Abstract:A novel cascade reaction was developed for the synthesis of diverse members of a series of benzofuro(3,2-d]pyrimidine derivatives. The process utilizes readily prepared 3-chlorochromenones and various commercially available amidines and their analogues as starting materials. This tandem reaction is promoted by using a simple copper(I) reagent and involves a chemoselective Michael addition-heterocyclization-intramolecular cyclization sequence.

Abstract:Here, a general copper-catalyzed radical cascade carbocyclization reaction with 2-arylbenzoimidazoles and a Togni reagent was realized. Structurally diverse CF3-containing tetracyclic core benzimidazo[2,1-a]isoquinoline-6(5H)-ones were obtained in moderate to good yields. The wide substrate scope, good functional group tolerance, and ease of scale-up of this method are expected to promote its potential applications in pharmacy and biotechnology.

Abstract:A Cu-catalyzed tandem synthesis of azole-fused pyrimido[1,2-c]quinazolines and imidazo[1,2-c]quinazolines has been developed. The reaction is based on a C-N cross-coupling/C-H functionalization reaction of 2-(2-bromophenyl)-1,4,5,6-tetrahydropyrimidines with azoles. Abstract:A selective copper(I)-catalyzed benzylic C(sp3)–H geminal difunctionalization reaction of a benzylic-type sp3 carbon was developed. This novel strategy allowed simultaneous introduction of amide and hydroxyl group in a highly selective and efficient way via successive oxidative intramolecular amidation and hydroxylation. This method was also applied to the synthesis of 3-hydroxy-2,3-dihydro-1H-pyrrolo[3,4-b]quinoxalinones from readily available 3-methyl-N-substituted quinoxaline-2-carboxamides in moderate to good yields. The five-membered cyclic hemiaminal moiety of the pyrrolo[3,4-b]quinoxalinones can serve as an intermediates for subsequent transformations into other useful functional groups.

Abstract:Herein, an atom-economic method for the synthesis of fused-ring pyrrolizine derivatives by a cycloaddition reaction of easily accessible N -substituted pyrrole-2-carboxaldehydes with N -substituted maleimides in the presence of di- tert -butyl peroxide has been successfully developed. A total of 23 compounds were obtained by using this method, with a maximum yield of 72%, providing a practical and efficient method for the synthesis of tricyclic pyrrolizine frameworks.
