing after mechanical disruption. These hydrogels can be injected in the gelation state through surgical needles and re-molded to the targeted geometries while protecting the encapsulated cells. Moreover, the weak host-guest crosslinking likely facilitate the infiltration and migration of cells into the hydrogels. The excess β-CDs in the hydrogels enable the hydrogel-tissue adhesion and enhance the loading and sustained delivery of hydrophobic drugs. The cell and animal studies show that such hydrogels support cell recruitment, differentiation, and bone regeneration, making them promising carrier biomaterials of therapeutic cells and drugs via minimally invasive procedures.

Abstract:Therapeutic proteins, such as growth factors (GFs), have been used in tissue engineering (TE) approaches for their ability to provide signals to cells and orchestrate the formation of functional tissue. However, to be effective and minimize off-target effects, GFs should be delivered at the target site with temporal control. In addition, protein drugs are typically sensitive water soluble macromolecules with delicate structure. As such, hydrogels, containing large amounts of water, provide a compatible environment for the direct incorporation of proteins within the hydrogel network, while their release rate can be tuned by engineering the network chemistry and density. Being formed by transient crosslinks, afforded by non-covalent interactions, supramolecular hydrogels offer important advantages for protein delivery applications. This review describes various types of supramolecular hydrogels using a repertoire of diverse building blocks, their use for protein delivery and their further application in TE contexts. By reviewing the recent literature on this topic, the merits of supramolecular hydrogels are highlighted as well as their limitations, with high expectations for new advances they will provide for TE in the near future.

Abstract:Peptide‐based hydrogel biomaterials have emerged as an excellent strategy for immune system modulation. Peptide‐based hydrogels are supramolecular materials that self‐assemble into various nanostructures through various interactive forces (i.e., hydrogen bonding and hydrophobic interactions) and respond to microenvironmental stimuli (i.e., pH, temperature). While they have been reported in numerous biomedical applications, they have recently been deemed as promising candidates to improve the efficacy of cancer immunotherapies and treatments. Immunotherapies seek to harness the body's immune system to preemptively protect against and treat various diseases, such as cancer. However, their low efficacy rates result in limited patient responses to treatment. Here, the immunomaterial's potential to improve these efficacy rates by either functioning as immune stimulators through direct immune system interactions and/or delivering a range of immune agents is highlighted. The chemical and physical properties of these peptide‐based materials that lead to immune‐modulation and how one may design a system to achieve desired immune responses in a controllable manner are discussed. Works in the literature that reports peptide hydrogels as adjuvant systems and for the delivery of immunotherapies are highlighted. Abstract:Self-assembled peptide-based hydrogels have shown great potential in bio-related applications due to their porous structure, strong mechanical stability, high biocompatibility, and easy functionalization. Herein, the structure and characteristics of hydrogels and the mechanism of action of several regular secondary structures during gelation are investigated. The factors influencing the formation of peptide hydrogels, especially the pH responsiveness and salt ion induction are analyzed and summarized. Finally, the biomedical applications of peptide hydrogels, such as bone tissue engineering, cell culture, antigen presentation, antibacterial materials, and drug delivery are reviewed.

Abstract:Self-assembly is a growth mechanism in nature to apply local interactions forming a minimum energy structure. Currently, self-assembled materials are considered for biomedical applications due to their pleasant features, including scalability, versatility, simplicity, and inexpensiveness. Self-assembled peptides can be applied to design and fabricate different structures, such as micelles, hydrogels, and vesicles, by diverse physical interactions between specific building blocks. Among them, bioactivity, biocompatibility, and biodegradability of peptide hydrogels have introduced them as versatile platforms in biomedical applications, such as drug delivery, tissue engineering, biosensing, and treating different diseases. Moreover, peptides are capable of mimicking the microenvironment of natural tissues and responding to internal and external stimuli for triggered drug release. In the current review, the unique characteristics of peptide hydrogels and recent advances in their design, fabrication, as well as chemical, physical, and biological properties are presented. Additionally, recent developments of these biomaterials are discussed with a particular focus on their biomedical applications in targeted drug delivery and gene delivery, stem cell therapy, cancer therapy and immune regulation, bioimaging, and regenerative medicine.

Abstract:Polyethylene glycol (PEG) is hydrophilic, biocompatible, and electrically neutral in an aqueous solution due to its unique internal structure. PEG is widely used to prepare hydrogels for applications in drug delivery and tissue engineering. In addition, PEG can be used as a functionalization agent of biomaterials to prolong the presence of drug systems in vivo in biomedical applications. The functionalization of PEG hydrogels using functional peptides gives PEG hydrogels the advantages of better biocompatibility, higher loading efficiency, more adjustable lifetime, and lower cost, which have exhibited promising applications in biomedical fields. In this review, recent advances in the functionalization of PEG hydrogels with peptides for biomedical applications are presented. For this aim, first, the basic properties of PEG hydrogels are described by introducing various preparation methods, and then, the functional regulation of PEG hydrogels is demonstrated with different motif-designed peptides to achieve specific properties. Finally, biomedical applications of peptide-engineered PEG hydrogels in controlled drug delivery, tissue engineering, medical diagnostics, bioimaging, and biosensing are described in detail. This work provides a systematic literature review on this promising topic and facilitates readers in the design of composite materials and functional tailoring of PEG hydrogels with biomolecules.

Abstract:The self-assembly of low-molecular-weight building motifs into supramolecular polymers has unlocked a new realm of materials with distinct properties and tremendous potential for advancing medical practices. Leveraging the reversible and dynamic nature of non-covalent interactions, these supramolecular polymers exhibit inherent responsiveness to their microenvironment, physiological cues, and biomolecular signals, making them uniquely suited for diverse biomedical applications. In this review, we intend to explore the principles of design, synthesis methodologies, and strategic developments that underlie the creation of supramolecular polymers as carriers for therapeutics, contributing to the treatment and prevention of a spectrum of human diseases. We delve into the principles underlying monomer design, emphasizing the pivotal role of non-covalent interactions, directionality, and reversibility. Moreover, we explore the intricate balance between thermodynamics and kinetics in supramolecular polymerization, illuminating strategies for achieving controlled sizes and distributions. Categorically, we examine their exciting biomedical applications: individual polymers as discrete carriers for therapeutics, delving into their interactions with cells, and in vivo dynamics; and supramolecular polymeric hydrogels as injectable depots, with a focus on their roles in cancer immunotherapy, sustained drug release, and regenerative medicine. Abstract:Protein hydrogels with three-dimensional network structure can be synthesized through both covalent and noncovalent cross-linking of protein monomers and assemblies, which have shown wide applications in various fields due to their low toxicity, higher biocompatibility, better bioactivity, and better flexibility compared to traditional inorganic and polymeric hydrogels. In this review, we present recent advances (2016 to current) in the synthesis, structural and functional regulation, as well as healthcare applications of protein hydrogels. We provide detailed information on the synthesis of protein hydrogels via chemical, physical, and biological stimulations, and then introduce their unique self-healing, stimuli-responsive, swelling, mechanical, biological, and injectable properties. Furthermore, we analyze the strategies that can be utilized to regulate the structures and functions of protein hydrogels through tailoring the coiled-coil, porous, and periodic structure, as well as adding various functional materials into protein hydrogels. Finally, the healthcare applications of protein-based hydrogels for wearable devices, biosensors, drug/gene delivery, disease treatment, tissue engineering, cancer therapy, and bioimaging are demonstrated and discussed. It is the first effort to demonstrate excellent contributions of protein hydrogels to healthcare fields, which will be valuable for the readers to design and synthesize functional bio-hydrogels rationally and further to develop novel hydrogels for advanced applications.

Abstract:Supramolecular hydrogels bound by hydrogen bonding, host-guest, hydrophobic, and other non-covalent interactions are among the most attractive biomaterials available. Supramolecular hydrogels have attracted extensive attention due to their inherent dynamic reversibility, self-healing, stimuli-response, excellent biocompatibility, and near-physiological environment. However, the inherent contradiction between non-covalent interactions and mechanical strength makes the practical application of supramolecular hydrogels a great challenge. This review describes the mechanical strength of hydrogels mediated by supramolecular interactions, and focuses on the potential strategies for enhancing the mechanical strength of supramolecular hydrogels and illustrates their applications in related fields, such as flexible electronic sensors, wound dressings, and three-dimensional (3D) scaffolds. Finally, the current problems and future research prospects of supramolecular hydrogels are discussed. This review is expected to provide insights that will motivate more advanced research on supramolecular hydrogels.

Abstract:Due to the similarity to extracellular matrix, hydrogels have been widely used in biomedical field such as drug delivery, tissue engineering and in vitro cell culture. Deoxyribonucleic acid (DNA) is known for the clear chemical composition, designable sequence and unique structural rigidity, which makes DNA an ideal material for construction of hydrogels. Based on our research, this review summarizes recent developments in DNA supramolecular hydrogels. Firstly, the construction strategies including the pure and hybrid DNA supramolecular hydrogels are introduced. While the construction of pure DNA supramolecular hydrogels is mainly based on base pairing or physical entanglement, covalent or non-covalent doping is also demonstrated to play important roles in preparing the hybrid hydrogels. Then, the physical properties of the DNA supramolecular hydrogels are discussed, such as the excellent permeability, tunable mechanical strength and dynamic properties. Next, the functionalization strategies are also summarized, including sequence design, chemical modification and network interpenetration. Furthermore, the applications of DNA supramolecular hydrogels in biomedicine are introduced, focusing on three-dimensional cell culture and printing, tissue engineering, immunotherapy and drug delivery. Finally, some challenges and future perspectives in the development of DNA supramolecular hydrogels are discussed. Abstract:Abstract Soft materials, such as hydrogels, are used as scaffolds in tissue engineering and regenerative medicine to help tissues regenerate and heal. Recently, supramolecular hydrogels, based on non-covalent interactions, have grown in popularity, especially in the development of materials for biomedical use. Their potential lies in the dynamic, reversible and temporary nature of their crosslinks, which can make them responsive to stimuli, injectable and suitable for 3D printing. Such versatility and processability is important when developing new biomaterials for drug delivery or as implantable scaffolds. The behavior and properties of such hydrogels are different compared to those of chemically crosslinked hydrogels. In this review, we give an overview on supramolecular hydrogels which contain hyaluronic acid (HA) as one of the building blocks. HA is particularly interesting, due to its hydrophilicity, biofunctionality and ease of chemical modification. Specifically, we focus on HA-based hydrogels that make use of hydrogen bonding, hydrophobic interactions, electrostatic interactions, metal–ion coordination and guest–host interactions, and are intended for applications in the biomedical field, with potential for clinical translation.

Abstract:Supramolecular hydrogels of self-assembling peptide-drug conjugates have been considered as effective self-delivery drug systems for cancer therapy in recent years. Here, a novel self-assembling peptide-based supramolecular hydrogel was developed by simultaneously conjugating small-molecule drug chlorambucil (CRB) and peptide drug tyroservatide (YSV) to the self-assembling peptide. The resulting hydrogel with a nanofiber structure showed enhanced stability against proteinase K degradation and an improved cellular uptake performance in comparison with the free molecules. As a consequence, it exhibited enhanced antitumor efficiency both in vitro and in vivo with favorable biocompatibility. This biocompatible self-delivery drug system could not only significantly improve the delivery efficiency of the small-molecule drugs but also adequately synergize the antitumor effect of CRB and YSV, inspiring the design of new strategies of cancer combination therapy.

Abstract:A short bioinspired octapeptide, GV8, can self-assemble under mild conditions into biodegradable supramolecular physical hydrogels with high storage modulus and good biocompatibility. GV8 hydrogels can encapsulate both single or multiple macromolecular protein-based therapeutics in a simple one-pot formulation manner, making it a promising candidate to address challenges faced by existing synthetic polymer or peptide hydrogels with complex gelation and drug-encapsulation processes. Alongside its versatility, the hydrogel exhibits concentration-dependent storage modulus and controlled drug-release action. We demonstrate that GV8 hydrogels loaded with adipose-derived mesenchymal stem cells (ADMSC) secretome remain mechanically robust, and exhibit promising potential for wound healing applications by preserving secretome activity while maintaining a constant supply of ADMSC secretome to promote epithelial cell migration. Overall, our work highlights the potential of GV8 peptide hydrogel as a versatile and safe carrier for encapsulation and delivery of macromolecular therapeutics.Statement of Significance: Supramolecular peptide hydrogels are a popular choice for protein-based macromolecular therapeutics delivery; however, despite the development of abundant hydrogel systems, several challenges limit their adaptability and practical applications. GV8 short peptide hydrogel circumvents these drawbacks and demonstrates the ability to function as a versatile growth factor (GF) encapsulant. It can encapsulate precise concentrations of complex adipose-derived mesenchymal stem cells secretome mixtures with a one-pot formulation approach and perform controlled release of GFs with preserved activity without compromising the self-assembly properties and mechanical properties of the hydrogel's supramolecular network. The significance of GV8 hydrogel lies in its gelation simplicity and versatility to encapsulate and deliver macromolecular therapeutics, thus representing a promising biomaterial for regenerative medicine applications.
