Abstract:Amino acid derivatives of a modified indole-3-acetic acid have been synthesised. Fourteen new dipepticle-like compounds 3-4 were obtained and their structures were elucidated based on the IR, H-1 NMR, MS spectra.

Abstract:Indole-2-carboxylic acid and indoline-2-carboxylic acid are common building block in natural products with bioactivities and anti-hypertension pharmaceuticals. The syntheses and transformation from each other were reviewed. Indoline-2-carboxylic acid can be got from the reduction of indole-2-carboxylic acid and the S enantiomer can be achieved from the resolution of the racemic mixture with chemical reagents or enzymes. The chiral pool synthesis, emerged recently, will play an important role in the future. Indole-2-carboxylic acid can be formed from Fischer way or substituted anilines and benzaldehydes.
Transformation of L -Phenylalanine to ( S )-Indoline-2-carboxylic Acid Without Group-Protection

Abstract:(S)-Indoline-2-carboxylic acid was synthesized by use of a nitro amination approach with l-phenylalanine as chiral pool. The first step of the synthesis was nitration of l-phenylalanine, with urea nitrate (UN)/H2SO4 as nitrating reagent, to give 2,4-dinitro-l-phenylalanine in 75.7 % yield in one-pot synthesis and 69.1 % yield by step-wise nitration. Intramolecular nitro amination of 2,4-dinitro-l-phenylalanine gave (S)-6-nitro-indoline-2-carboxylic acid in 65.7 % yield and more than 99.5 % enantiomeric excess (ee). The title compound, (S)-indoline-2-carboxylic acid, was obtained in 85.9 % yield and high ee by one-pot transformation of (S)-6-nitroindoline-2-carboxylic acid. The total synthesis consisted of three operations and gave the title compound in 42 % yield and more than 99.5 % ee.

Abstract:We report the construction of C5‐indole unnatural amino acid derivatives via the diastereoselective Pd(II)‐catalyzed prochiral β‐C(sp3)‐H arylation of racemic (DL), and enantiopure L‐ and D‐carboxamides of amino acids with 5‐iodoindoles. Independently, indole‐based compounds and amino acid motifs are important small molecules in organic synthesis and drug discovery research. This report aimed to contribute to enriching the library of indole‐based unnatural amino acid derivatives. The synthesis of examples of C5‐indole motif‐installed amino acid derivatives comprising norvaline, phenylalanine, leucine, norleucine, 2‐aminooctanoic acid having anti‐stereochemistry was accomplished. The synthesis of the C5‐indole motif‐containing non‐α‐amino acid (aminoalkanoic acid) derivatives was also reported. The C‐H arylation reactions of racemic and enantiopure carboxamides with 5‐iodoindoles afforded the corresponding indole unnatural amino acid derivatives with good diastereoselectivity (anti, dr >95:5 and enantiopurity (er up to 99:1). Removal of the directing group (8‐aminoquinoline), phthalimide group and preparation of indole amino acid esters and free amino group‐containing indole unnatural amino acid motifs were also shown. The utility of the methodology was shown by synthesizing indole motif‐containing pyrrolidone and peptide molecules. The stereochemistry of the major (anti) diastereomers was ascertained from the X‐ray structure of a representative compound.

Abstract:We report the synthesis and theoretical investigations of three novel pseudo-peptide molecules derived from 2-amino-2-(1H-indole-2-yl) acetamides. The compounds were subjected to spectroscopic characterization ($^1$H, $^{13}$C-NMR and MS) and their chemical, electronic, and optical properties have been investigated. To ascertain their potential pharmacological applicability, the prospective reactive centers and molecular sites prone to interaction with water were identified along with possible sensitivity to autoxidation. Further, we have studied the optical response in the presence of different solvents and compared the electronic and optical properties of the pristine molecules. We highlight the subtle dependence of the properties on the structure and composition of these pseudo-peptides. Our results indicate that these molecules have high pharmaceutical potential and could serve as lead components in new drug formulations.
[Synthesis and AchE Inhibitory Activity of 2-Phenoxy-indan-1-one Derivatives].

Abstract:AIM:To design and synthesize novel AchE inhibitors.METHODS:The condensation of 2-bromo-5, 6-dimethoxy-indan-1-one with various aminoalkyl phenols in the presence of K2CO3 and acetonitrile gave the corresponding title compounds, and the in vitro AchE and BchE inhibitory activities were evaluated by the modified Ellman method.RESULTS:Sixteen novel target compounds 8a - p were synthesized, their structures were confirmed by 1H NMR, MS, IR and elemental analysis. Preliminary pharmacological test demonstrated that most of these compounds displayed high AchE inhibitory activities, the IC50 of the most potent inhibitor 8h was 50.0 nmol x L(-1), similar to that of Huperzine A (IC50 = 53.0 nmol x L(-1)), while all the compounds were almost inactive against BchE.CONCLUSION:2-Phenoxy-indan-1-one derivatives exhibit high activities of AchE inhibition and are worthy of further investigation.

Abstract:A new practical approach to prepare diethyl N-{4-[N-methyl-N-(3-(2,4-dioxo-1,2,3,4-tetrahydropyrimidin-5-yl)amino-2-hydroxypropyl)amino]benzoyl}-L-glutamate and its derivatives has been realized from ethyl 4-aminobenzoate (4a) and diethyl N-(4-aminobenzoyl)-L-glutamate (4b). Six novel 5-(substituted)amino pyrimidines 9 similar to 11 were synthesized via methylation, allylation, bromohydroxylation, epoxidation, ring-opening, deprotection reaction and so on, and were characterized by (1)H NMR, (13)C NMR and MS techniques. The inhibitory activities against human dihydrofolate reductase of the compounds have been determined. The results showed that the bioactivities of the compounds were related with L-glutamate side chain.

Abstract:Herein, we report an unprecedented intramolecular cross-nucleophile coupling strategy of indole tethered β-amino acrylates using a catalyst system combining λ3-iodanes and Lewis acids to achieve the chemodivergent synthesis of three unique alkaloid skeletons. It was worth noting that the acquisition of spiroindolenines and azepino[4,5-b]indoles derivatives was switchable with choice of the Lewis acids. Moreover, the polycyclic spiroindolines containing a lactone fragment could also be accessed for the first time via cross-nucleophile coupling cascade intramolecular condensation sequence.

Abstract:Chiral N-alkylated indoles are privileged structural motifs found in many natural products and pharmaceuticals and a search for effective methods with which to prepare chiral N-alkylated indoles is ongoing. Efficient syntheses of enantioenriched N-alkylated indoles has been developed but not with standing these advances, the development of a facile method to prepare chiral N-alkylated indoles from simple and available starting materials under mild conditions is still needed. In this paper, we report a nitrogen replacement process driven by dearomatization that directly incorporates natural a-amino acids into 2-alkynylanilines leading to the formation of a variety of multi-functional chiral N-alkylated indoles.

Abstract:Indoles, a group of alkaloids, showed a wide range of biological activities. Recently, there is a growing tendency to develop some new methods for the synthesis of novel indoles derivatives and to study their biological properties. In this paper, we like to report the design and synthesis of a novel series of tricyclic indoles through several efficient reactions, such as acylation, Pictet-Spengler reaction (P-S reaction) and so on. All these new target compound were synthesized in seven steps with the total yield of 11%-15%, All the products and important intermediates were characterized by 1HNMR and LC-MS spectrum.

Abstract:An simple and efficient procedure for the synthesis of novel 6H‐furo[3,2‐e]indole derivatives via one‐pot three component reactions. A one‐pot, three‐component reaction of ethyl 5‐aminobenzofuran‐2‐carboxylate, arylglyoxals and cyclic 1,3‐dicarbonyl compounds in the presence of acetic acid as an organocatalyst in ethanol medium under reflux conditions provided a series of new 6H‐furo[3,2‐e]indole derivatives in good yields.

Abstract:Objective: In this paper we have reported the synthesis of novel chromeno[2,3- d ]pyrimidine-2,4(3 H )dione derivatives ( IVa–IVh ) by three component reaction between barbituric acid, indole and aromatic aldehydes useing L-Proline as a catalyst. Methods: The structures of synthesized compounds were confirmed by spectroscopic techniques. The synthesized compounds were examined for Acetyl CoA inhibitory activity. Results and Discussion: Compound ( IVb ) showed good activity as compared to other compounds. Also, the synthesized compounds were examined for Anti-inflammatory activity against MMP-2 and MMP-9 using gelatin zymography; in which compound ( IVd ) exhibited very good efficacy viz inhibited MMP-2 and MMP-9 to 80 and 50%, respectively. Furthermore, the molecular docking studies were carried out using a 1PG4 receptor. Conclusions: From the molecular docking study it was found that compound IVa exhibits a best dock confirmation with higher binding affinity (−11.0 kcal/mol) by establishing 3 hydrogen bonds.

Abstract:A new procedure has been developed for synthesizing enantiomerically pure beta-amino acids, alpha-hydroxy-beta-amino acids and certain alkaloids from aspartic acid. By protection, anhydride formation and regioselective reduction, L-aspartic 10 is converted to the N-tosylamino lactone 12. Hydroxylation of 12 by an oxaziridine gives the trans-2-hydroxy-3-N-tosylamino derivative 20. Opening of 12 and 20 by trimethylsily iodide and ethanol affords the iodo-homoserine esters 13 and 21 respectively. Submission of 13 and 21 to Gilman reagents followed by saponification and deprotection gives 4-substituted 3-amino and 3-amino-2-hydroxybutyric acids (15 and 23), exemplified by the syntheses of cyclohexylnorstatine (25), and the components of bestatin (23, R=Ph) and microginin (14). Certain beta-amino acids are transformed into solenopsin A (33) and indolizidine 209D (41).

Abstract:We have developed a high yielding synthesis of indolizine and directly elaborated the molecule into three optically active indolizinylalanine regioisomers. The protocols exploit metal catalyzed coupling of indolizinyl-halides with organozinc reagents derived from carbamoylated iodoalanine esters. The scalable protocols provide products in a form amenable to solid-phase peptide synthesis (SPPS). When incorporated into peptides, the indolizine heterocycle is more basic and markedly less nucleophilic than tryptophan. Its protonated vinylpyridinium form is deeply colored in solution while the neutral heterocycle is highly fluorescent. The fluorescence quantum yield of indolizine exceeds that of indole and aza-indoles in water, suggesting that indolizinylalanines could be powerful optical probes of protein structure and dynamics, functioning as true tryptophan isosteres.

Abstract:Herein, we report a one-pot greener methodology for the synthesis of 3-(1 H -indol-3-yl)-2-phenyl-1 H -benzo[ f ]indole-4,9-dione derivatives by the multicomponent reaction of arylglyoxal monohydrate, 2-amino-1,4-naphthoquinone, and indole in acetonitrile medium under reflux conditions in the presence of 10 mol% sulfamic acid as a catalyst in 20–30 min of reaction time. Three new bonds have formed (2 C-C, 1 C-N) in this methodology. Bioactive moieties such as indole, pyrrole and naphthoquinone are present in our product. This methodology is also applicable in gram-scale synthesis. A wide variety of substrates were tested to find the generality of the methodology and good yield of the products were obtained in a very short reaction time. Along with the operational simplicity of the methodology, purification process of the products is easier by simple recrystallization process. All the synthesized products were characterized by spectroscopic techniques such as FTIR, 1 H NMR, 13 C NMR, and HRMS.
