Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:A direct method for the preparation of phosphonamidate- and phosphonate-linked phosphonopeptides has been developed. Using this method, both phosphonopeptides were prepared in acceptable yields directly from simple and commercially available chemicals in one-pot reactions of benzyl carbamate, aldehydes, and methyl dichlorophosphite, followed by aminolysis with amino acid esters or alcoholysis with hydroxy esters.

Abstract:Optically active 1-aminoalkylphosphonic acid derivatives were synthesized in moderate yields and optical purities via Mannich-type reactions of benzyl carbamate, aldehydes, and optically pure chlorophosphites. Side chain-functionalized depsiphosphonopeptides were also prepared in satisfactory yields directly from one-pot reactions of benzyl carbamate, aldehydes, and diethyl (R,R)-2-chloro-1,3,2-dioxaphospholane-4,5-dicarboxylate.

Abstract:Phosphonopeptides are important phosphorus analogs of natural peptides and have been widely applied as enzyme inhibitors and antibacterial agents. A series of phosphonodipeptides containing C-terminal α-aminoalkylphosphonic acids was synthesized in satisfactory to good yields conveniently from 2-(N-benzyloxycarbonylamino)alkanamides, aldehydes, and phosphorus trichloride via Mannich-type reaction and subsequent sequential hydrolysis. The reaction mechanism was proposed and verified by (31)P NMR tracing experiments. The current method is an efficient and convenient method for preparation of phosphonopeptides containing C-terminal α-aminoalkylphosphonic acids.

Phosphorus Sulfur and Silicon and the Related Elements

Abstract:Mannich-type condensation of benzyl carbamate, aldehydes, and chlorophosphite has been widely used in the synthesis of 1-(N-benzyloxycarbonylamino)arylmethyl-phosphonic derivatives. Reactions of benzyl carbamate, aldehydes, and cyclic chlorophosphite 2-chloro-1,3,2-dioxaphospholane were conducted in benzene to afford a mixture of benzyl [(2-oxido-1,3,2-dioxaphospholane-2-yl)arylmethyl]-carbamates and 2-hydroxyethyl 1-(N-benzoxycarbonylamino)arylmethylphos-phonate monoesters. 2-hydroxyethyl 1-(N-benzoxycarbonylamino)arylmethylphos-phonate monoesters were obtained exclusively after the addition of water under stirring. The reaction mechanism was discussed. Their mass spectrometric fragmentations were also described.

Abstract:An efficient and straightforward procedure toward the synthesis of amides-substituted 2H-benzopyrans H-benzopyrans has been explored through palladium-catalyzed one-pot aminocarbonylation reaction. By using nitro compounds as attractive nitrogen sources, with Mo(CO)6 6 as both CO source and reductant, a variety of amides-containing 2H- H- benzopyrans were obtained in moderate to high yields with good functional group tolerance. This protocol has advantages including atom-economy, no manipulation of toxic CO gas, and broad substrate scope.

Abstract:Four phosphonodipeptides containing 1-aminoalkylphosphonate were synthesized by applying four different coupling methods. One of them was then converted into monoacid, ethyl hydrogen 1-(N-acetyl-prolyl)amino-2-phenylethyl phosphonate. Three phosphonodipeptides containing ethyl 1w-amino-2-phenylethylphosphonamidate were also prepared by converting phosphono monoacid into phosphonochloridate and reacting it with amino acid ester. Benzyl ethoxyphosphonyl-glycyl-glycine ethyl ester also was synthesized from ethyl glycylglycinate and ethyl benzylphosphonochloridate derived from direct chlorination of diethyl benzylphosphonate with phosphorus oxychloride.

Abstract:Here an unprecedented formal deformylative phosphonylation is disclosed. Under the catalysis of 1 mol % 1,2,3,5-tetrakis(diphenylamino)-4,6-dicyanobenzene (4DPAIPN) and the presence of 200 mol % triethylamine as additive, 4-alkyl-1,4-dihydropyridines (DHPs), derived from aldehydes, underwent phosphonylation smoothly with 9-fluorenyl o-phenylene phosphite, allowing facile synthesis of alkylphosphonates. This strategy is applicable to primary, secondary, and even tertiary DHPs, and exhibits a broad substrate scope and excellent functional group tolerance, thereby enabling the late-stage phosphonylation of a series of naturally-ocurring bioactive and biorelevant molecules.

Abstract:Various alpha,beta-unsaturated (E)-alkenylphosphonamidates were prepared in excellent stereo- and regioselectivities via the electrophilic hydrophosphonylation of cyclic and acyclic aldimines with alkylphosphonochloridates in the presence of base. For unsaturated aldimines, only their respective imine bonds were hydrophosphonylated regiospecifically. The reaction provided a new strategy for the synthesis of (E)-alk-1-enylphosphonamidates via a tandem phosphorylation, deprotonation, and intramolecular 1,5-hydride transfer sequence process from readily available aldimine and alkylphosphonochloridate starting materials. The strategy established the first one-pot electrophilic rather than nucleophilic hydrophosphonylation of aldimines without external hydride donors.

Abstract:An efficient method for the synthesis of organophosphonates through palladium-catalyzed coupling of α-diazo phosphonates with benzyl or allyl halides has been developed. Trisubstituted alkenylphosphonates bearing versatile functional groups can be easily accessed in good yields and with excellent stereoselectivity through this method. Moreover, with similar strategy α-substituted vinylphosphonates can also be attained by the palladium-catalyzed coupling reaction of N-tosylhydrazones and aryl bromides. Migratory insertion of palladium carbene is proposed as the key step in this reaction.

Abstract:(2‐Aminoaryl)iminophosphanes were found to be versatile intermediates for the synthesis of 1‐alkyl‐2‐functionalized benzimidazoles. Readily prepared from o‐nitroaniline derivatives, they reacted efficiently in situ with a variety of electrophiles to form common heterocyclic scaffolds suitable for the synthesis of more complex, biologically interesting molecules. N‐Alkyl‐2‐nitroanilines deoxygenated with tributylphosphine form intermediate 2‐(alkylamino)aryliminophosphoranes, which were subjected, without isolation, to various cyclocondensation reactions with CS2, CO2, alkyl isocyanates, acyl chlorides, anhydrides, or esters. A simple, convenient, one‐pot procedure provided derivatives of unsymmetrically substituted 1‐alkylbenzimidazoles functionalized at C2 in good to excellent yields. The method does not require the use of metals, sensitive catalysts, or pressure.
