Phosphorus Sulfur and Silicon and the Related Elements

Abstract:Mannich-type condensation of benzyl carbamate, aldehydes, and chlorophosphite has been widely used in the synthesis of 1-(N-benzyloxycarbonylamino)arylmethyl-phosphonic derivatives. Reactions of benzyl carbamate, aldehydes, and cyclic chlorophosphite 2-chloro-1,3,2-dioxaphospholane were conducted in benzene to afford a mixture of benzyl [(2-oxido-1,3,2-dioxaphospholane-2-yl)arylmethyl]-carbamates and 2-hydroxyethyl 1-(N-benzoxycarbonylamino)arylmethylphos-phonate monoesters. 2-hydroxyethyl 1-(N-benzoxycarbonylamino)arylmethylphos-phonate monoesters were obtained exclusively after the addition of water under stirring. The reaction mechanism was discussed. Their mass spectrometric fragmentations were also described.

Abstract:Unsymmetrical N-benzyloxycarbonyl-protected 1-amino-1-arylalkylphosphonate mixed diesters were synthesized using a one-pot reaction involving benzyl carbamate, aromatic aldehydes and alkoxydichlorophosphine, followed by treatment with alcohols in the presence of triethylamine. The reactions were followed by 31P NMR and a mechanism is proposed.

Abstract:Optically active 1-aminoalkylphosphonic acid derivatives were synthesized in moderate yields and optical purities via Mannich-type reactions of benzyl carbamate, aldehydes, and optically pure chlorophosphites. Side chain-functionalized depsiphosphonopeptides were also prepared in satisfactory yields directly from one-pot reactions of benzyl carbamate, aldehydes, and diethyl (R,R)-2-chloro-1,3,2-dioxaphospholane-4,5-dicarboxylate.

Abstract:A straightforward method for the synthesis of depsiphosphonopeptides via a Mannich-type multicomponent condensation of simple starting materials, Such as benzyl carbamate, aldehydes, and 1-carbetboxyalkyl phosphorodichloridites, was developed. Compared to previous methods, our strategy provides a more efficient, convenient, and practical route for the preparation of depsiphosphonopeptides under mild reaction conditions with good yields. Such a strategy avoids the initial synthesis of 1-ammoalkylphosphonic acid or 1-aminoalkylphosphonous acid derivatives as starting materials.

Abstract:Four phosphonodipeptides containing 1-aminoalkylphosphonate were synthesized by applying four different coupling methods. One of them was then converted into monoacid, ethyl hydrogen 1-(N-acetyl-prolyl)amino-2-phenylethyl phosphonate. Three phosphonodipeptides containing ethyl 1w-amino-2-phenylethylphosphonamidate were also prepared by converting phosphono monoacid into phosphonochloridate and reacting it with amino acid ester. Benzyl ethoxyphosphonyl-glycyl-glycine ethyl ester also was synthesized from ethyl glycylglycinate and ethyl benzylphosphonochloridate derived from direct chlorination of diethyl benzylphosphonate with phosphorus oxychloride.

Abstract:Methyl 3-deoxy-3-(diphenylphosphino)-4,6-O-benzylidene-α-D-altropyranoside (1) and methyl 2-deoxy-2-(diphenylphosphino)-4,6-O-benzylidene-α-D-altropyranoside (2) were prepared from methyl 2,3-anhydro-4,6-O-benzylidene-O-D-mannopyranoside and methyl 2,3-anhydro-4,6-O-benzyl-idene-α-D-allopyranoside,respectively,via regioselective and stcreospecific ring-opening reactions in high yields.Compounds 1 and 2 were oxidized to give the corresponding phosphine oxides (3 and 4).

Abstract:Phosphinopeptides are a class of unnatural peptides containing a tetrahedral phosphorus atom and have potential for use as enzyme inhibitors. A series of phosphinopeptides were synthesized via the Mannich-type reaction of aryldichlorophosphines, aldehydes, and N-protected amino amides or peptide amides and subsequent aminolysis with amino esters or peptide esters. The current method named the Mannich ligation is an efficient route to synthesis of phosphinopeptides through convergent condensation.

Abstract:Phosphonopeptides are important phosphorus analogs of natural peptides and have been widely applied as enzyme inhibitors and antibacterial agents. A series of phosphonodipeptides containing C-terminal α-aminoalkylphosphonic acids was synthesized in satisfactory to good yields conveniently from 2-(N-benzyloxycarbonylamino)alkanamides, aldehydes, and phosphorus trichloride via Mannich-type reaction and subsequent sequential hydrolysis. The reaction mechanism was proposed and verified by (31)P NMR tracing experiments. The current method is an efficient and convenient method for preparation of phosphonopeptides containing C-terminal α-aminoalkylphosphonic acids.

Abstract:A direct method for the preparation of phosphonamidate- and phosphonate-linked phosphonopeptides has been developed. Using this method, both phosphonopeptides were prepared in acceptable yields directly from simple and commercially available chemicals in one-pot reactions of benzyl carbamate, aldehydes, and methyl dichlorophosphite, followed by aminolysis with amino acid esters or alcoholysis with hydroxy esters.

Abstract:The negative ion mass spectrometry of N-benzyloxycarbonyl and N-ethoxycarbonyl 1-aminoarylmethylphosphonic methyl, ethyl, and phenyl monoesters was investigated under electrospray ionization conditions. Their fragmentation pathways are proposed and supported by collisional activated dissociation product-ion spectrometry. All of the deprotonated molecules preferentially eliminate a molecule of benzyl alcohol or ethanol first to yield isocyanato-alkylphosphonate anions, which further generate phosphonate ions by the loss of carbon monoxide, phenol or alcohol, carbon monoxide plus arenes. The isocyanato-sulfonate anions can further cyclize to generate 3-aryl-2-phenoxy-1,2,4-oxaphosphazolidine-2,5-dione amide anions, which can undergo rearrangements by the loss of carbon dioxide, metaphosphorous acid phenyl ester, or carbon dioxide plus metaphosphorous acid phenyl ester, respectively, to give rise to nitrogen-containing anions. The title compounds show an obviously different fragmentation in the negative ionmode from that in the positive ionmode under the electrospray ionization conditions.

Abstract:Chiral hydroxyl monophosphane 3 [(2S,3S,4S,5S)-3,4-dihydroxy-2, 5-dimethyl-1-phenylphospholane] and bisphospholanes 5a [1,2-bis[(2S, 3S,4S,5S)-3,4-dihydroxy-2,5-dimethylphospholanyl]benzene] and 5b [1, 2-bis[(2S,3S,4S,5S)-2,5-diethyl-3,4-dihydroxyphospholanyl]benzene] were synthesized from readily available D-mannitol in high yields. Strategies for protection and deprotection of OH-groups in the presence of phosphines have been explored. Rate acceleration in the Baylis-Hillman reaction was observed when a hydroxyl phosphine was used as the catalyst. Rhodium complexes with chiral bisphospholanes are highly enantioselective catalysts for the asymmetric hydrogenation of various kinds of functionalized olefins such as dehydroamino acid derivatives, itaconic acid derivatives, and enamides. An interesting feature of the hydroxyl phospholane system is that hydrogenation of some substrates can be carried out in water with >99% ee and 100% conversion (e.g., itaconic acid).

Abstract:The palladium-catalyzed coupling-cyclization of monoesters of 1,2-allenyl phosphonic acids with allylic halides was studied. 4-Allyl-2,5-dihydro[1,2]oxaphosphole 2-oxides were smoothly afforded. Multi substituted 1-ethoxy-1,3-dihydro[2,1]benzoxaphosphole 1-oxides were prepared from 3,4-diallyl-2,5-dihydro[1,2]oxaphosphole 2-oxides by a one-pot tandem reaction.

Abstract:A phosphate ester reactive emulsifier is synthesized via multi steps reaction which uses phosphorus oxychloride, hydroxypropyl acrylate(HPA), and triethylamine as reactants. The optimum technical conditions are acquired by discussing different feed ratios, reaction time and temperature. The product is characterized by IR spectroscopy. The results indicate that the yield of methyl acryloyl oxygen propyl phosphate ester will reach 98.80% when the feed ratio is: n(POCl3)∶n(triethylamine) : n(HPA) = 1.1∶1.3∶1, the reaction time is 1.5h, the reaction temperature is 25℃, and the second step hydrolysis reaction temperature is 0 to 15℃, and the hydrolyzed material ratio is n(intermediate product)∶n(triethylamine)∶n(water) = 1∶1. 4∶1. 2.
