Abstract:As a key building block for 2,4-disubstituted pyridines, 2-bromo-4-iodopyridine was synthesized conveniently from 2-bromopyridine using LDA and I-2 via 'halogen dance'. 2-Bromo-4-iodopyridine was then converted to other 2,4-diaryl pyridines and 4-aryl-2,2'-bipyridines in one-pot reactions.

Abstract:A novel tandem one-pot method for the synthesis of polysubstituted pyridine derivatives has been developed via Knoevenagel-type reaction and subsequent 675 electrocyclization of the Blaise reaction intermediates and 3-formylchromones. Abstract:By means of selective cross-coupling of ArMgBr with 2-bromo-4-iodo-pyridine in the presence of a catalytic amount of Pd(PPh3)(4), 4-Ar-2-bro-mopyridines were facilely one-step synthesized. These pyridine compounds were smoothly transformed into their 2,2'-bipyridine derivatives via Stille-type cross-couplings.

Abstract:An efficient and concise method is hereby reported for the synthesis of pyrido[3,4- c ]pyridazin-8(7H)-ones from the commercially available material, methyl 4,6-dichloropyridazine-3-carboxylate. The four-step reaction process involves Suzuki coupling, Buchwald coupling, ammoniation, and intramolecular cyclization. The advantage of this route represents the development of a novel C5 site-selective Suzuki monocoupling of 3,5-dichloropyridazine, with an overall yield of over 73 %.

Abstract:A copper-catalyzed process for the construction of pyrrolobenzoxazepinones from 2-(2-iodophenoxy)-1-(1H-pyrrol-2-yl)ethanones is described. The reaction afforded a variety of products in good to excellent yields.

Abstract:Reactions of easily accessible chloropyrimidinyl hydrazones with bromine were investigated. Oxidative cyclization followed by concomitant bromination led to the formation of [1,2,4]triazolo[4,3-c]pyrimidines, which then underwent the Dimroth rearrangement to afford highly functionalized 2-substituted [1,2,4]triazolo[1,5-c]pyrimidines.

Abstract:An efficient procedure for the synthesis of 7‐(aryl)‐8‐nitro‐2,3,6,7‐tetrahydroimidazo[1,2‐a]pyridinones, 8‐(aryl)‐9‐nitro‐3,4,7,8‐tetrahydropyridone[1,2‐a]pyrimidines and 9‐(aryl)‐10‐nitro‐2,3,4,5,8,9‐hexahydropyridone[1,2‐a]diazepine via one‐pot three component reaction of diamine, nitroketene dithioacetal (1,1‐bis(methylsulfanyl)‐2‐nitroethene), and coumarine‐3‐ carboxylic acid derivatives in EtOH under reflux conditions is reported. The advantages of this procedure are simplicity, easy purification, good yields, and catalyst‐free conditions. All products were confirmed by 1H‐ and 13C‐NMR, IR, MS, and X‐ray crystal structure analyses.

Abstract:We report the high-yielding, large-scale, one-pot synthesis of two versatile building blocks (1-Cl and 1-Br) for the regioselective synthesis of a variety of 2,3,5-trisubstituted pyridines from inexpensive materials. These molecules are readily derivatized at positions 2, 3, and 5. These building blocks can also be used for the synthesis of fused pyrido-oxazines and for the synthesis of 2,3,4,5-tetrasubstituted pyridines.
