Abstract:α-Hydroxy amides are a class of important intermediates in organic synthesis. It plays an important role in the field of science and medicine. However, the preparation methods of α-hydroxy amides are relatively few, and they are usually enzyme-or Lewis acid-catalyzed reactions. These types of methods are not ideal in operation, yield and/or stereoselectivity. We found a new way to synthesize α-hydroxy amides by transforming amino acids into α-hydroxyl acids, and then by amination of the α-hydroxy acids.

Abstract:The (4E)-N-(4-chlorophenyl)-5-(3-chlorophenyl)-2-diazo-3-oxopent-4-enoic acid amides 5a~j were synthesized with N-(4-chlorophenyl)-2-diazo-3-oxobutyramide 4 from p-chloroaniline and various arylaldehydes. The yielded products 5a~j were investigated with NMR, MS, IR, and X-ray crystallographic techniques.

Abstract:A variety of novel N-t-butyl-N′-aminocarbonyl-N-(substituted)benzoylhydrazines containing α-aminoalkylphosphonate groups were synthesized by the addition of N-t-butyl-N-(substituted)benzoylhydrazines to α-isocyanatoalkylphosphonates, which were synthesized by the reaction of α-aminoalkylphosphonates with triphosgene in good yields. The structures of products including by-products were confirmed by 1H NMR, 31P NMR, IR, mass spectroscopy, and elemental analysis.

Abstract:A series of novel substituted 2-amino-4H-benzo[h]chromene-3-carbonitrile derivatives was synthesised by condensation of different halogen derivatives with the hydroxy-group of 2-amino-8-hydroxy-4-styryl- 4H-benzo[h]chromene-3-carbonitrile and 2-amino-8-hydroxy-4-phenethyl-4H-benzo-[h]chromene-3-carbonitrile, which were prepared from cinnamaldehyde, malononitrile and naphthalene-1,6-diol through Knoevenagel condensation and cyclization reaction and then reduction with hydrogen in the presence of Pd/C at room temperature. Each intermediates and target compounds were obtained in good yields.

Abstract:Highly conformationally-constrained novel α-amino acid derivatives ((2R/3S) and (2S/3R)-2-(tert-butoxycarbonylamino)-3-cyclohexyl-3-phenylpropanoic acids) have been synthesised with high stereoselectivity (> 90% de) and in 36–37% overall yields. In the synthesis, Evans’ auxiliary (4(R/S)-4-phenyl-oxzaolidin-2-one) was used to control the stereoselectivity via the key reactions of asymmetric Michael addition, azidation and catalytic hydrogenolysis.

Abstract:3-Formacyl-9-ethyl carbazole and 3,6-diformacyl-9-ethyl carbazole were synthesized by the reaction of N-alkylation and formylation from carbozole.The structures were characterized by 1H NMR,IR and MS.Influences of reaction conditions on formylation were studied.The results showed that the appropriate formylation conditions were 1(10 mmol) and DMF[n(DMF) ∶n(1)=10 ∶1] in 1,2-dichloroethane(25 mL) under refluxing for 8 h～48 h.The yield and selectivity of product was improved by controlling the conditions for sing-formylation or di-formylation.

Abstract:Tetrabutyl ammonium bromide (TBAB) catalyzed one-pot synthesis of 4 H -chromene via a three-component cyclocondensation of aryl aldehydes, cyanoacetate and dimedone in green media–water. Then, 4 H -chromene reacted with acyl chloride in acetonitrile at refluxing with catalytic amount of DMAP to yield the title compounds in good yields. The structures of all title compounds were confirmed by 1 H NMR, MS and elementary analyses. The X-ray crystallography of compound 3n indicated that there were strong intermolecular hydrogen bonds. Abstract:The condensation of N-acylated amino acids with polymer-supported cyclic malonic acid ester was carried out in the presence of DCC/DMAP. The cyclisation of the intermediate resin, by heating in an organic solvent, gave the N-protected tetramic acid derivatives in good yields and high purity.

Abstract:Aim To synthesize protected aminoalkyl sulfmyl dilactones which were useful as the synthetic intermediates or the Cterminal pharmacophores of potential peptidomimetic proteasome inhibitors. Methods Organic reactions such as reduction, oxidation, olefination, and dihydroxylation were used. Results A convenient synthetic procedure to afford a series of aminoalkyl sulfinyl dilactones was presented, which would be useful in the synthesis of five- or six-member sulfinyl dilactones. Conclusion Four aminoalkyl sulfinyl dilactones connecting different α-amino acids were synthesized.

Abstract:A new route for the synthesis of( 3S,4S)-4-amino-3-hydroxy-5-phenylpentanoic acid derivatives was reported. The target compounds with overall yields of 5. 8% ~ 6. 7% were synthesized by condensation with Weinreb amide,reduction,aldol condensation reaction and bromine imitation reaction,using amino-protected L-phenylalanine as starting material. The structures were confirmed by1 H NMR,13 C NMR and ESI-MS. The exploration of reaction conditions showed that the loading amount of D-proline had no evident effect on the yield,but on stereoselectivity. Larger size protecting groups in substrate can enhanced the stereo-selectivity.
