Abstract:A new method for the solid phase synthesis of benzothiazoles and 2-arylamino-3-carboxyl-4-hydroxy-5-arylthiophenes is described. Resin bound cyclic malonic acid ester was reacted with aryl isothiocyanates and then with bromine or α-bromoketones, followed by treatment with perchloric acid or sodium methoxide to afford substituted 2-methylbenzothiazoles or 2-arylamino-3-carboxyl-4-hydroxy-5-arylthiophenes, respectively, in high yields and purities.

Abstract:As an intermediate for COX-2 inhibiters,4-(methylthio)benzeneacetic acid(Ⅳ) was prepared via Friedel-Crafts and Willgerodt-Kindler reaction from thioanisole(Ⅰ).When the molar ratio of Ⅰ to acetyl chloride and AlCl_3 was 1∶1.1∶1.3 and CS_2 was used as solvent,after reacting at(0°C) for 6 h the yield of 4-(methylthio) acetophenone(Ⅱ) was 75.9%.Ⅱ was refluxed 5 h with morpholine and sulfur to give thioacetmorpholide(Ⅲ),then Ⅲ was refluxed in 10% NaOH for 10 h to give Ⅳ with an overall yield of 63.5% and purity of 99.0%(by HPLC).The structure was confirmed by IR and()~1H NMR.

Abstract:A convenient and efficient synthesis of 2-substituted benzo[b]thiophenes is described. This one-step reaction of alkylthiobenzaldehyde and activated electrophiles proceeded smoothly in toluene at 120 degrees C with magnesium oxide as a catalyst. The target 2-substituted benzo[b]thiophenes are obtained in moderate yields and the side reaction is inhibited via azeotropic distillation. According to the experiments, a plausible mechanism is presented.

Abstract:The resin-bound cyclic malonic acid ester 2 reacted with aryl isothiocyanate and alkyl halides to afford the key intermediate arylthioaminomethylene cyclic malonic ester resin 3. Subsequently, resin 3 proceeded thermal cyclization giving the 2-alkylthio-4(1H)-quinolones in good yields and excellent purities.

Abstract:The resin-bound 5-monosubstituted cyclic malonic ester 3 was generated and reacted with an alpha-bromoketone to give the corresponding 5,5-disubstituted cyclic malonic ester resin (4). Subsequent reaction with hydrazine resulted in cyclisation with concomitant cleavage from the polymeric support to release the final products, 4,6-disubstituted 4,5-dihydro-3(2H)-pyridazinones, in good yield and high purity.

Abstract:A convenient method has been developed for the preparation of a variety of 1,3-benzothiazole-2(3H)-thiones. The reaction proceeds from an o-haloaniline derivative and carbon disulfide in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene at 80-140 degrees C to give the corresponding 1,3-benzothiazole-2(3H)-thione derivatives in good-to-excellent yields.

Abstract:A novel protocol has been successfully established for the efficient synthesis of benzothiazole-tethered chromanone/coumarin scaffolds via Claisen rearrangement using a solid state melt reaction in a one-pot manner. Benzothiazole formation and Claisen rearrangement involve the cleavage of S-S and C-O bonds and formation of C-S, C═N, and C-C bonds in a single operation without using a catalyst or solvent.

Abstract:Many nitrogen- and sulfur-containing heterocyclic compounds exhibit biological activity. Among these heterocycles are benzo[4,5]thiazolo[2,3-c][1,2,4]triazoles for which two main synthetic approaches exist. Here we report a new synthetic protocol that allows the preparation of these tricyclic compounds via the oxidation of a mercaptophenyl moiety to its corresponding disulfide. Subsequent C-H bond functionalization is thought to enable an intramolecular ring closure, thus forming the desired benzo[4,5]thiazolo[2,3-c][1,2,4]triazole. This method combines a high functional group tolerance with short reaction times and good to excellent yields.

Abstract:A facile and efficient method to synthesize thioethers from sulfonyl chlorides and benzylic trimethylammonium salts is presented. This protocol provides a simple and direct approach to realize the reductive coupling reaction of readily available sulfonyl chlorides with benzyl ammonium salts in the presence of triphenylphosphine along with base via C‐N cleavage of benzyl ammonium salts. This reaction features mild conditions and good substrates compatibility to afford thioethers in moderate to good yields.

Abstract:The solid-phase synthesis of 1,3-disubstituted and 1,3,5-trisubstituted 1,3,5-triazine-2,4,6-triones from MBHA and Wang resin is described. Reaction of resin-bound amino acids with isocyanates yield resin-bound ureas, which further react with chlorocarbonyl isocyanate in toluene at 65 degreesC to selectively afford the resin-bound 1,3-disubstituted 1,3,5-triazine-2,4,6-triones. Selective alkylation at the N-5 position of the resin-bound 1,3-disubstituted 1.3,5-triazine-2,4,6-triones was accomplished by treatment with alkyl halides in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU). The desired products were cleaved from their solid support and obtained in good yield and purity. The method can be employed in production of toltrazuril analogue libraries for identification of new anticoccidial agents.

Abstract:The reactions of N -[2-(cyclohex-1-en-1-yl)-6-methylphenyl]arenesulfonamides and N -[2-(cyclohex-1-en-1-yl)phenyl]methanesulfonamide with molecular bromine in the presence of ethylene chlorohydrin afforded mixtures of N -{2-[6-(2-chloroethoxy)cyclohex-1-en-1-yl]phenyl} sulfonamides and 9-(arenesulfonyl)- or 9-(methanesulfonyl)-2,3,9,9a-tetrahydro-1 H -carbazoles. Intramolecular condensation of the obtained 2-chloroethyl ethers in the presence of potassium hydroxide and benzyl(triethyl)ammonium bromide in THF gave 8-(arenesulfonyl)-9-methyl-2,4,4a,6,7,8-hexahydro-3 H -dibenzo[ e , g ][1,4]oxazocines as a result of formation of new nitrogen–carbon bond. The presence of a methyl group at C 9 of the aromatic ring gives rise to axial isomerism of the synthesized benzoxazocines. The major conformer of the cyclocondensation products has a S *, R * configuration of the heterocycle, and this conformer slowly isomerizes to the a R *, R * conformer at room temperature.

Abstract:A facile solid phase synthesis of 2-substituted 1,3-oxazin-6-ones using polymer-supported Meldrum's acid has been reported. Reaction of the resin-bound cyclic malonic acid ester with triethyl orthoformate and subsequent double substitution with amide, afforded the corresponding polymer-supported acylaminomethylene cyclic malonic acid ester, which upon thermal treatment led to 1, 3-oxazin-6-ones in good yields and with high purity.

Abstract:An efficient method for the preparation of polymer-bound cyclic malonic acid ester starting from Merrifield resin has been developed. Reaction of the resin-bound cyclic malonic acid ester with triethyl orthoformate and subsequent double substitution with nucleophilic reagents, such as arylamine, urea, thiourea, 2-aminobenzothiazoles, or isothiosemicarbazones, afforded the corresponding polymer-bound substituted aminomethylene cyclic malonic acid esters, which upon thermal treament led to 4(1H)-quinolones, 3-substituted uracils and thiouracils, 4H-pyrimido[2,1-b]benzothiazol-4-ones, and 1-(N-alkylidene or benzylideneamino)-1,6-dihydro-2-methylthio-6-oxo-pyrimidines, depending on the structures of the nucleophilic reagents.
