Abstract:We reported that for a series of amines with different structures and functional groups, the deprotection of the dialkylphosphoryl group from the N-alkyl-N-dialkyl- phosphoramidates can be achieved under very mild conditions as compared to the corresponding sulfonyl derivatives. From the kinetic study, we find that the diisopropylphosphoramidates derivatives have substantial stability in the acid media. The O, O-diisopropyl-N-phenyl-phosphoramidates has a half life of 53 min in 6M HCl at 50℃. Also, the P-N bond persists during the Lewis acid catalyzed Freidel-Crafts acylation of N-dialkylphosphoryl-N-substituted phenethylglycine (4) to give the stable 2, 3, 4, 5-tetrahydro-7, 8-dialkoxyl-N-diisopropylphosphoryl- 3-benzazepine-1-one (5). 5 can be quantitatively transformed into the amine HCl salt under very mild condition. Thus. we have shown that diisopropyl phosphite can be used as an economical amino protection reagent, the steric hindrance of the diisopropyl group slows down the rate of acid hydrolysis, therefore, increases its stability in the acid media. This chemical property will be useful in organic synthesis.
