Abstract:A new method for the solid phase synthesis of benzothiazoles and 2-arylamino-3-carboxyl-4-hydroxy-5-arylthiophenes is described. Resin bound cyclic malonic acid ester was reacted with aryl isothiocyanates and then with bromine or α-bromoketones, followed by treatment with perchloric acid or sodium methoxide to afford substituted 2-methylbenzothiazoles or 2-arylamino-3-carboxyl-4-hydroxy-5-arylthiophenes, respectively, in high yields and purities.

Abstract:An efficient method for the preparation of polymer-bound cyclic malonic acid ester starting from Merrifield resin has been developed. Reaction of the resin-bound cyclic malonic acid ester with triethyl orthoformate and subsequent double substitution with nucleophilic reagents, such as arylamine, urea, thiourea, 2-aminobenzothiazoles, or isothiosemicarbazones, afforded the corresponding polymer-bound substituted aminomethylene cyclic malonic acid esters, which upon thermal treament led to 4(1H)-quinolones, 3-substituted uracils and thiouracils, 4H-pyrimido[2,1-b]benzothiazol-4-ones, and 1-(N-alkylidene or benzylideneamino)-1,6-dihydro-2-methylthio-6-oxo-pyrimidines, depending on the structures of the nucleophilic reagents.

Abstract:The resin-bound 5-monosubstituted cyclic malonic ester 3 was generated and reacted with an alpha-bromoketone to give the corresponding 5,5-disubstituted cyclic malonic ester resin (4). Subsequent reaction with hydrazine resulted in cyclisation with concomitant cleavage from the polymeric support to release the final products, 4,6-disubstituted 4,5-dihydro-3(2H)-pyridazinones, in good yield and high purity.

Abstract:An approach for the solid-phase synthesis of 4,81 disubstituted-8,9-dihydropyrazino[2,3-g]quinazolin-7(6H)-ones from 4-chloro-7-fluoro-6-nitroquinazoline scaffold is described. A chemoselective reaction of resin-bound arylamines (3) with 4-chloro-7-fluoro-6-nitroquinazoline (8) yielded resin-bound 4-arylamino-7fluoro-6-nitroquinazolines (4), which were reacted with amino acid methyl esters to afford the corresponding resin-bound compound (5). Following the reduction of nitro group and intramolecular cyclization of 5, resin-bound 4,8-disubstituted-8,9-dihydropyrazino[2,3g]quinazolin-7(6H)-one (6) was yielded. The desired products (7) were obtained in good yields and purities after cleavage from the resin. (c) 2007 Wiley Periodicals, Inc.

Abstract:Solid-phase synthesis is greatly dependent on the solid support. Here, we report the use of a new hydrophilic grafted surface on SynPhase lanterns in solid-phase organic chemistry. A convenient and facile solid-phase synthesis of disubstituted 1,4-benzodiazepine-2-ones on polyamide SynPhase lanterns is described. The key step of the synthesis involved a reduction-cyclization of a nitroaryl methyl ester with a mixture of tin(II) chloride dihydrate and ammonium acetate in water and ethanol at elevated temperature to give the desired target compounds. A library of 21 disubstituted 1,4-benzodiazepine-2-ones was prepared.

Abstract:Pyridinium 1,4-zwitterionic thiolates, as a novel kind of sulfur-containing synthon, have been applied to the synthesis of 12aH-benzo[f]pyrido[1,2-d][1,4]thiazepines and benzo[b]thiophenes. Benzopyridothiazepines were produced through a 1,5-dipolar cycloaddition reaction from pyridinium 1,4-zwitterionic thiolates with arynes, whereas benzothiophenes as side products were generated via a [3 + 2] cascade cyclization reaction. The [5 + 2] reaction mode of pyridinium 1,4-zwitterionic thiolates is disclosed for the first time.

Abstract:Practical and efficient parallel methods have been developed for the synthesis of 7,8-disubstituted 2,3-dihydro-1,5-benzothiazepin-4(5H)-ones and 3,7,8-trisubstituted 2,3-dihydro-1,5-benzothiazepin-4(5H)-ones. This benzothiazepin-4(5H)-one skeleton possesses three or four diversity points. Furthermore, three novel tricycles integrating a benzothiazepin-4(5H)-one scaffold with other privileged structures, such as benzimidazole, benzimidazolone, and thio-benzimidazole, were also developed. The synthetic strategy provides an efficient way to access the benzothiazepinone core, starting from commercially available 1,5-difluoro-2,4-dinitrobenzene (DFDNB), and also allows further derivation of the strategically anchored functionalities.

Abstract:The [4+3] annulation of aza‐o‐quinone methides and pyridinium 1,4‐zwitterionic thiolates has been developed for the one‐step synthesis of functionalized 2,3‐unsaturated 4,1‐benzothiazepines under mild and metal‐free conditions. Abstract:The solid phase synthesis of 2-aminoimidazolidin-4-ones from resin-bound amino acids is described. Resin-bound thioureas were treated with Mukaiyama's reagent (2-chloro-1-methypyridinium iodide) to form, via intramolecular cyclization, resin-bound 2-iminoimidazolidin-4-ones. Following alkylation and cleavage, the corresponding 1,5-disubstituted 2-aminoimidazolidin-4-ones were provided in good yield and purity.

Abstract:The main objective of this work is the synthesis of polycyclic molecules of benzo[4,5]imidazo[1,2- a ]pyrimidine hybrids by condensation of 2-aminobenzimidazole with the precursor 4-hydroxy-6-methylpyran-2-one utilizing various alcoholic solvents. Structural characterization of the synthesized compounds was conducted using diverse spectroscopic methods (NMR, MS, IR, and CHN). The formation of the obtained structures is explained by a detailed reaction mechanism.
