Abstract:A simple and efficient procedure for the synthesis of pyridazin-3(2H)-ones through copper-catalyzed dehydrogenation of a single C-C bond of 4,5-dihydropyridazin-3(2H)-ones to a C=C bond with oxygen as the terminal oxidant is described. Functional groups including hydroxy, carboxylic, bromo, chloro, cyano, nitro and alkoxy were all tolerated under the reaction conditions. Moreover, this methodology was applied to the preparation of a series of structurally similar N-substituted 6-phenylpyridazinone compounds containing fluorine. The dehydrogenation reactions exhibit good yields and selectivity.

Abstract:A novel and efficient copper-catalyzed cascade method for the synthesis of quinazoline derivatives has been developed. The protocol uses readily available substituted (2-bromophenyl)methylamines and amidine hydrochlorides as the starting materials, inexpensive CuBr as the catalyst, and economical and environment friendly air as the oxidant, and the corresponding quinazoline derivatives were obtained in moderate to good yields. The procedure underwent sequential intermolecular N-arylation, intramolecular nucleophilic substitution and aerobic oxidation.

Abstract:A novel FeCl2·4H2O-mediated internal-oxidant relay cascade reaction has been developed by functionalization of O-substituted benzohydroxamic acids or N-chloro-arylamides with [60]fullerene. Depending on the nature of the N-substituted groups, fullerenooxazoles or rare hydroxyfullerenyl amides could be obtained in a straightforward and flexible manner. Such a new transformation provides a unique strategy for the synthesis of fullerenooxazoles or hydroxyfullerenyl amides.

Abstract:A novel strategy involving Cu-catalyzed oxidative transformation of ketone-derived hydrazone moiety to various synthetic valuable internal alkynes and diynes has been developed. This method features inexpensive metal catalyst, green oxidant, good functional group tolerance, high regioselectivity and readily available starting materials. Oxidative deprotonation reactions were carried out to form internal alkynes and symmetrical diynes. Cross-coupling reactions of hydrazones with halides and terminal alkynes were performed to afford functionalized alkynes and unsymmetrical conjugated diynes. A mechanism proceeding through a Cu-carbene intermediate is proposed for the CC triple bond formation.

Abstract:A sequential multi-component reaction has been achieved via aerobic copper-catalyzed intermolecular C-N/C-O bonds cascade formation process. This mild, operationally simple method enables the transformation of commercially available aldehydes, along with aryl hydrazines, into the corresponding N',N'-diaryl acylhydrazines with high regioselectivity in moderate to good yields. This transformation employs aryl hydrazine as a dual synthon, a hydrazine and an aryl synthon. The value of this efficient synthetic protocol has been demonstrated through gram-scale experiment, late-stage modification of bioactive natural products and drug molecules. Mechanistic investigations support the generation of an acyl diazene intermediate and an aryl radical species, along with the dual role of air as the sole oxidant and source of oxygen functionality in this aerobic C-N bond coupling reaction.

Abstract:On the basis of our recent discovery of high valent organocopper compounds, we have designed and achieved efficient copper(II)-catalyzed oxidative arene CH bond azidation under very mild aerobic conditions by using NaN3 as an azide source. In the presence of a Cu(II) catalyst, a number of azacalix[1]arene[3]pyridines underwent direct arene CH bond cupration through an electrophilic aromatic metalation pathway to form an arylcopper(II) intermediate. Oxidized by a free copper(II) ion, the arylcopper(II) intermediate was transformed into an arylcopper(III) species that subsequently cross-coupled with azide to furnish the formation of aryl azide products with the release of a copper(I) ion. Under ambient catalytic reaction conditions, the copper(I) species generated was oxidized by air into copper(II), which entered into the next catalytic cycle. Application of the method was demonstrated by the synthesis of functional azacalix[1]arene[3]pyridines by means of simple and practical functional group transformations of azide. The showcase of the Cu(II)ArCu(II)ArCu(III)Cu(I) catalytic cycle would provide a new strategy for the design of copper(II)-catalyzed aerobic oxidative arene CH bond activation and transformations.
