Abstract:A short and facile synthesis of a series of 1,2-dihydroquiazolin-4(3H)-ones was accomplished in good yields via the novel reductive cyclisation of o-nitrobenzamides with aldehydes or ketones promoted by the TiCl4/Sm system. Structures were established on the basis of elemental analysis, IR, H-1 NMR and confirmed by a single-crystal X-ray diffraction analysis. The advantages of our method are easily accessible starting materials, convenient manipulation and moderate to high yields.

Abstract:Under the action of a low-valent titanium reagent, substituted imidazolidines were synthesized in moderate yields via cross-coupling between imines and triethyl orthoformate, and the yields of product 3 with different substituted groups as well as the corresponding isomer distributions were also discussed. Related NMR spectra were used to define the configuration of the products.

Abstract:This paper reports the reactions of o-nitrobenzamides with ketones induced by titanium reagent (TICl4-SM system). The results show that the low-valent titanium promoted a novel reductive cyclization of o-nitrobenzamides and ketones to give 1, 2-dihydroquinazohn-4 3H)-ones. The products were characterized by elementary analysis, IR, H-1 NMR and X-ray single crystal diffraction. The crystal of compound 3b belongs to monoclinic system with space group P2(1)/n, a=1.191 7(2) nm, b=0.691 10) nm, c= 1.782 1(4) nm, beta=98.81degrees, V=1.455 04(5) nm(3), Z=4, D-c=1.220 g/cm(3), mu=0.077 mm(-1), F(000) =568, and final R=0.043 3 and wR=0.107 6 for reflections [I>2sigma(I)]. In summary, a series of 1,2-dihydroquinazolin-4 3H) -ones was synthesized via reductive cyclization of o-nitrobenzamides with ketones induced by the TiCl4-Sm system. Our method possesses the advantages of easily accessible starting materials, convenient manipulation and moderate to high yield.

Abstract:We report the concise and protecting-group-free enantioselective total syntheses of circumdatins F and H. In view of the extreme importance of analogs of quinazolinone alkaloids in drug research and discovery, four analogs of bioactive quinazolinobenzodiazepine alkaloids, including demethoxycircumdatin H (12) and N-demethyl-benzomalvin A (13), have been synthesized. The method is based on the low-valent titanium-promoted intramolecular reductive coupling of imides with o-nitrobenzimides, which yielded quinazolino[3,2-a][1,4] benzodiazepines under mild conditions. In addition, heptacyclic dehydraasperlicin E (16) has been synthesized from asperlicin C by a NCS-mediated dehydra-cyclization reaction.

Abstract:2-Fluoro-5-(trifluoromethyl)aniline was found to be a suitable monodentate transient directing group (MonoTDG) to enable Ru(II)-catalyzed intermolecular direct ortho-C(sp2)–H imidation of benzaldehydes. N-Tosyloxyphthalimide was used as an alternative azide-free amidation reagent to achieve high efficiency and good functional group tolerance. Abstract:An iodine‐promoted one‐pot cascade oxidative annulation reaction has been developed for the synthesis of [1,2,3]triazolo[1,5‐a]quinolines from methyl azaarenes and N‐tosylhydrazines. The reaction has a broad substrate scope and can be easily scaled up to gram‐scale. 1,2,3‐Triazoles are an important skeletal structure for the construction of C‐C and C‐P bonds, 2‐cyclopropylquinolines and imidazo[1,5‐a]quinolines, for which different synthetic applications were explored.

Abstract:An efficient and simple one-pot method was developed for the synthesis of two kinds of N-fused quinazolinone derivatives using nano TiO2 particles as catalyst from isatoic anhydride, 2-carboxy benzaldehyde, hydrazides or aromatic hydrazines. Six quinazolino [2,1-b] phthalazin-8-ones and ten 3-acylamino isoindolo [2,1-a] quinazolines were obtained in excellent yields, and the reaction refluxed in aqueous ethanol were environmentally friendly. Attractive features of this methodology were its versatility, mild condition and high efficiency in creating two different structures in a single operation.

Abstract:A linear-organic-polymer-supported iridium complex Cp*, which is designed and synthesized by the coordinative immobilization of [Cp*IrCl2]2 on poly(4-vinylpyridine), was proven to be an efficient heterogeneous autotandem catalyst for synthesizing quinazolinones via selective hydration/acceptorless dehydrogenative coupling from o-aminobenzonitriles.
