Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:A concise synthesis of 8, 9-dihydo-2H-pyrano-[5,6-g] quinolin-2-one derivatives was reported, using 3-amino-phenol as starting material. Treatment of 3-amino-phenol with ketones bearing alpha C - H at the presence of a catalyst PTS .H2O affords compound 2a similar to 2d, followed by treatment with ethyl acetoacetate, gives target compound 3a similar to 3d with regioselectivity.

Abstract:Quinone methides, generated in situ from the reaction of 4-hydroxycoumarin with benzaldehyde, formaldehyde and pentanal, underwent the Diels-Alder reaction with silyl enol ethers to give a series of 3,4-dihydro-2H,5H-pyrano[3,2-c][1]benzopyran derivatives in high yields. Steric configurations of five compounds were determined by X-ray diffraction experiments. The regioselectivity of the reactions was investigated by frontier molecular orbital interactions between the reactants as calculated by a density functional theory (DFT) method, which well rationalized the observed data.

Abstract:This study provides straightforward synthesis of 4 H -pyran-annulated heterocycles that serve as drug-like candidates for biological purposes. It was found that these multicomponent reactions performed under green conditions in the presence of sodium benzoate as an efficient, environmentally benign, and commercially available organo-salt catalyst. The target 2-amino-4-aryl-5-oxo-5,6,7,8-tetrahydro-4 H -chromene-3-carbonitriles, 7-amino-5-aryl-2,4-dioxo/(4-oxo-2-thioxo)-1,3,4,5-tetrahydro-2 H -pyrano[2,3- d ]pyrimidine-6-carbonitriles, and 2-amino-4-aryl-5-oxo-4,5-dihydropyrano[3,2- c ]chromene-3-carbonitriles, which are estimated to be important in organic synthetic and medicinal chemistry, were obtained in high yields and relatively shorter reaction times through a simple filtration. The method is versatile and amenable to many substrates, as it requires no specialized devices such as microwave, ultrasound and ball-milling. The aim of the work is the synthesis of pyran-annulated compounds in the presence of sodium benzoate as the organo-catalyst. This procedure has several further advantages, such as isolation of heterocyclic products without column chromatography or other chromatographic methods, atom-economy, step-economy, and clean reaction profiles.

Abstract:An effective route to novel 4-(alkylamino)-1-(arylsulfonyl)-3-benzoyl-1,5-dihydro-5-hydroxy-5-phenyl-2H-pyrrol-2-ones 10 is described (Scheme 2). This involves the reaction of an enamine, derived from the addition of a primary amine 5 to 1,4-diphenylbut-2-yne-1,4-dione, with an arenesulfonyl isocyanate 7 Some of these pyrrolones 10 exhibit a dynamic NMR behavior in solution because of restricted rotation around the C-N bond resulting from conjugation of the side-chain N-atom with the adjacent alpha,beta-unsaturated ketone group, and two rotamers are in equilibrium with each other in solution (10 reversible arrow 11; Scheme 3). The structures of the highly functionalized compounds 10 were corroborated spectroscopically (IR, H-1- and C-13-NMR, and EI-MS), by elemental analyses, and, in the case of 10a, by X-ray crystallography. A plausible mechanism for the reaction is proposed (Scheme 4).
