Abstract:The rapid development of single-photon detection technology offers the possibility for an imaging system to go beyond traditional limits. Recently, single-photon imagers based on the upconversion technique have achieved unprecedentedly high resolution and noise tolerance in 3-D imaging. Nevertheless, the limited imaging range, complicated optical path, and long data acquisition time impose remaining challenges to practical applications. In this work, we build an upconversion single-photon imager for full-range and accurate 3-D imaging. A compact all-fiber design allows our system to work robustly in an open environment. Compared to the previous imagers, the imaging speed is increased by three orders of magnitude due to the high upconversion efficiency and photon-efficient algorithm. With the application of a high-repetition laser and a nanosecond optical delay line (ODL), we achieve full-range imaging under the upconversion scheme against high solar background noise. For close-up scenes, our imager achieves ultrahigh axial resolution up to 20 mu m, which is not affected by the time jitter or pileup effect of the single-photon detector. Our proposal can stimulate practical applications in sophisticated scenarios.

Abstract:Mode-locked Yb-doped fiber lasers around 1 μm are attractive for high power applications and low noise pulse train generation. Mode-locked fiber lasers working in soliton and stretched-pulse regime outperform others in terms of the laser noise characteristics, mechanical stability and easy maintenance. However, conventional optical fibers always show a normal group velocity dispersion around 1 μm, leading to the inconvenience for necessary dispersion management. Here we show that optical microfibers having a large anomalous dispersion around 1 μm can be integrated into mode-locked Yb-doped fiber lasers with ultralow insertion loss down to −0.06 dB, enabling convenient dispersion management of the laser cavity. Besides, optical microfibers could also be adopted to spectrally broaden and to dechirp the ultrashort pulses outside the laser cavity, giving rise to a pulse duration of about 110 fs. We believe that this demonstration may facilitate all-fiber format high-performance ultrashort pulse generation at 1 μm and may find applications in precision measurements, large-scale facility synchronization and evanescent-field-based optical sensing.

Abstract:Developing compact and stable 2 $\mu \text{m}$ ultrafast laser sources is important for generating mid-inferred frequency combs. Here, we report on a simple, compact and reliable thulium-doped fiber laser based on figure-of-9 mode-locking. Self-starting and stable mode-locked operation is achieved with 0.30 nJ pulse energy and 128 fs pulse width under 121.3 MHz repetition rate. The generated pulses centered at 1990 nm have a 3 dB bandwidth of 39.4 nm. The laser has been assembled into a $197\times 78\times 20$ mm3 fiber box under temperature-control. The repetition rate was phase locked over 20 h with a fluctuation standard deviation of 192.5 $\mu $ Hz. Such compact and highly-reliable mid-inferred laser could be used beyond laboratory and promote numerous critical applications including dual-comb spectroscopy, ranging and frequency calibration.

Simultaneous Generation of Ultrabroadband Noise-Like Pulses and Intracavity Third Harmonic at 2 Μm

Abstract:We demonstrated that ultrabroadband noise-like pulses (NLPs) spanning from below 1600 nm to beyond 2300 nm can be generated in Tm-doped fiber lasers enabled by an optical microfiber. Meanwhile, pronounced red light around 660 nm was also observed, which was attributed to the intracavity third harmonic generation (THG) of ultrashort pulses by harnessing the intermodal phase matching in the optical microfiber. As far as we know, it is the first time to simultaneously observe the ultrabroadband NLPs and the intracavity THG in ultrafast fiber lasers, and it is anticipated that these ultrabroadband NLPs can be adopted as a compact broadband source for optical spectroscopy around 2 µm and a wonderful potential seed for supercontinuum generation in the mid-infrared. Moreover, the THG of the intracavity high peak power NLPs in ultrafast fiber lasers provides a new kind of fiber-format visible light source.
Efficient, broadly-tunable source of megawatt pulses for multiphoton microscopy based on self-phase modulation in argon-filled hollow-core fiber

Abstract:An exciting recent development for deep-tissue imaging with cellular resolution is three-photon fluorescence microscopy (3PM) with excitation at long wavelengths (1300 and 1700 nm). In the last few years, long-wavelength 3PM has driven rapid progress in deep-tissue imaging beyond the depth limit of two-photon microscopy, with impacts in neuroscience, immunology, and cancer biology. However, wide adoption of 3PM faces challenges. Three-photon excitation (3PE) is naturally weaker than two-photon excitation, which places a premium on ultrashort pulses with high peak power. The inefficiency, complexity, and cost of current sources of these pulses present major barriers to the use of 3PM in typical biomedical research labs. Here, we describe a fiber-based source of femtosecond pulses with multi-megawatt peak power, tunable from 850 nm to 1700 nm. Compressed pulses from a fiber amplifier at 1030~nm are launched into an antiresonant hollow-core fiber filled with argon. By varying only the gas pressure, pulses with hundreds of nanojoules of energy and sub-100 fs duration are obtained at wavelengths between 850 and 1700 nm. This approach is a new route to an efficient, robust, and potentially low-cost source for multiphoton deep-tissue imaging. In particular, 960-nJ and 50-fs pulses are generated at 1300 nm with a conversion efficiency of 10\%. The nearly 20-MW peak power is an order of magnitude higher than the previous best from femtosecond fiber source at 1300~nm. As an example of the capabilities of the source, these pulses are used to image structure and neuronal activity in mouse brain as deep as 1.1 mm below the dura.

Abstract:Compact and high-energy femtosecond fiber lasers operating around 900-950 nm are desirable for multiphoton microscopy. Here, we demonstrate a >40 nJ, sub-100 fs, wavelengthtunable ultrafast laser system based on chirped pulse amplification (CPA) in thulium-doped fiber and second-harmonic generation (SHG) technology. Through effective control of the nonlinear effect in the CPA process, we have obtained 92-fs pulses at 1903 nm with an average power of 0.89W and a pulse energy of 81 nJ. By frequency doubling, 95-fs pulses at 954 nm with an average power of 0.46 W and a pulse energy of 42 nJ have been generated. In addition, our system can also achieve tunable wavelength from 932 nm to 962 nm (frequency doubled from 1863 nm to 1919 nm). A pulse width of similar to 100 fs and sufficient pulse energy are ensured over the entire tuning range. Finally, we applied the laser in a two-photon microscope and obtained superior imaging results. Due to a relatively low repetition rate (similar to 10 MHz), similar imaging quality can be achieved at significantly reduced average power compared with a commercial 80MHz laser system. At the same time, the lower average power is helpful in limiting the thermal load to the samples. It is believed that such a setup, with its well-balanced optical characteristics and compact footprint, provides an ideal source for two-photon microscopy. Abstract:We report an all-fiberized 1840-nm thulium-fiber-laser source, comprising a dissipative-soliton mode-locked seed laser and a chirped-pulse-amplification system for label-free biological imaging through nonlinear microscopy. The mode-locked thulium fiber laser generated dissipative-soliton pulses with a pre-chirped duration of 7 ps and pulse energy of 1 nJ. A chirped-pulse fiber-amplification system employing an in-house-fabricated, short-length, single-mode, high-absorption, thulium fiber delivered pulses with energies up to 105 nJ. The pulses were capable of being compressed to 416 fs by passing through a grating pair. Imaging of mouse tissue and human bone samples was demonstrated using this source via third-harmonic generation microscopy.

Abstract:We demonstrate a compact and efficient femtosecond laser operating at ~950 nm by directly frequency-doubling of a mode-locked Tm-doped fiber laser. The laser is based on a highly compact architecture, with a dispersion-managed oscillator together with only one-step frequency conversion. The output pulse width is ~270 fs, with a repetition rate of 22.1 MHz and a pulse energy of 12.9 nJ. It is believed that the greatly simplified setup makes the current laser well suited for two-photon microscopy and other biomedical and clinical applications.
Low-cost Custom Fabrication and Mode-locked Operation of an All-normal-dispersion Femtosecond Fiber Laser for Multiphoton Microscopy

Abstract:A protocol is presented to build a custom low-cost yet high-performance femtosecond (fs) fiber laser. This all-normal-dispersion (ANDi) ytterbium-doped fiber laser is built completely using commercially available parts, including $8,000 in fiber optic and pump laser components, plus $4,800 in standard optical components and extra-cavity accessories. Researchers new to fiber optic device fabrication may also consider investing in basic fiber splicing and laser pulse characterization equipment (~$63,000). Important for optimal laser operation, methods to verify true versus apparent (partial or noise-like) mode-locked performance are presented. This system achieves 70 fs pulse duration with a center wavelength of approximately 1,070 nm and a pulse repetition rate of 31 MHz. This fiber laser exhibits the peak performance that may be obtained for an easily assembled fiber laser system, which makes this design ideal for research laboratories aiming to develop compact and portable fs laser technologies that enable new implementations of clinical multiphoton microscopy and fs surgery.

Abstract:An ytterbium-doped, single-stage, double-pass nonlinear fiber amplification system was fabricated for amplifying an 1100-nm mode-locking fiber laser. Pre-chirp managed amplification (PCMA) was applied in realizing the nonlinear amplification process with an all-polarization-maintaining (PM) fiber construction. The system can deliver 19.8-nJ, 58.7-fs, 24.4-MHz amplified signal pulses with a 10-dB spectral range spanning from 1049 nm to 1130 nm. Further experimental investigations were conducted in exploring the dynamics of the double-pass nonlinear amplification process. This compact 1100-nm ultrafast fiber laser can be implemented for multi-photon microscopy (MPM) with deep penetration depth.

Abstract:Fiber lasers operating at 1.7 μm have very important applications in biomedicine, optical imaging, laser welding, optical communication and other fields because of their rich spectral characteristics in the near-infrared band. We designed and experimentally implemented a 1.7 μm all-fiber figure-9 (F9) mode-locked laser, with a fiber Bragg grating (FBG) acting as both the mirror and the spectrum filter. The all-fiber F9 design made the laser work in the mode-locking state more efficiently. We obtained mode-locked pulses with a central wavelength of 1724.76 nm and a repetition rate of 14.39 MHz when the pump power was 1.1 W, and the pulse width was about 54 ps. Limited by the bandwidth of the FBG, the 3 dB bandwidth of the mode-locked spectrum was about 0.18 nm. The output power was 52 mW at a pump power of 2.5 W. The multi-pulse dynamics were studied by adjusting the pump power and the polarization controllers, and pulse trains of up to six pulses in a group were achieved. The 1.7 μm narrow-bandwidth all-fiber F9 mode-locked laser is simple in structure and easy to build, with potential application as a seed source in high-energy ultrashort pulse lasers.

Abstract:We demonstrate a robust high power 930 nm femtosecond Nd:fiber laser system with hollow-core photonic bandgap fiber (HC-PBGF) as the output delivery, which can be easily integrated into compact two-photon microscopy system for bio-imaging. The whole laser system can deliver up to 17.4 nJ, 220-fs pulses at 930 nm with repetition rate of 46 MHz. In this paper, this laser was demonstrated as the light source for volumetric imaging of zebrafish blood vessel.

Abstract:Solid-state femtosecond lasers have stimulated the broad adoption of multiphoton microscopy in the modern laboratory. However, these devices remain costly. Fiber lasers offer promise as a means to inexpensively produce ultrashort pulses of light suitable for nonlinear microscopy in compact, robust and portable devices. Although encouraging, the initial methods reported in the biomedical engineering community to construct home-built femtosecond fiber laser systems overlooked fundamental aspects that compromised performance and misrepresented the significant financial and intellectual investments required to build these devices. Here, we present a practical protocol to fabricate an all-normal-dispersion ytterbium (Yb)-doped femtosecond fiber laser oscillator using commercially-available parts (plus standard optical components and extra-cavity accessories) as well as basic fiber splicing and laser pulse characterization equipment. We also provide a synthesis of established protocols in the laser physics community, but often overlooked in other fields, to verify true versus seemingly (partial or noise-like) mode-locked performance. The approaches described here make custom fabrication of femtosecond fiber lasers more accessible to a wide range of investigators and better represent the investments required for the proper laser design, fabrication and operation.

Abstract:By combining nonlinear polarization rotation (NPR) and semiconductor saturable absorber, we report a hybrid mode-locked Er:ZBLAN fiber oscillator at 2.8 µm. Stable 325-fs mode-locked pulses with an average power of 131 mW and a record signal-to-noise ratio of 79 dB at the fundamental frequency of 55.4 MHz are generated. Numerical simulations are carried out based on the modified coupled nonlinear Schrödinger equations, and offer new insights into the underlying dynamics of pulse generation. The simulations indicate that compared with Er:ZBLAN fiber lasers mode-locked by NPR alone, the hybrid mode-locked Er:ZBLAN fiber oscillator allows a wider range and a lower threshold of the pump power while maintaining the ultrashort pulse width. Moreover, we numerically demonstrate that the hybrid mode-locked oscillator is less sensitive to the variation of polarization states, which will increase its robustness against environmental disturbance. This is the first time that the hybrid mode-locking technique is applied in the mid-infrared, opening up new opportunities for the development of stable ultrafast mid-infrared laser sources and practical applications outside the laboratory.
Compact two-photon fluorescence microscope based on a single-mode fiber coupler

Abstract:We present a two-photon fluorescence microscope based on a three-port single-mode optical fiber coupler. It is found that the coupler behaves as a low-pass filter that can deliver an ultrashort-pulsed laser beam of as much as 150 mW of power in the wavelength range from 770 to 870 nm as well as collect a two-photon fluorescence signal in the visible range. As a result of using the fiber coupler, the new two-photon imaging system exhibts a number of advantages, including a compact arrangement, freedom from vibration from lasers and electronic devices, self-alignment, reduction of multiple scattering, and an enhanced optical sectioning effect. The effectiveness of the new instrument is demonstrated with a set of three-dimensional images of biological samples. This instrument may make two-photon fluorescence endoscopy possible for in vivo medical applications.

Abstract:We have demonstrated widely tunable Yb:fiber-based laser sources, aiming to replace Ti:sapphire lasers for the nJ-level ultrafast applications, especially for the uses of nonlinear light microscopy. We investigated the influence of different input parameters to obtain an expansive spectral broadening, enabled by self-phase modulation and further reshaped by self-steepening, in the normal dispersion regime before the fiber damage. We also discussed the compressibility and intensity fluctuations of the demonstrated pulses, to reach the transform-limited duration with a very low intensity noise. Most importantly, we have demonstrated clear two-photon fluorescence images from UV-absorbing fluorophores to deep red dye stains.

Abstract:We demonstrate a compact ultrafast source centered at 850 nm with >200 nm bandwidth (full width at half-maximum) based on a 3 GHz Yb-fiber master-oscillator-power-amplifier system. The output pulses (with up to 13 W average power) from the laser system are coupled into short (<50 mm) pieces of photonic crystal fibers to excite broadband fiber-optic Cherenkov radiation; the resulting broad phase-matching bandwidth due to short fiber length produces an upconverted spectrum spanning in the wavelength range of 750-950 nm with average power of 94, 184, and 380 mW for fiber length of 28, 37, and 48 mm, respectively. The spectrum generated from the 37 mm fiber is then dechirped by eight double-chirped mirrors, leading to compressed pulses ~14 fs in duration. Such an ultrafast source is a promising substitute of multigigahertz mode-locked Ti:sapphire lasers for applications in optical frequency metrology and multiphoton coherent microscopy.

Abstract:We report an all-fiber ultra-short pulse burst laser operating at around 1.98 µm that is obtained through a nonlinear wavelength converter and Tm-doped fiber amplifier. A mode-locked Er-doped fiber laser was first built and then amplified in subsequent amplifiers to an average power of 1.3 W. Ultra-short pulse burst output was achieved through a pulse multiplier and a fiber-pigtailed acousto-optic modulator. It was then injected into an all-fiber nonlinear wavelength converter constructed from P-doped fiber and Tm-doped fiber, obtaining an ultra-short pulse burst laser of 540 mW around 1.98 µm. Its average output power was then amplified to 4.33 W in a Tm-doped fiber amplifier with an intra-burst pulse repetition frequency of 0.9 GHz, a burst repetition frequency of 200 kHz, and a duty cycle of 2%, corresponding to about 200 pulses within each burst. This 1.98 µm pulse burst laser has enormous potential to be applied in bio-medical areas.
