Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:A new synthesis of L-norvaline is described. Using valeric acid as the raw material, alpha-brominevaleryl chloride was achieved by acyl chlorination and alpha-position bromination in one-pot with the yield of 84.7%. The yields of the following ammoniation of alpha-brominevaleryl chloride and resolution of DL-norvaline were 88.7% and 26.7%, respectively. D-Norvaline was also obtained in a similar method from the waste resolution solution. Other optical active amino acids, valine, alpha-aminobutyric acid and alanine were also synthesized in similar ways.

Abstract:The sodium salts of(S)-2-(1-imidazolyl)alkanoic acids(2) were prepared by the cyclization of natural amino acids with formaldehyde aqueous solution and glyoxal aqueous solution under basic condition.The sodium salts(2) were esterified to form(S)-2-(1-imidazolyl)alkanoates(3a~3g).The(S)-2-(1-imidazolyl)alkanols(4a~4e) were obtaind by reduction of 3a~3e via NaBH_(4) with satisfactory yields.The structures of the compounds(3a~3g,4a~4e) were confirmed by IR,~(1)H NMR,MS,and elemental analysis.

Abstract:A practical and cost-effective synthesis of (2-aminopyrimidin-5-yl) boronic acid 1b has been developed. Key features of the synthesis include the inexpensive in situ protection of the amine via bis-silylation using TMSCl followed by metal–halogen exchange using n-BuLi and trapping with B(Oi-Pr)3. The water-soluble boronic acid is isolated by a well-designed acid–base sequence providing the target in 80% yield and high purity for the two-step process. The large-scale (15 kg) implementation of a Suzuki–Miyaura borylation to form the pinacol boronic ester is also described.

Abstract:An efficient method for the synthesis of N-Cbz-β-aminoalkanesulfonamides was described. N-Cbz-β-aminoalkanesulfona-mides were readily prepared in good yields from a variety of amino alcohols, including optically active ones, via N-Cbz protection with benzyl chloroformate, Mitsunobu esterification reaction with thiolacetic acid, N-chlorosuccinimide oxidation, and ammonolysis process.

Abstract:Enantiopure N-benzyloxycarbonyl-protected and free 3-substituted homotaurines were synthesized from naturally occurring amino acids via N-benzyloxycarbonyl protection, Arndt–Eistert homologation, reduction, esterification with thioacetic acid, and oxidation with performic acid. The current method is a convenient, practical, and salt-free method for the synthesis of enantiopure 3-substituted homotaurine with moderate to good yields.

Abstract:Abstract: A protocol for the Chemoselective N-Boc protection of various types of amines has been developed. This includes heteroaryl, aliphatic, and alicyclic amines. The process makes use of malic acid as a catalyst and operates efficiently at ambient temperature without the need for solvents. This technique has been proven to effectively protect a wide range of functionalized amines containing both electron-donating and electron-withdrawing substituents. The benefits of this method include its fast reaction rate, high selectivity, excellent yield, catalyst recyclability, and environmentally friendly conditions. background: A green, efficient, and solvent-free reaction enlarge for Chemoselective N-Boc protection of aromatic, aliphatic, and heterocyclic amine using malic acid-catalyzed under ambient temperature. The described technique successfully performs the N-Boc on a wide range of functionalized aliphatic, aromatic, and hetero-aromatic amines. The advantage of this method has studied the different types of amines that contain electron-donating as well as electron-withdrawing substituents. The reaction works by rapid reaction rate, high selectivity, solvent-free, excellent yield, catalyst recycle, and greener route. This work presents a green, economical, and eco-friendly protocol. result: In the present work to investigate the scope and disadvantages of this solvent-free N-ter-butoxycarbonylation of amine derivatives catalyzed under malic acid, we can study with the model the reaction of aniline react with di-ter-butyl carbonate (1:1 mmol) and malic acid (15 mol%) under solvent-free reaction conditions to give ter-butyl-phenyl carbonate. (Scheme 2) The reaction was completed within 5 min at room temperature and the corresponding N-Boc protected aniline was obtained in 98 % yield. To appreciate the role of malic acid on the N-Boc protection of amine, we have analyzed the reaction of aniline with di-ter-butyl carbonate in catalyst-free conditions at room temperature. It should be noted that the catalyst-free condition and reaction time of 12 h, were obtained in a 25 % yield. Whereas after 24 hours, it was observed to increase only 37 % yield. All amines derivatives undergo reaction at room temperature without a catalyst for 24 h. The N-Boc protection of amine under the malic acid applies to increases yield and reaction time as compared to the absence of a catalyst.

Abstract:Chiral α-aminoboronic acids and their derivatives are generally useful as bioactive compounds and some have been approved as therapeutic agents. Here we report a NiH-catalysed asymmetric hydroamidation process that with a simple amino alcohol ligand can easily produce a wide range of highly enantioenriched α-aminoboronates from alkenyl boronates and dioxazolones under mild conditions. The reaction is proposed to proceed by an enantioselective hydrometallation followed by an inner-sphere nitrenoid transfer and C–N bond forming sequence. The synthetic utility of this transformation was demonstrated by the efficient synthesis of a current pharmaceutical agent, Vaborbactam.

Abstract:N-Benzyloxycarbonyl (Cbz)-protected 2-aminoalkanesulfonyl chlorides are useful building blocks for the synthesis of sulfonopeptides, which are receptor ligands and enzyme inhibitors, and are prepared by the coupling reaction of N-protected aminoalkanesulfonyl chlorides with amino acid or peptide esters. Various N-Cbz-protected 2-aminoalkanesulfonyl chlorides with functionalized side chains were synthesized through the radical addition of different xanthates to benzyl N-allylcarbamate and subsequent oxidative chlorination with tert-butyl hypochlorite under neutral conditions. A mechanism for the oxidative chlorination is proposed. This is a useful and convenient strategy for the synthesis of N-Cbz-protected 2-aminoalkanesulfonyl chlorides with diverse functionalized side-chains.

Abstract:Aim To synthesize the tripepide Weinreb amide Boc-Asp(OBzl)-β-Ala- Asp(OBzl)-N(OMe)Me (7) as a useful precursor of aspartyl peptide aldehyde derivatives; Methods DCC, IBCF method was used for preparation of Weinreb amide; N-hydroxysuccinimide activated ester was used in peptide synthesis; and Boc as N-protecting group of amino acid. Results Boc-Asp(OBzl)-N(OMe)Me (3), Boc-β-Ala-Asp(OBzl)-N(OMe)Me (5), and Boc-Asp(OBzl)-β-Ala-Asp (OBzl)-N(OMe)Me (7) were synthesized successfully. Conelusion An useful precursor of tripeptide aspartyl aldehydes was synthesized.

Abstract:The direct catalytic α−hydrocarbylation of readily available amino acids with halohydrocarbons is one of the most straightforward methods leading to α,α−disubstituted non-proteinogenic α−amino acid compounds. However, all the reported methodologies depend on N-unprotected amino acids as starting materials. Herein, we report on three highly efficient aldehyde-catalyzed direct α−hydrocarbylations of N-unprotected amino acid esters with aryl-, allyl-, and benzyl halides. By promoting a simple chiral BINOL-aldehyde catalyst or combining catalysts of a chiral aldehyde and Lewis acid ZnCl2, the asymmetric α−arylation, α−allylation, and α−benzylation of amino acid esters with the corresponding halohydrocarbons proceed smoothly, producing α,α−disubstituted α−amino acids in moderate-to-high yields and good-to-excellent enantioselectivities. The asymmetric α−arylation reaction can be applied in the formal synthesis of the clinical candidate compound (+)-AG-041R. Based on the results given by control experiments, three reaction models are proposed to illustrate the stereoselective-control outcomes.
