Abstract:This paper presents a novel method to fabricate a normally open micro pneumatic valve in rigid poly(methyl methacrylate) (PMMA) chips. The control and fluid substrates of the valve were prepared with PMMA sheets via hot embossing. After both control and fluid substrates were silanized with 3-aminopropyltriethoxysilane and treated with glow discharge plasma, each of them was irreversibly bonded to a plasma-treated PDMS membrane, forming a control half or a fluid half. Relying on the native adhesive force of the PDMS membrane, the control half and fluid half were then reversibly sealed to form a full microchip with the four-layer structure of PMMA-PDMA center dot center dot center dot PDMS-PMMA. The irreversible bonding between rigid PMMA substrate and elastic PDMS membrane in combination with the reversible sealing between the control half and fluid half not only eases the difficulties in chip bonding, but also allows the control half and fluid half to be replaced individually in routine analyses. The performance of the developed micro pneumatic valves was characterized, and potential applications of the valves in droplet generation and micro flow injection analysis were demonstrated.

Abstract:On-chip valves can simplify a microfluidic chip and make it easy to operate. However, most on-chip valves already reported still need complicated manufacture and sophisticated supporting devices. In this work, we present a straightforward on-chip valve, which can be serially connected, to form an on-chip pump. The liquid can horizontally flow one way by the regular deformations of flexure strips in the two valves at both sides of the chamber under pressure changes in microchannels generated by repeated vertical movements of linear actuators. The volume of this system including the chip and the supporting device is 0.65 cubic decimeters, which is much smaller than that of reported systems with a volume of at least 12 cubic decimeters, and the weight of this system is only 0.56 kg, making it possible for point-of-care testing. We carry out an immunoassay of folic acid on chip, and the results show satisfactory reproducibility with acceptable coefficients of variation. We determine 163 clinical human serum samples for folic acid. Furthermore, we detect transferrin, cobalamin and folic acid simultaneously on one chip with both sandwich and competitive binding immunoassay methods. We anticipate that this on-chip valve and pump can be applied in immunoassays and other biosensing applications.

Microfluidic Time-Delay Valve Mechanism on Paper-Based Devices for Automated Competitive ELISA

Abstract:Paper-based technologies have been drawing increasing attentions in the biosensor field due to their economical, ecofriendly, and easy-to-fabricate features. In this paper, we present a time-delay valve mechanism to automate a series of procedures for conducting competitive enzyme-linked immunosorbent assay (ELISA) on a paper-based device. The mechanism employs a controllable time-delay valve, which has surfactants to dissolve the hydrophobic barriers, in a fluid pathway. The valves can regulate the liquid and sequentially deliver the sample flow for automating ELISA procedures in microchannels. Competitive ELISA is achieved in a single step once the sample, or small molecule pesticide (e.g., Imidacloprid), is applied onto the paper-based device with a comparable sensitivity to plate-based competitive ELISA. The results further demonstrate the appositeness of using paper-based devices with the valve designs for on-the-go ELISA detection in agriculture and biomedical applications.

Abstract:Hydrogel-based microfluidic chips are more biologically relevant than conventional polydimethylsiloxane (PDMS) chips, but the inherent swelling of hydrogels leads to a decrease in mechanical performance and deformation of the as-prepared structure in their manufacture and application processing. Non-swelling hydrogel has, for the first time, been utilized to construct microfluidic chips in this study. It was fabricated by covalently cross-linking the biocompatible copolymer of di-acrylated Pluronic F127 (F127-DA). Thanks to their non-swelling property, the hydrogel-based microfluidic chips maintain their as-prepared mechanical strength and channel morphology when equilibrated in aqueous solution at 37 degrees C. Moreover, the microfluidic chips are autoclavable and show an appropriately slow degradation rate by remaining stable within 21 days of incubation. Based on these properties, a vessel-on-a-chip was established by seeding human umbilical vein endothelial cells (HUVECs) onto the microchannel surfaces inside the microfluidic chip. Under 6 days of perfusion culture with a physiologically relevant shear stress of 5 dyne per cm(2), the HUVECs in the chip show responsivity to fluid shear stress and express higher endothelial functions than the corresponding static culture. Therefore, non-swelling hydrogel-based microfluidic chips could potentially be applicable for cell/tissue-related applications, performing much better than conventional PDMS or existing hydrogel based microfluidic chips.

Abstract:A novel polymethylmethacrylate(PMMA)/polydimethylsiloxane(PDMS)hybrid polymer microfluidic chip is reported.The chip has 4-layer structure of PMMA-PDMS…PDMS-PMMA to integrate the pneumatic microvalves.The fluid PMMA substrate and control PMMA substrate are irreversibly bonded to an elastic PDMS membrane to respectively form the fluid half chip and control half chip.Relying on the adhering property of the PDMS membranes,the fluid and control halves are reversibly bonded to form a complete chip with micro valves.This method solves the bonding problems occurred in fabricating PMMA-PDMS-PMMA 3-layer microfluidic chip and the bonding process is simple and reliable.The control half and fluid half are allowed to be replaced individually,so the operation cost can be further reduced,which makes this microfluidic chip especially suitable for disposable use.The preliminary experiment results show that the microvalve has excellent switching performance and durability.

Abstract:Several microvalves utilizing stimuli-responsive hydrogel materials have been developed. The hydrogel components are fabricated inside microchannels using a liquid phase polymerization process. In-channel processing greatly simplifies device construction, assembly, and operation since the functional components are fabricated in situ and can perform both sensing and actuation functions. Two in situ photopolymerization techniques, “laminar stream mode” and “mask mode,” have been explored. Three two-dimensional (2-D) valves were fabricated and tested (response time, pressure drop, maximum differential pressure). In addition, a hydrogel/PDMS three-dimensional (3-D) hybrid valve that physically separates the sensing and regulated streams was demonstrated. Analytical modeling was performed on the 3-D valve. Hydrogel-based microvalves have a number of advantages over conventional microvalves, including relatively simple fabrication, no external power requirement, no integrated electronics, large displacement (185 $\mu$ m), and large force generation (22 mN).$\hfill{\hbox{[597]}}$

Abstract:In this paper we present a bulk micromachined check valve with very high frequency and extremely low leak rates. The valve is designed to have a hexagonal orifice, a hexagonal membrane flap and three flexible tethers. The three elbow-shaped flexible tethers are used to secure the membrane flap to the valve seat and to obtain large flap displacement in the forward flow direction. A silicon-on-insulator wafer and deep reactive ion etching technology are used to implement this microvalve. A very simple fabrication process has been developed, and only two photolithographic masks are required. Preliminary fluidics testing on a 1.44 mm size check valve was performed. A maximum flow rate (deionized water) of 35.6 ml min(-1) was obtained at a forward pressure of 65.5 kPa, and only negligible leakage rate was observed at a reverse pressure of up to 600 kPa. The frequency response of the valve in air was also measured and its first resonance frequency was found at 17.7 kHz.

Abstract:In recent years, microvalves have become increasingly significant in microfluidic structures. In microfluidic devices, these microvalves are crucial to controlling fluid flow. Due to their ability to precisely manipulate and adjust the fluid, these structures are more efficient in applications, particularly in medicine, drug delivery, protein analysis, microscale communication, and chemical processes. Utilizing a numerical model for the design and fabrication of a microvalve results in the development of a more optimized and efficient structure. Microvalves can be generally divided into active and passive categories. This research focuses on the design and fabrication of a pneumatic microvalve, which falls under the category of active microvalves. This structure has two distinct chambers that allow for microscale communication. The microvalve is constructed using two layers, where the upper layer controls the lower layer, which contains fluid flow through pneumatic pressure. The performance of the system was evaluated through experimental analysis. The microchannel passage remained obstructed when the flow rates were set to 0.2 ml/h and when maintaining a constant pump pressure of 15 psi. However, the fluid flow is connected even when the pressure is cut off. We believe this structure will impact diverse applications, including molecular communication.

Abstract:Abstract Microfluidics has enabled low volume biochemistry reactions to be carried out at the point-of-care. A key component in microfluidics is the microfluidic valve. Microfluidic valves are not only useful for directing flow at intersections but also allow mixtures/dilutions to be tuned real-time and even provide peristaltic pumping capabilities. In the transition from chip-in-a-lab to lab-on-a-chip, it is essential to ensure that microfluidic valves are designed to require less peripheral equipment and that they are transportable. In this paper, a thermally-actuated microfluidic valve is presented. The valve itself is fabricated with off-the-shelf components without the need for sophisticated cleanroom techniques. It is shown that multiple valves can be controlled and operated via a power supply and an Arduino microcontroller; an important step towards transportable microfluidic devices capable of carrying out analytical assays at the point-of-care. It is been calculated that a single actuator costs less than $1, this highlights the potential of the presented valve for scaling out. The valve operation is demonstrated by adjusting the ratio of a water/dye mixture in a continuous flow microfluidic chip with Y-junction channel geometry. The power required to operate one microfluidic valve has been characterised both theoretically and experimentally. Cyclical operation of the valve has been demonstrated for 65 h with 585 actuations. The presented valve is capable of actuating rectangular microfluidic channels of 500 μm × 50 μm with an expected temperature increase of up to 5 °C. The fastest actuation times achieved were 2 s for valve closing (heating) and 9 s for valve opening (cooling).

Fabrication of PMMA microfluidic chips using disposable agar hydrogel templates.

Abstract:A novel method based on disposable agar hydrogel temples have been developed for the fabrication of PMMA microfluidic chips. Molten agar hydrogel was sandwiched between a glass plate and a PMMA template bearing negative relief of microstructure. After cooling, the negative PMMA template could be easily separated from the solidified agar hydrogel and a layer of agar hydrogel temple bearing high-fidelity positive relief of the microstructure was left on the glass plate. Prepolymerized methyl methacrylate molding solution containing ultraviolet-initiator was subsequently sandwiched between the agar hydrogel template and a PMMA plate and was allowed to polymerize under ultraviolet light to fabricate the PMMA channel plate at room temperature. Complete microchips could be obtained by bonding the channel plates with covers. The prepared microfluidic microchips have been successfully employed in the electrophoresis separation and detection of several ions in connection with contactless conductivity detection.

Abstract:A novel method for fabricating nano- or submicro-fluidic PMMA chips using photoresist-free UV-lithography and UV-assisted low-temperature bonding was developed. The nano- or submicro-channels were fabricated by exposing the PMMA substrate to the UV-light through a mask for a certain time. The PMMA substrate with channels and another flat PMMA cover sheet were pretreated with the UV-light for 2 h before they were brought together in running water. The bonding was carried out under a pressure of (1.19 ± 0.12) × 105 Pa and at a temperature of 45°C for 35 min. The chips bonded in this way could bear a tensile of 6.71 ± 2.50 MPa, and the deformation of the bonded channel was about 13%. A hybrid micro- and nano-fluidic PMMA chip fabricated with the developed method was demonstrated for the test of the electrokinetically driven ion enrichment and ion depletion.
Design and fabrication of a microfluidic detection chip

Abstract:A new type of plastic microfluidic detection chip was developed for analyzing and testing samples. The size of the chip is 86 mm×60 mm×4.5 mm. Gas is used to drive the fluid. The chip was fabricated using precision machining method. A simple hydrophilic modification method was applied for microchannels. Experiment results prove that the hydrophilicity is markedly improved. The mechanism of the hydrophilicity improvement is also analyzed. The chip was bonded in room temperature with a dichloroethane-alcohol mixture mixed in a ratio of 1:1. The influence to the bonding result is analyzed for different mixture ratio, bonding time and bonding pressure. Several valves were integrated in the chip. The adhesion technology of the valve membrane was studied.
Preparation of thermoswitchable micro capillary force valves inside polydimethylsiloxane microfluidic channels

Abstract:This article presents a simple approach for preparation of thermoswitchable capillary force valves on full PDMS microfluidic chips. The highly hydrophobic PDMS surface was tailored to hydrophilic or hydrophilicity/hydrophobicity thermoswitchable by surface grafting of poly (acrylic acid) (PAA) or Poly (N-isopropylacrylamide) (PNIPAAm) via benzophenone initiated surface photopolymerization techniques. A thermoswitcable capillary force valve with a hydrophobicity/hydrophilicity switchable PNIPAAm patch in otherwise hydrophilic PAA grafted PDMS channel was designed, and a two-step region-selective grafting method was established to prepare the valve inside bonded PDMS microchannels. With a micro Peltier heating/cooling device used to regulate the valve temperature, aqueous solution was allowed to pass the valve at room temperature while the steam could be stopped at the edge of the PNIPAAm patch that was at 40°C. The capability of the developed thermoswitchable capillary force valve for manipulation of aqueous flow was demonstrated on full PDMS chips with a T-shaped micro channel network.
Fabrication of full glass chips with hybrid micro- and nanochannels and their application to protein concentration

Abstract:In this paper, we proposed an alternative method that is simple and cost-effective to fabricate full glass chips with hybrid micro- and nanochannels. Double-lithography and double wet-etching technique was used for the fabrication of hybrid micro and nanostructures. The microchannels were firstly constructed on a glass substrate by standard UV photolithography and wet etching technique. After washing away the rest photoresist and spinning a new layer of photoresist on the microchannel-structured glass substrate, nanochannels with the depth less than 100 nm were produced on the same substrate by second lithography and wet etching. Then the channels-structured glass substrate was sealed to a cover glass plate by using previously developed room temperature bonding technique. Without using clean room facilities and expensive instruments, micro- and nanofluidic chips with delicate structures were well fabricated. The good nature of those nanofluidic chips was demonstrated by the successful concentration of FITC-labeled bovine serum albumin via electrokinetic ion trapping.

Abstract:An array of thermo-actuated poly(N-isopropylacrylamide) (PNIPAAm) multivalves was designed and fabricated to perform volume-based sample injection for microflow injection analysis on a glass microfluidic chip. The PNIPAAm monolithic plug valves were prepared inside the vinylized glass channels by photopolymerization in water–ethanol (1:1) medium using 2-hydroxy-2-methyl propiophenone (Darocure-1173) as the initiator and a photo-mask for micropattern transferring. Experimental conditions for the photopolymerization were studied, and the thermo-responsive behavior of the synthesized monolithic plug valves was investigated. To perform active heating and cooling of the on-chip integrated thermo-actuated valves, micro-Peltier devices were used and operation times of 3-s for opening and 7-s for closing were obtained. In the close status, a 2-mm long monolithic plug valve could endure a pressure of no higher than 0.45MPa. The volume-based sample and reagent injector was composed of two groups of valves (total valve number of 5) and two loops. When the two groups of valves were alternatively opened and closed via thermo-actuation, the sampling loops were able to be switched between loading and injection position without any mechanical moving parts. Cooperating with syringe pumps, the microfluidic chip with the integrated sample injector has been demonstrated for microflow injection chemiluminescence detection of hydrogen peroxide. For a sampling volume of 6nL, linear response was observed over the H2O2 concentration range of 0–2mmolL−1, and a precision of 0.6% (RSD, n=11) was achieved for a standard H2O2 solution 2mmolL−1.
Fabrication method and performance of a light-responsive hydrogel microvalve in a microfluidic chip

Abstract:Microfluidic technology has potential advantages in the complex manipulation of microfluidics on small-sized chips. However, it is difficult to integrate microvalves with complex flow channel structures, and this has limited the miniaturization of microfluidic systems and their portable applications. Light-responsive hydrogel (LRH) materials can rapidly change their volume under laser irradiation and can be used to prepare flexible microvalves to realize the integrated control of microfluidics. A simple fabrication method for an LRH microvalve on a microfluidic chip is proposed. The microspheres, as control elements of the microvalve based on an LRH modified with Laponite RD nanoclay and ferriferous oxide (Fe3O4) nanoparticles, are prepared through a T-shaped flow channel. The microvalve is assembled on the microfluidic chip with a normally closed circulation channel. The open/close performance of the microvalve is represented by the color change of the photonic crystal material. The results show that the LRH microspheres shrink and the flow channel opens after laser irradiation for 2 s. After stopping the laser at 18 s, the valve core swells and the flow channel closes.
Weak solvent based chip lamination and characterization of on-chip valve and pump

Abstract:Using polystyrene as a fabrication material and pure acetonitrile as a bonding solvent, we have developed an innovative and inexpensive weak-solvent-based chip lamination process to produce highly functional, completely plastic, microfluidic chips with a 3-layer structure. This simple, scalable and rapid method allows active components, such as multiple valves and pumps, to be constructed on chip with a thin, deflectable film as the middle layer sandwiched between two polystyrene layers. Our irreversible bonding method achieves uniform lamination under mild conditions (35–45°C and 10–50 KPa) without damage to the underlying micro-features. The on-chip valve and pump structures have been systematically characterized and the pumping rate has been compared against theoretical rates predicted by mathematical modeling studies. A wide range of pumping rates (0.33–10 μL/s) can be achieved, with the integral pumps maintaining a constant pumping rate and depending on pumping frequency and pump diaphragm size. Valve leakage of less than 0.02 μL/min is noted under pressures of 41 kPa. Utilizing various configurations of on-chip valves and pumps, the fully automated flow control of an integrated chip for sample lysis, nucleic acid purification and PCR is demonstrated. The present technology and chip have been heavily evaluated internally and externally for rapid biomedical diagnosis of HPV, HIV, etc., and they are currently in the process of commercialization.

A valve-less microfluidic peristaltic pumping method.

Abstract:We demonstrate a valve-less microfluidic peristaltic pumping method which enables the delivery of continuous nanoliter-scale flow with high precision. The fluid is driven by squeezing the microchannels embedded in a poly(dimethylsiloxane) device with rolling cams or bearings. We achieve continuous and uniform flow with velocity range from 1 to 500 nl/s, with outflow volume error within 3 nl. The devices show enhanced backpressure resistance up to 340 kPa. This method also shows great flexibility. By altering the channels' layout, emulsions and plugs can be generated easily. These low-cost and easy-to-fabricate micro-pumps offer novel approaches for liquid actuation in various microfluidic applications. (c) 2015 AIP Publishing LLC.
Configurable 3D Printed Microfluidic Multiport Valves with Axial Compression

Abstract:In the last decade, the fabrication of microfluidic chips was revolutionized by 3D printing. It is not only used for rapid prototyping of molds, but also for manufacturing of complex chips and even integrated active parts like pumps and valves, which are essential for many microfluidic applications. The manufacturing of multiport injection valves is of special interest for analytical microfluidic systems, as they can reduce the injection to detection dead volume and thus enhance the resolution and decrease the detection limit. Designs reported so far use radial compression of rotor and stator. However, commercially available nonprinted valves usually feature axial compression, as this allows for adjustable compression and the possibility to integrate additional sealing elements. In this paper, we transfer the axial approach to 3D-printed valves and compare two different printing techniques, as well as six different sealing configurations. The tightness of the system is evaluated with optical examination, weighing, and flow measurements. The developed system shows similar performance to commercial or other 3D-printed valves with no measurable leakage for the static case and leakages below 0.5% in the dynamic case, can be turned automatically with a stepper motor, is easy to scale up, and is transferable to other printing methods and materials without design changes.

Abstract:This paper introduces the fabrication of microfluidic chips based on integrating the agarose structure and their application to bacterial culture.Two designs of microfluidic chips were devised.One was a chemostat in microfluidic chip,applied to the killing kinetics of cells.The microfluidic chip showed several advantages,measuring the data on-line,simplify the experimental manipulation and feasible output of tolerant cells.Therefore it holds potential for research of the killing kinetics of cells.The other had three different high micro-culture chambers and the bacterial population was measured by two ways-fluorescent intensity and OD value,by which more information about the bacterial population was acquired.The experiments indicated that the measuring tendency of bacterial growth by OD value was consistent with one by fluorescent intensity.There are other advantages in such microfluidic chip that it is easy to fabricate the microfluidic chips and the agarose confines bacteria in the chambers and cell nutrients are diffusible in agarose,which offer a good means to research the motile bacteria.
