Abstract:An improved synthesis technology of 1-methyl-3-trifluoromethyl-4-pyrazole carboxylic acid which is an important intermediate of pesticides was studied.The title compound was synthesized by sodium cyanoacetate via condensation,acylation,cyclization and hydrolysis.The suitable reaction conditions were found as follows.The condensation reaction was carried out at 40~45℃ for 5 h,and in which the ethanol was used as solvent,the molar ratio of sodium cyanoacetate to N-(methoxymethylene)-N,N-dimethylaminium methanesulfonate is 1:2.5,and under above conditions the yield was improved from 67%(reported in the literatures) to 93.7%.During acylation,TEA was used as acid-capturer,trifluoroacetyl chloride was used to replace phosgene as acylating reagent and the yield of 78.6% was obtained when the molar ratio of 3-(dimethylamino)acrylonitrile,trifluoroacetyl chloride and TEA is 1:1.5:2.The cyclization reaction was carried out in a mixed solvent of methanol and water,and the product yield of 74.3% was obtained.The structures of the intermediates and product were confirmed by 1H-NMR.The proposed process has quite good industrial value because of its high yields,cheap and easily available raw materials,moderate reaction conditions and convenient operations.

Abstract:α-Hydroxy amides are a class of important intermediates in organic synthesis. It plays an important role in the field of science and medicine. However, the preparation methods of α-hydroxy amides are relatively few, and they are usually enzyme-or Lewis acid-catalyzed reactions. These types of methods are not ideal in operation, yield and/or stereoselectivity. We found a new way to synthesize α-hydroxy amides by transforming amino acids into α-hydroxyl acids, and then by amination of the α-hydroxy acids.

Abstract:Synthesis of poly(α-amino acid)s bearing carboxyl groups is a critical pathway to prepare biomaterials to simulate functional proteins. The traditional approaches call for carboxyl-protected monomers to prevent degradation of monomers or wrong linkage. In this contribution, we synthesize N -carboxypentyl glycine N -thiocarboxyanhydride (CPG-NTA) and iminodiacetic acid N -thiocarboxyanhydride (IDA-NTA) without protection. Initiated by amines, CPG-NTA directly polymerizes into polyCPG bearing unprotected carboxyl groups with controlled molecular weight (2.8–9.3 kg mol −1 ) and low dispersities (1.08–1.12). Block and random copolymerizations of CPG-NTA with N -ethyl glycine N -thiocarboxyanhydride (NEG-NTA) demonstrate its versatile construction of complicated polypeptoids. On the contrary, IDA-NTA transforms amines into cyclic IDA dimer-capped species with carboxyl end group in decent yields (>89%) regio-selectively. Density functional theory calculation elucidates that IDA repeating unit is prone to cyclize to be the six-membered ring product with low ΔG . The polymer is a good adhesive reagent to various materials with adhesive strength of 33–229 kPa.

Abstract:Three new compounds were first synthesized by the reaction of N-benzoyl-N~α-solanesyl glutaminic acid with different amines or amino acid ester hydrochloride by the method of the activated N-hydroxysuccinimide ester in the presence of N, N-dicyclohexylcarbodiimide. Their structures were confirmed by IR, ()~1H NMR,MS and elemental analyses.

Abstract:A Trost Pd-catalyzed [3+2] cycloaddition between a p -benzyloxy cinnamate bearing an Evans auxiliary and 1-trimethylsilyl-2-acetoxymethylpropene is disclosed leading to, after functional group manipulation, previously unreported diastereoisomeric 3-amino-5-arylcyclopentane 1-carboxylic acids via the corresponding 3-hydroxy and 3-azido precursors. The availability of these conformationally restricted cyclic amino acids may find utility in the context of CNS-active compounds related to GABA, or as peripheral units of bioactive pharmaceuticals. An expedient alternative 4-step synthesis of 3 S -amino-5 S - p -hydroxyphenyl-1 S -cyclopentane carboxylic acid methyl ester was achieved starting with the (–)-Vince lactam and utilizing a regio- and diastereoselective Pd-catalyzed hydroarylation reaction. It was found that many problems occurred preparing amino acid derived N-acylsulfonamides with the commonly used coupling approach in our previous studies. In this paper, we report an efficient approach to synthesize various amino acids derived N-acylsulfonamides in high yields without any racemization.

Abstract:Aim To synthesize protected aminoalkyl sulfmyl dilactones which were useful as the synthetic intermediates or the Cterminal pharmacophores of potential peptidomimetic proteasome inhibitors. Methods Organic reactions such as reduction, oxidation, olefination, and dihydroxylation were used. Results A convenient synthetic procedure to afford a series of aminoalkyl sulfinyl dilactones was presented, which would be useful in the synthesis of five- or six-member sulfinyl dilactones. Conclusion Four aminoalkyl sulfinyl dilactones connecting different α-amino acids were synthesized.

Abstract:Objective: To synthesize N-[(1-methyl-β-carboline-3-yl)-formyl]-glycine. Methods: The target compound was obtained from L-tryptophan via 6-step reactions, including the Pictet-Spengler condensation, methylation, aromatization, hydrolyzation, coupling reaction and hydrolyzation. Results: The yields of the Pictet-Spengler condensation, aromatization and coupling reaction were 95%, 65% and 37%, respectively. The inhibition rate of N-[(1-methyl-β-carboline-3-yl)-formyl]-glycine benzyl ester 10, 1, 0.1μmol·L~(-1) on the MCF-7 human tumor cell line was 61.50%, 12.40% and 7.24%, respectively. Conclusion: The target amino acid and its ester prepared by this convenient synthetic process potentiated an anti-tumor activity.

Abstract:A new route for the synthesis of( 3S,4S)-4-amino-3-hydroxy-5-phenylpentanoic acid derivatives was reported. The target compounds with overall yields of 5. 8% ~ 6. 7% were synthesized by condensation with Weinreb amide,reduction,aldol condensation reaction and bromine imitation reaction,using amino-protected L-phenylalanine as starting material. The structures were confirmed by1 H NMR,13 C NMR and ESI-MS. The exploration of reaction conditions showed that the loading amount of D-proline had no evident effect on the yield,but on stereoselectivity. Larger size protecting groups in substrate can enhanced the stereo-selectivity.

Abstract:Optically active 1-aminoalkylphosphonic acid derivatives were synthesized in moderate yields and optical purities via Mannich-type reactions of benzyl carbamate, aldehydes, and optically pure chlorophosphites. Side chain-functionalized depsiphosphonopeptides were also prepared in satisfactory yields directly from one-pot reactions of benzyl carbamate, aldehydes, and diethyl (R,R)-2-chloro-1,3,2-dioxaphospholane-4,5-dicarboxylate.

Abstract:In this paper, we reported the development of an efficient and highly productive synthetic protocol for preparing new 2,5-diamino-1,3,4-thiadiazole derivatives functionalized with N -(2,2,2-trichloroethyl)carboxamide group. The method of their preparation was based on the reaction of oxidative dehydrosulfurization of N -(2,2,2-trichloro-1-(2-(phenylcarbamothioyl)hydrazine-1-carbothioamido)ethyl)carboxamides and N -(1-(2-carbamothioylhydrazine-1-carbothioamido)-2,2,2-trichloroethyl)benzamide under the action of a mixture of iodine and triethylamine in DMF. This reaction took place at room temperature for two hours and allowed obtaining the target products with a 69-75% yield. Using the developed protocol, we obtained ten new derivatives of 1,3,4-thiadiazole. Their structure was proven by IR, 1 H NMR, 13 C NMR, 1 H- 1 H COSY, 1 H- 13 C HSQC, 1 H- 13 C HMBC spectroscopy, and LC/HRMS spectrometry.

Abstract:Four phosphonodipeptides containing 1-aminoalkylphosphonate were synthesized by applying four different coupling methods. One of them was then converted into monoacid, ethyl hydrogen 1-(N-acetyl-prolyl)amino-2-phenylethyl phosphonate. Three phosphonodipeptides containing ethyl 1w-amino-2-phenylethylphosphonamidate were also prepared by converting phosphono monoacid into phosphonochloridate and reacting it with amino acid ester. Benzyl ethoxyphosphonyl-glycyl-glycine ethyl ester also was synthesized from ethyl glycylglycinate and ethyl benzylphosphonochloridate derived from direct chlorination of diethyl benzylphosphonate with phosphorus oxychloride.
