Abstract:A general and transition-metal-free method for the construction of the carbazol-4-amine motif via a vinylogous Michael addition/cyclization/isomerization/elimination reaction of 3-nitroindoles with alkylidene malononitriles has been developed. This novel methodology allows the facile synthesis of a series of di- and trisubstituted carbazol-4-amine derivatives in moderate to good yields. A gram-scale experiment was successfully performed, highlighting the practicability of this method. Moreover, this strategy is also applicable to 3-nitrobenzothiophene, affording the corresponding dibenzo[ b, d]thiophen-1-amine derivatives in moderate yields.

Abstract:We describe here a Cu-catalyzed and 4-OH-TEMPO-mediated sequential dehydrogenation/aza-Michael addition/annulation cascade reaction for the construction of N-alkyl 2-arylindoles from facilely available saturated ketones and 2-arylethynylanilines. This reaction shows high regioselectivity and tolerates a variety of functional groups. Moreover, 3-alkyl-substituted indoles can also be achieved when using 2-alkylethynylanilines as starting materials.

Abstract:2-Alkynylanilines are attractive starting materials in indole synthesis because of their ready availability. Herein, a one-pot stepwise procedure is reported for efficient construction of multisubstituted oxocino[4,3,2-cd]indoles from 2-alkynylanilines and 2-alkynylbenzaldoximes. The method comprises the oxidative dearomatization of 2-alkynylanilines, the silver-catalyzed [3+3] cycloaddition with 2-alkynylbenzaldoximes, and subsequent thermal radical skeletal rearrangement and aromatization.

Abstract:Dearomatized indole derivatives bearing a C3- or C2-stereocenter exist ubiquitously in natural products and biologically active molecules. Despite remarkable advances in their synthesis, stereoselective and regio-divergent methods are still in a high demand. Herein, a Pd-catalyzed intermolecular asymmetric spiroannulation of 2,3-disubstituted indoles with internal alkynes has been developed for the efficient construction of indoline structures with a C2-quaternary stereocenter. Stereospecific aza-semipinacol rearrangement of these indoline derivatives under acidic conditions afforded indolenine products bearing a C3-quaternary stereocenter, where the migrating group could be controlled by the reaction sequence. The asymmetric spiroannulation together with the subsequent aza-semipinacol rearrangement enabled a divergent access to dearomatized indole derivatives with either a C3- or a C2-quaternary stereocenter.

Abstract:Chiral N-alkylated indoles are privileged structural motifs found in many natural products and pharmaceuticals and a search for effective methods with which to prepare chiral N-alkylated indoles is ongoing. Efficient syntheses of enantioenriched N-alkylated indoles has been developed but not with standing these advances, the development of a facile method to prepare chiral N-alkylated indoles from simple and available starting materials under mild conditions is still needed. In this paper, we report a nitrogen replacement process driven by dearomatization that directly incorporates natural a-amino acids into 2-alkynylanilines leading to the formation of a variety of multi-functional chiral N-alkylated indoles.

Abstract:A process that rapidly assembles 4,7-difunctionalized indoles from 2-alkynycyclohexadienimines, sulfinamides, and nucleophiles (amines or alcohols) was developed. The process involves imino exchange, cascade cyclization/1,4-nucleophilic addition/aromatization, and 1,3-migration of the sulfinyl group. The 7-sulfinyl group is easy to convert into the sulfonyl or the thioether group through a simple oxidation and reduction reaction.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:The oxidative dearomatization of para-substituted o-alkynylanilines afforded 2-alkynyl cyclohexadienimines, which can act as active substrates for reaction with electron-rich styrenes. Abstract:The direct coupling of 2-carbonyl-anilines and diazo-cyclopentanones, promoted by a rhodium catalyst and diphenyl phosphate, is reported for the divergent generation of both carbazolones and indolones. Abstract:Three-component reactions of 2-alkynylbenzaldehydes, amines, and indoles catalyzed by AgOTf under mild conditions afford the 1-(1H-indol-3-yl)-1,2-dihydroisoquinolines in good yields. This silver-catalyzed tandem reaction is found to be workable with various indoles, anilines or alkyl amines, and 2-alkynylbenzaldehydes with electron-withdrawing groups attached on the aromatic backbone.

Abstract:The search for efficient and selective methods for the divergent synthesis of multi-substituted aminotetralins is of much interest and importance. We report herein for the first time the diastereoselective [4+2] annulation of 2-methyl aromatic aldimines with alkenes via benzylic C(sp3)−H activation by half-sandwich rare-earth catalysts, which constitutes an efficient route for the divergent synthesis of both trans and cis diastereoisomers of multi-substituted 1-aminotetralin derivatives from readily accessible aldimines and alkenes. The use of a scandium catalyst bearing a sterically demanding cyclopentadienyl ligand such as C5Me4SiMe3 or C5Me5 exclusively afforded the trans-selective annulation products in the reaction of aldimines with styrenes and aliphatic alkenes. In contrast, the analogous yttrium catalyst, whose metal ion size is larger than that of scandium, yielded the cis-selective annulation products. This protocol features 100% atom-efficiency, excellent diastereoselectivity, broad substrate scope, and good functional group compatibility. The reaction mechanisms have been elucidated by kinetic isotope effect (KIE) experiments and the isolation and transformations of some key reaction intermediates.

Abstract:The synthesis of two stereoisomers of hydroindole derivatives was achieved through [3 + 2] annulations of p‐quinamines with nitroalkenes. Catalyst‐free or in the presence of DMAP, the annulation reactions were employed to produce the target products containing four contiguous stereogenic centers. This method enables efficient assessment of multisubstituted and functionalized hydroindole derivatives using readily accessible building blocks and reagents. Furthermore, the anti‐inflammatory activity of some resulting hydroindoles was evaluated, with three derivative products showing effective inhibition of NLRP3 inflammasome activation.

Abstract:Presented herein is a switchable synthesis of naphthalenone spiroindanediones or 4‐acyl‐1‐naphthols via condition‐controlled divergent cascade reactions of aryl enaminones with diazoindanediones. When the reaction was carried out in the presence of an oxidative additive, naphthalenone spiroindanedione was formed through C(sp2)–H alkylation followed by simultaneous spiroannulation and elimination of dimethylamine. When the reaction was carried out in the presence of an acid additive, on the other hand, 4‐acyl‐1‐naphthol was formed through ring opening of the spiroindanedione moiety attacked by the in situ released dimethylamine. In addition, the reactions are suitable for gram‐scale applications and compatible with a variety of substrates. Moreover, the naphthol products allow structural elaborations and hybridization with clinic drug, thus further enhancing the value of this developed method.
