Abstract:Elemental sulfur and isothiocyanates S8/RNCS couple was found to undergo redox condensation with o-halonitrobenzenes in the presence of N-methylpiperidine as a base in N-methylpyrrolidin-2-one to provide step- and redox- economical access to 2-aminobenzothiazoles. Alternatively, dithiocarbamate salts generated in situ from addition of amines and CS2 could be used in place of the S8/RNCS couple. Benzothiazole with a 2-amino substituent have frequently been used as core structures in the development of bioactive molecules.1 Consequently, they are important intermediates and broadly found in biological chemistry and medicinal chemistry.2 Several thiazole derivatives have entered into clinical trials showing a wide range of biological activities (Scheme 1).3 Successful examples include antimosquito agent AB1, phosphodiesterase 10 inhibitor AB2,4anti-inflammatory agent or Talarozole AB3 used osteoarthritis treatment,5 anticancer molecules AB46 and AB5,7 and inhibitor of p38α MAP kinase AB6.8 Traditionally, the synthesis of 2-aminobenzothiazoles9 is heavily based on the starting materials in which the resulting endocyclic nitrogen atom is in the lowest oxidation state (i.e. -3 as in aniline derivatives or simpler benzothiazoles.10 These methods usually require multi-step and harsh reaction conditions for the synthesis of the desired products from commercially available aniline derivatives. Additionally, it should be noted that the introduction of the sulfur atom into the aromatic ring was mainly relied on transition-metal catalyzed Ullmann-type coupling reactions of o-haloanilines.11 The required substrates for these strategies are generally available in only a limited number of simple structures, and thus hinders their synthetic application, especially in drug discovery research wherein a large set of candidate molecules are highly desirable for structure–activity relationship study.
