Catalyst-free synthesis of phytosterol diacid monoesters and their competitive effect on the solubilization of cholesterol in model bile mixed micelles

Abstract:Phytosterols have received an extensive attention owing to their excellent cholesterol-lowering activity and the role in cardiovascular diseases prevention. Phytosterol diacid monoesters, the important intermediates in the structural modification of free phytosterols, were usually obtained by chemical method with catalyst. The aim of this research was to provide an eco-friendly approach for the preparation of phytosterol diacid monoester and explore the possible mechanism of their hypocholesteremic effect using model bile mixed micelles. Catalyst-free synthesis of β-sitosterol with succinic anhydride, maleic anhydride, and glutaric anhydride was investigated and optimized. The yields of three β-sitosterol diacid monoesters all reached above 94% under optimum reaction conditions and their chemical structures were confirmed further by mass spectroscopy, Fourier transform infrared spectroscopy, and nuclear magnetic resonance spectroscopy. Compared with β-sitosterol, β-sitosterol diacid monoesters possessed the increased thermal stability (up to an above 355.28°C) and improved solubility in binary mixed micelle system (exceeding 1.7 mM) and similar cholesterol-lowering activity (7.78%-11.05%). Moreover, β-sitosterol diacid monoesters showed the competitive effect on solubilization of cholesterol due to their enhanced micellar incorporation capacity. Overall, the competition with cholesterol in vitro gives useful information about the cholesterol-lowering activity of phytosterol diacid monoesters, which will further expand their applications in the food industry and health. PRACTICAL APPLICATION: Modified phytosterols, phytosterol diacid monoesters, were successfully synthesized in petroleum ether without catalyst. These products enhanced the thermal stability of free phytosterols and reduced the dissolution capacity of cholesterol in model bile mixed micelles due to competitive solubilization. These results indicated that phytosterol diacid monoesters represented cholesterol-lowering potential, which might further expand the applications in the food industry and health.

Abstract:A new intermediate containing dihydrochalcone moiety, i.e., bis(dihydrochalcone) diphenyl ether (BHCDPE), was first synthesized by the Friedel-Crafts alkylation reaction of chalcone and diphenyl ether (DPE). Subsequently, a series of novel poly(aryl ether ketone) (PAEK) polymers were prepared by further Friedel-Crafts polycondensation of BHCDPE and DPE with isophthaloyl dichloride (IPC). It was observed that BHCDPE exhibited strorkg reactivity during Friedel-Crafts polycondensation, i.e., one non-conjugated gamma-carbonyl would not decrease the Friedel-Crafts acylation's reactivity of corresponding benzene rings, such that the dihydrochalcone moiety could be successfully incorporated into the backbone of PAEK to form novel PAEKs with dihydrochalcone moiety. The synthesis route was a continuous procedure and displayed inexpensive and commercially available of the raw materials, mild reactive conditions, as well as high yields due to the absence of complicated process of intermediate treatment. The polymers' structures were characterized by FT-IR, H-1-NMR and XRD analysis. Moreover, obtained polymers maintained good thermal properties, and incorporation of a dihydrochalcone moiety significantly improved polymer solubility in organic solvents, which was advantageous for further functionalization, processing and interactions with other substances. Thus, the designed synthesis route was appropriate to obtain novel soluble PAEKs containing dihydrochalcone moieties, which were expected as new potential candidates for more useful materials in various fields.

Abstract:Here an unprecedented formal deformylative phosphonylation is disclosed. Under the catalysis of 1 mol % 1,2,3,5-tetrakis(diphenylamino)-4,6-dicyanobenzene (4DPAIPN) and the presence of 200 mol % triethylamine as additive, 4-alkyl-1,4-dihydropyridines (DHPs), derived from aldehydes, underwent phosphonylation smoothly with 9-fluorenyl o-phenylene phosphite, allowing facile synthesis of alkylphosphonates. This strategy is applicable to primary, secondary, and even tertiary DHPs, and exhibits a broad substrate scope and excellent functional group tolerance, thereby enabling the late-stage phosphonylation of a series of naturally-ocurring bioactive and biorelevant molecules.

This method has the merits of easy operation and high yields. Abstract:Three single chain amphiphilic cholesterol derivatives with amino acid as hydrophilic groups, I, II and III, were synthesized. When they were hydrated in water, they can all form lamellae structures as evidenced by DSC measurement. The lamellae states of II and III can be converted to vesicles upon ultrasonication of the diluted aqueous solutions. The vesicles showed very slow release rate of the encapsulated water-soluble dyes, indicating that they are stable and belong to the least permeable vesicles.

Abstract:Choline hydroxide (ChOH) is an effective ionic liquid (IL) that enables the straightforward construction of β-phosphonomalonates in basic medium. A variety of compounds, including aromatic/heterocyclic and aliphatic aldehydes ( 1a–p ) were reacted with dicyanomethane ( 2 ) and diethyl phosphonate ( 3 ) to produce the targeted products in good yields ( 4a–p ). The constructed compounds as well as the IL were characterized using spectral 1 H, 13 C, and 31 P NMR.

Abstract:A novel approach to synthesize pitavastatin calcium (1), an effective HMG-CoA reductase inhibitor, based on readily available and attractively functionalized (R)-3-chloro-1,2-propanediol is reported. Abstract:We report a facile synthetic strategy toward CH2-substituted phosphothreonine mimetics. Herein, inexpensive valine with a directing group was converted into homothreonine via palladium-catalyzed gamma-methyl C(sp(3))-H bond activation, followed by construction of a phosphorus-carbon bond via the well-developed Appel reaction and Michaelis-Becker reaction with a total yield of 30%. Furthermore, the derived mimetic was applied for solid-phase synthesis of two phosphopeptide inhibitors. This efficient synthesis provides a chance to prepare not only phosphopeptides but also phosphoproteins resistant to phosphatases.

Abstract:A simple and efficient synthesis of two new hydroximinosteroidal derivatives (11, 13) from cholesterol was described. The cytotoxicity of these compounds, their intermediates were evaluated on human cancer cell lines including Hep-G2, HeLa and T98G. Compound 13 presented a potent antiproliferative effect to T98G cancer cell at 2.9 mM of IC50. In addition, the antimicrobial assay showed that two new oximes 11 and 13 were active against E. coli and S. aureus strain (MIC :50 mg/mL).
