Abstract:Copper-catalyzed aerobic methyl/methylene oxygenation of substituted indoles and pyrroles was developed using 1,4-diazabicyclo[2.2.2]octane (DABCO) as an additive in dimethyl sulfoxide (DMSO). Similar aerobic catalytic conditions could also be utilized for direct C–H formylation of C(3) on indoles and C(2) on pyrroles.

Abstract:An efficient copper-catalyzed tandem oxidation/alpha-formylation reaction of 2,3-dihydro-1H-pyrroles in an oxygen atmosphere has been developed. The new reaction is conducted in dimethyl sulfoxide (DMSO), which serves as both the carbonyl carbon source and solvent, in the presence of trifluoroacetic acid to directly generate structurally diverse alpha-formylpyrrole derivatives in one pot in moderate to good yields. Sensitive aldehyde groups remain intact in the presence of an oxidant, trifluoroacetic acid and a transition metal salt. A possible mechanism is presented.

Abstract:A tandem coupling-ketooxygenation reaction of readily accessible enediyne-carboxylic compounds with inner alkynes has been developed that utilizes the PdCl2/CuBr2 catalytic system under an O2 atmosphere and assembles a class of isoindolinones and o-acylbenzoic acids. The two oxygen atoms are regioselectively incorporated into enediyne units at the 1- and 6-positions from atmospheric molecular oxygen and H2O, respectively, during the present process. This study uncovered a formal [4C + 2C] benzannulation-diketonization of enediynes and alkynes via a coupling and decoupling strategy.

Abstract:An efficient copper‐catalyzed ethoxycarbonyldifluoromethylation of indoles and pyrroles with commercially available BrCF2CO2Et is reported. The developed protocol allows the site‐selective introduction of the CF2CO2Et moiety at the C2‐position of N‐heteroarenes with the assistance of a pyrimidyl group using CuI as the catalyst and 4,7‐diphenyl‐1,10‐phenanthroline as the ligand. This procedure is scalable and tolerates various important functional groups.

Abstract:We report a safe and convenient open-flask copper-catalyzed selective oxidation/functionalization methodology for tetrahydrocarbolines and tetrahydro-β-carbolines that employs atmospheric O<sub>2</sub> as the terminal oxidant. The system is applicable to oxidative rearrangement of tetrahydro-β-carbolines, tetrahydrocarboline oxidation to α-alkoxy carbazoles and spirooxindoles, and Witkop oxidation. Mechanistic experiments indicated that a single-electron oxidation process is responsible for the tunable selectivity control. This copper-catalysis protocol represents a significant advance in the field of indole oxidation.

Abstract:Base-switched methylenation and formylation using tetramethylethylenediamine (TMEDA) as a carbon source have been achieved under mild conditions, catalyzed by CuCl2, with atmospheric oxygen as oxidant. Bisindolylmethanes, diphenylmethanes and 3-formylindoles were synthesized with excellent regioselectivity and good yield.
