Abstract:By using a D-fructose-derived chiral ketone or a D-mannitol-derived chiral amine as catalyst, the organocatalytic asymmetric epoxidation of cis-1-propenylphosphoric acid with 30% aqueous hydrogen peroxide in H(2)O-CH(3)CN mixture (similar to 80:20) afforded (1R, 2S)-(-)-(1,2)-epoxypropyl phosphoric acid (fosfomycin). Asymmetric epoxidation was carried out at 0 degrees C for 72 h to achieve 100% conversion with a maximum enantiomeric excess (e.e.) of 74%. (c) 2008 Elsevier B.V. All rights reserved.

Abstract:A palladium-catalyzed reaction of an aryl aryl imidazolylsulfonate with an H-phosphonate diester in the presence of DPPP and (Pr2NEt)-Pr-i is described. The reactions proceed efficiently to generate the corresponding arylphosphonates in good to excellent yields. The advantages of this method include good substrate generality, high efficiency, and experimental ease.

Abstract:Addition of P-H species to carbonyl groups, namely the Pudovik reaction, normally delivers hydroxyl phosphorus compounds, along with phosphate byproducts in some cases. A few controllable systems starting from phosphites were set up to mainly provide the phosphates. Herein, we present a highly selective protocol starting from phosphonate precursors leading to phosphinate derivatives. Enantioenriched phosphinates were successfully achieved from chiral phosphine oxide precursors. Experimental and theoretical investigations were conducted to understand the mechanistic details.

Abstract:Here an unprecedented formal deformylative phosphonylation is disclosed. Under the catalysis of 1 mol % 1,2,3,5-tetrakis(diphenylamino)-4,6-dicyanobenzene (4DPAIPN) and the presence of 200 mol % triethylamine as additive, 4-alkyl-1,4-dihydropyridines (DHPs), derived from aldehydes, underwent phosphonylation smoothly with 9-fluorenyl o-phenylene phosphite, allowing facile synthesis of alkylphosphonates. This strategy is applicable to primary, secondary, and even tertiary DHPs, and exhibits a broad substrate scope and excellent functional group tolerance, thereby enabling the late-stage phosphonylation of a series of naturally-ocurring bioactive and biorelevant molecules.

Abstract:A direct method for the preparation of phosphonamidate- and phosphonate-linked phosphonopeptides has been developed. Using this method, both phosphonopeptides were prepared in acceptable yields directly from simple and commercially available chemicals in one-pot reactions of benzyl carbamate, aldehydes, and methyl dichlorophosphite, followed by aminolysis with amino acid esters or alcoholysis with hydroxy esters.

Abstract:A convenient synthetic method was reported for the preparation of N-phthaloyl dehydroamino acid esters from easily accessible vinyl bromides (or vinyl tosylate) and potassium phthalimide. Rh-catalyzed asymmetric hydrogenation of these substrates with TangPhos gave the products with good to excellent enantioselectivities (up to 99% ee).

Abstract:We successfully achieved the phosphorylation of secondary aromatic alcohols with H-phosphine oxides (less developed system) using phosphotungstic acid as a catalyst in dimethyl carbonate. Abstract:A general and practical cross-dehydrogenative coupling protocol between readily available trisubstituted α,β-dehydro α-amino carboxylic esters and H-phosphites is described. This C(sp2)–H phosphorylation reaction proceeds with absolute Z-selectivity promoted by silver salt in a radical relay manner. Abstract:A convenient enzymatic method is presented for the preparation of optically pure 2-hydroxy-2-arylethanephosphonates. It is proved that 2-butyryloxy-2-arylethanephosphonates can be enantioselectively hydrolyzed in diisopropyl ether equilibrated with water to give both isomers in high yield and excellent enantiomeric excess.

Abstract:A protocol for the acid-catalyzed synthesis of phosphorylated dihydrodibenzoxa-/thiazepine derivatives from dibenzo[b,f][1,4]oxa-/thiazepines has been established. The reaction appears compatible with a range of functional groups in both the dibenzo[b,f][1,4]oxa-/thiazepine, H-phosphonate and especially diarylphosphine oxide phosphorylation partners, affording moderate to excellent yields of the corresponding products. The present method further expands the utility of Pudovik reaction.

Abstract:Aryl, heteroaryl, and benzylic electrophiles can be coupled to in situ-generated alkylphosphinates (ROP(O)H-2) in moderate to good yields using palladium catalysis. For the first time, electrophiles other than simple aryl iodides can be employed in an experimentally straightforward, one-pot reaction. The direct formation of H-phosphinic esters avoids the separate esterification of H-phosphinic (phosphonous) acids and is particularly useful in the case of nitrogen-containing heteroaromatics. Abstract:Coupling of N-phosphoamino acids 2 and amino acid methyl esters by dichlorotriphenylphosphorane 3 produced N-phosphopeptide methyl esters 5, which were sequentially hydrolyzed in triethylamine or dilute NaOH aqueous solution. The target products N-phosphodipeptides 6 were obtained in reasonable yields and high purity after isolation using extraction method. The convenient and efficient approach could be generally used for synthesis of polypeptides.

Abstract:We report a facile synthetic strategy toward CH2-substituted phosphothreonine mimetics. Herein, inexpensive valine with a directing group was converted into homothreonine via palladium-catalyzed gamma-methyl C(sp(3))-H bond activation, followed by construction of a phosphorus-carbon bond via the well-developed Appel reaction and Michaelis-Becker reaction with a total yield of 30%. Furthermore, the derived mimetic was applied for solid-phase synthesis of two phosphopeptide inhibitors. This efficient synthesis provides a chance to prepare not only phosphopeptides but also phosphoproteins resistant to phosphatases.

Abstract:Chiral organophosphorus compounds possessing a P-stereogenic center have been widely used as agricultural chemicals, pharmaceuticals, organocatalysts, and ligands for transition-metal catalysis. P-Stereogenic intermediates bearing a tert -butyl(methyl)phosphino group are important for the preparation of several commonly used diphosphine ligands. However, previously reported synthetic methods used hazardous starting materials and are difficult to operate. In order to overcome these limitations, a new and convenient synthesis for a number of P-stereogenic intermediates possessing a tert -butyl(methyl)phosphino group has been developed. The reported route relies on an air- and moisture-stable secondary phosphine oxide prepared from a readily available starting material, trichlorophosphane.

Abstract:A microwave-assisted synthesis of dialkyl and cyclic H-phosphonates via bis(2,2,2 trifluoroethyl) phosphonate (BTFEP) is described. This method enables the synthesis of various cyclic H-phosphonates and hetero-substituted dialkyl H-phosphonates by simple alcoholysis under non-inert and additive-free conditions. Short reaction times and the requirement for only stoichiometric amounts of alcohol render this method attractive for synthetic applications.
