Abstract:The marine peptides have been a fertile source for new bioactive molecules, and many possess potent, activity, across a broad spectrum of targets. Their diversity in both biological activity and in chemical complexity has made these secondary metabolites being the focus of much work in recent years. As a fragment, (R)-2-methyloctanoic acid-7-yne was always found in those nature materials. In this article, (R)-2-methyl-7-octynoic acid was synthesized with pentane-1, 5-diol as the start material. Stereogenic carbon(C-2) was obtained through alkylation induced with Evans' chiral auxiliary, and alkyne 13 was got by use of Ohira-Bestmann reagent. Through 13-step reactions the target molecule was prepared with over yield of 17. 1%, and e. e. beyond 99%.

Abstract:α-Hydroxy amides are a class of important intermediates in organic synthesis. It plays an important role in the field of science and medicine. However, the preparation methods of α-hydroxy amides are relatively few, and they are usually enzyme-or Lewis acid-catalyzed reactions. These types of methods are not ideal in operation, yield and/or stereoselectivity. We found a new way to synthesize α-hydroxy amides by transforming amino acids into α-hydroxyl acids, and then by amination of the α-hydroxy acids.

Abstract:A new route for the synthesis of( 3S,4S)-4-amino-3-hydroxy-5-phenylpentanoic acid derivatives was reported. The target compounds with overall yields of 5. 8% ~ 6. 7% were synthesized by condensation with Weinreb amide,reduction,aldol condensation reaction and bromine imitation reaction,using amino-protected L-phenylalanine as starting material. The structures were confirmed by1 H NMR,13 C NMR and ESI-MS. The exploration of reaction conditions showed that the loading amount of D-proline had no evident effect on the yield,but on stereoselectivity. Larger size protecting groups in substrate can enhanced the stereo-selectivity.

Abstract:The synthesis of a marine cytotoxic cyclic depsipeptide obyanamide has been accomplished. The key steps include assembling liner pentapeptide via Yamaguchi esterification and HATU-promoted ring closing. The structure of the synthetic sample was identified by H-1 and C-13 NMR, H-H COSY, HMQC, HMBC, and HRESIMS, but appears to be different from that of the marine natural product.

Abstract:Unnatural polar -aminoxy acid residue with proteingenous hydroxymethyl side chain, a building block of the peptidomimetic foldamer of -aminoxy peptide, was synthesized starting from natural amino acid L-serine. The starting material, L-serine, undergoes a reaction sequence to produce compound 1 in three steps: (1) the neighboring carboxyl group participates in diazotization/bromination to transform the amino group to a bromo group, (2) the C-terminal carboxyl group is protected, and (3) bromide is SN2-displaced by N-hydroxyl phthalimide to introduce a N-O bond. After several conventional deprotection/coupling reactions, compound 1 is easily transformed to an -aminoxy diamide, which can be widely used in peptidomimetics design.

Abstract:The synthesis of 3-(N-ethyl-N-methoxycarbonylmethyl) amino acetanilide via two-step reactions was reported,with 3-aminoacetanilide as starting material.The designed product was yielded by the ethylated compound(from 3-aminoacetanilide reacting with acetaldehyde) reacting with methyl chloroacetate.The optimized reaction conditions were as follows: in the first step,the yield of 3-(Nethyl) amino acetanilide was 90.7% and its purity was up to 97.5%,when the molar ratio of acetaldehyde to 3-aminoacetanilide was 1.0∶1.2 and when 5% Pd/C was used as catalyst under 3.5 MPa of hydrogen atmosphere at 70 ℃.In the second step,the designed product was obtained in 94.6% yield and 96.8% purity with 0.6 equiv.of sodium carbonate as deacid reagent under 70 ℃.

Abstract:Based on the structural features of obyanamide, the target molecule was disconnected into four fragments by applying retrosynthetic analysis method, i.e., N-Me-Phe-N-Me-Val (fragment A), thiazole carboxy acid derivative (fragment B), lactic acid derivative (fragment Q and unnatural 3-aminopentanoic acid (fragment D). The total synthesis of a natural marine cytotoxic cyclic depsipeptide obyanamide was accomplished successfully from synthesized the fragments A, B, C and D. The structures of the synthesized intermediates and product was identified by H-1 NMR, C-13 NMR, HMBC and HRMS (FAB) spectra.

Abstract:2-{[(2 Z )-4-Aryl-2-hydroxy-4-oxobut-2-enoyl]amino}benzoic acids have been synthesized through the reaction of methyl esters of aroylpyrovinic acids with 2-aminobenzoic (anthranilic) acid in glacial acetic acid in the presence of anhydrous sodium acetate. Abstract:On the basis of the total synthesis of obyanamide, 20 analogues of this marine cyclic depsipeptide have been synthesized by (i) preparation of the tripeptide fragments in the western hemisphere using Z/OtBu protocol; (ii) preparation of the dipeptide fragments in the eastern hemisphere using Boc/OMe protocol; and (iii) fragments coupling, removal of protecting groups (Boc and OtBu, in one pot), and macrocyclizaion in the last step. The cytotoxic test showed that three synthetic compounds exhibited moderate activities against HL-60, KB, LOVO, and A549 cell lines. According to the results, the β-amino acid residue was found to play a critical role in the biological activities. Additionally, the ester bond along with the Ala(Thz) moiety was also essential for biological activities. However, it seems too early to draw a conclusion that the N-methylation of Val/Phe can lead to higher or lower cytotoxic activities.

Abstract:Objective: ( S )-2-{3-[(Benzyloxy)carbonyl]-5-oxooxazolidin-4-yl}acetic acid is an useful intermediate in the preparation of β-branched peptide derivatives of aspartic acid. However, we and others observed the formation of side products in the reaction of activated esters of this compound with amino acid derivatives. This suitably protected aspartic acid derivative exists as a mixture of E and Z rotamers that complicates the interpretation of NMR spectra of the products obtaind from it. Therefore, some of the failures in the preparation of β-branched aspartic acid derivatives could be due to misinterpretation of spectral data. Methods: The selectivity of the reaction of pentafluorophenyl ( S )-2-{3-[(benzyloxy)carbonyl]-5-oxooxazolidin-4-yl}acetate with amino acid esters was explored with particular emphasis on the rigorous structure assignment of the products using high-temperature NMR experiments. Results and Discussion: The isomeric products of the reaction were isolated and identified. One of the isomers is an expected dipeptide, and its isomer is an N -(hydroxymethyl-amino)succinimide formed by methylene bridge cleavage. The steric bulkiness of amino acid esters favors dipeptide formation, but the reaction selectivity is quite unpredictable. Conclusions: The protection of the α-carboxyl group of aspartic acid in the form of (5-oxooxazolidin-4-yl)acetic acid derivatives has a number of limitations that must be taken into account, when this derivative is considered for use in the preparation of β-branched peptides.

Abstract:(S)-3-Benzyloxycarbonyl-5-oxo-4-oxazolidineacetic acid (IIa) and (S)-3-benzyloxycarbonyl-5-oxo-4-oxazolidinepropionic acid (IIb) were utilized for the selective protection of side-chain carboxyl group of corresponding α-aminodicarboxylic acids and the synthesis of β-aspartyl and γ-glutamyl peptides as an α-carboxyl protected intermediates. It was found that 5-oxazolidinone ring, the α-carboxyl protecting group, was easily removed under not only the condition of alkaline hydrolysis but also catalytic hydrogenolysis. Thus a facile synthesis of β-aspartyl and γ-glutamyl peptides was achieved by the coupling of IIa and IIb with benzyl ester of amino acids and peptides followed by hydrogenolytic removal of all the protecting groups.

Abstract:A new procedure has been developed for synthesizing enantiomerically pure beta-amino acids, alpha-hydroxy-beta-amino acids and certain alkaloids from aspartic acid. By protection, anhydride formation and regioselective reduction, L-aspartic 10 is converted to the N-tosylamino lactone 12. Hydroxylation of 12 by an oxaziridine gives the trans-2-hydroxy-3-N-tosylamino derivative 20. Opening of 12 and 20 by trimethylsily iodide and ethanol affords the iodo-homoserine esters 13 and 21 respectively. Submission of 13 and 21 to Gilman reagents followed by saponification and deprotection gives 4-substituted 3-amino and 3-amino-2-hydroxybutyric acids (15 and 23), exemplified by the syntheses of cyclohexylnorstatine (25), and the components of bestatin (23, R=Ph) and microginin (14). Certain beta-amino acids are transformed into solenopsin A (33) and indolizidine 209D (41).

Abstract:(S)-4-Amino-5-mercaptopentanoic acid (glutamate thiol, GluSH) is a derivative of glutamic acid with alpha-carboxyl group substituted by methanethiol group. Meanwhile, it could be considered as a derivative of cysteine. This amino acid is an important fragment of marine cytotoxic cyclic depsipeptide apratoxin E. Two facile synthetic methods of (S)-4-amino-5-mercaptopentanoic acid are described. For the first strategy, protected D-cysteine derivate was used as starting material. After coupled with Meldrum's acid, the resulting beta-keto ester underwent reductive elimination and thermal decarboxylative ring closure to afford 5-substituted pyrrolidinone (gamma-lactam). After removal of protecting groups and ring-open, the target compound was obtained in 76.0% overall yield. For the second method, commercial available Boc-Glu(OBzl)-OH was used as starting material. The thiol group was introduced by Mitsunobu reaction after converting the carboxylic acid to its corresponding alcohol. Then the protecting groups of amino, thiol, and carboxylic acid were removed smoothly in one pot to yield (S)-4-amino-5-mercaptopentanoic acid.

Abstract:Aim To synthesize the tripepide Weinreb amide Boc-Asp(OBzl)-β-Ala- Asp(OBzl)-N(OMe)Me (7) as a useful precursor of aspartyl peptide aldehyde derivatives; Methods DCC, IBCF method was used for preparation of Weinreb amide; N-hydroxysuccinimide activated ester was used in peptide synthesis; and Boc as N-protecting group of amino acid. Results Boc-Asp(OBzl)-N(OMe)Me (3), Boc-β-Ala-Asp(OBzl)-N(OMe)Me (5), and Boc-Asp(OBzl)-β-Ala-Asp (OBzl)-N(OMe)Me (7) were synthesized successfully. Conelusion An useful precursor of tripeptide aspartyl aldehydes was synthesized.

Abstract:L-Aspartic acid by tosylation, anhydride formation, and reduction with NaBH4 was converted into (3S)-3-(tosylamino)butan-4-olide (8; Scheme 1). Treatment of 8 with ethanolic trimethylsilyl iodide gave the N-protected deoxy-iodo-P-homoserine ethyl ester 9. The latter, on successive nucleophilic displacement with lithium dialkylcuprates (-->10a-e), alkaline hydrolysis (-->11a-e), and reductive removal of the tosyl group, produced the corresponding 4-substituted (SR)-9-aminobutanoic acids 12a-e (ee > 99%). Electrophilic hydroxylation of 8 (-->19; Scheme 3), subsequent iodo-esterification (-->21; Scheme 4), and nucleophilic alkylation and phenylation afforded, after saponification and deprotection, a series of 4-substituted (2S,3R)-3-amino-2-hydroxybutanoic acids 24 including the N-terminal acids 24e (=3) and 24f (=4) of bestatin and microginin (de > 95%), respectively.

Abstract:2-Azabicyclo[3.1.0]hexane-3-carboxylic acids were obtained via amino protection, 4-dimethylaminopyridine (DMAP) catalytic cyclization, reduction-dehydration into alkene, asymmetric Simmons-Smith reaction and hydrolysis reaction from glutamic acid. The intermediates and target products were characterized by 1H NMR and 13C NMR. (1R,3S,5R)-2-(tert-Butoxycarbonyl)-2-azabicyclo[3.1.0]hexane-3-carboxylic acid and (1S,3S,5S)-2-(tert-butoxycarbonyl)-2-azabicyclo-[3.1.0]hexane-3-carboxylic acid were analyzed by X-ray diffraction as well. The process conditions about DMAP catalytic cyclization reaction, Simmons-Smith reaction and hydrolyzation were discussed. The results show that the yield of (R)-1,2-di-tert-butyl 5-oxopyrrolidine- 1,2-dicarboxyate was 82% at the ratio of n(DMAP)∶n(di-tert-butyl dicarbonate)∶n(pyridine)=0.40∶4.0∶1.0. The ratio of cis/trans-form varied with the reaction time in Simmons-Smith reaction, which gave a best ratio of 6∶1 at 19.5 h. The total yield is 30%, and the de value is 72%.
