Abstract:A new method is proposed to perform the multiscale simulation of nanoindentation with wedge indenter. We designed a potential field to model the wedge indenter based on the repulsive force-field approach which was used to model round indenters. A smooth curve representing the indenter tip is used to avoid the singularity of an ideal wedge indenter. Simulation with our approach correctly shows essential features of wedge nanoindentation. The load responses during retraction is confirmed by elastic model based on the Oliver-Pharr approach. Significant indentation size effect that the contact hardness decreases with the increase of contact depth is compared with existing models. The atomic configurations of the film shows that perfect dislocations nucleate under the indenter with Burges vectors perpendicular to the indentation direction. The strain distribution under the indenter shows that the length of plastic zone in the indentation direction is much larger than the contact width.

Abstract:Molecular dynamics simulations of contact mode form measurement process on a soft substrate with a special surface profile using a diamond AFM tip are preformed to investigate the contact behavior and the surface profile damages or distortions of the measured substrate. The process of contact mode imaging of the AFM can be treated as the process of nano-scratching. The simulation-predicted interaction force, including scratching force and normal force, characterizes the saw-tooth pattern, which is referred to as atomic stick-slips. The shape of measured substrate surface, especially the surface underneath the AFM tip, is distorted by the interaction force between the AFM tip and the substrate surface.

Abstract:We have attempted to apply the computer-based finite element analysis (FEA) method to accurately measure the mechanical properties (e.g., hardness and elasticity) of a soft material by an indentation test. First, an axisymmetric model has been developed using commercially available FEA code ANSYS. The FEA model consisted of a thin Al-film resting on Si-substrate. A spherical indenter has been used to indent the Al-film, which traveled a predefined depth during the loading and unloading cycles. First, numerical simulations were conducted to get the force vs. displacement plot, which was later used to determine the modulus of elasticity and hardness of Al-film. The effects of substrate modulus and indentation depth were thoroughly investigated to determine the modulus and hardness of Al-film. The effect of friction, considered at the interface of indenter and Al-film, was found to offer minimum impact for relatively small indentation depth. The induced force on the Al-film by the indenter has been found to be higher with increasing indentation depth when friction was considered. However the contact stiffness, represented by the slope of the unloading curve, has been found almost the same with and without considering friction. The variation of substrate modulus has been found to be ineffective to capture the Al-film modulus for relatively small indentation depth. However for higher indentation depth, the substrate modulus has been found to offer profound effect to capture the film modulus. The hardness of the Al-film has also been found to be relatively unaffected with variation of substrate modulus. However, the hardness of the Al-film has been found to be higher with friction for relatively high indentation depth. Results obtained from this preliminary research are important to continue further investigation and to characterize the mechanical properties of other soft-materials, e.g., biofilms to minimize its detrimental effects and utilize its favorable aspects in industrial and biomedical applications.

Abstract:The characterization of the mechanical properties of soft films is of great importance for their applications. In our previous research, we demonstrate the existence of a maximum load occurring during the indentation process of a perforated film by a spherical indenter. Based on this result, an approach to obtain the shear modulus of the film material using the method of finite element analysis has been proposed. However, our previous work does not consider the effect of friction between the film and the indenter, which has a significant influence on the value of the maximum load. Here, a theoretical model is presented which takes into account the role of friction. The reliability and accuracy of the theoretical model are validated by comparison with simulations and experimental results. In addition, the indenter eccentricity and round hole shape deviations which commonly occur in actual indentation tests, are investigated by combining indentation test measurements with finite element analysis. The performance of this method on porous films is also analyzed experimentally and numerically. The results reveal that this indentation method is still effective for porous films. This work provides a fundamental understanding of the mechanism of the indentation method and is expected to provide a new perspective for local characterization of films, even with multiple holes.
In Situ Nanoindentation Mechanical Property of Films by Atomic Force Microscope

Abstract:An in situ nanoindentation test system was established by combining atomic force microscope (AFM) and scan electron microscope (SEM). This system enabled the indentation experiment using the AFM cantilever with Cube corner diamond indenter, and acquired the curve of load versus displacement. The experimental process can be observed in real time by in situ SEM simultaneously. A mechanical property testing method, based on the AFM cantilever loading curve and the in situ SEM indentation of images analysis, was well developed. The hardness and elastic model of the plastic thin film were obtained by measuring the maximum loading during the indentation and the residual area of the indentation by measuring SEM image. This method was demonstrated by conducting the indentation experiment on the sliver film deposited on the Si substrate using magnetron sputtering. Furthermore, the experiment conducted by the Nanoindenter G200 was presented here for comparison. The results show that the in situ AFM indentation possesses higher resolution of loading and displacement, and can achieve the indentation experiment scaled from nN to mN of the applied load. The hardness of the plastic thin film can be obtained by measuring the residual area of indentation. The pile-up effect of the soft film deposited on the hard substrate, which exists in the Oliver-pharr method, can be reduced by SEM image analyses of residual area and the calculation has a higher testing accuracy and reliability as well.
On the Determination of Elastic Moduli of Cells by AFM Based Indentation

Abstract:The atomic force microscopy (AFM) has been widely used to measure the mechanical properties of biological cells through indentations. In most of existing studies, the cell is supposed to be linear elastic within the small strain regime when analyzing the AFM indentation data. However, in experimental situations, the roles of large deformation and surface tension of cells should be taken into consideration. Here, we use the neo-Hookean model to describe the hyperelastic behavior of cells and investigate the influence of surface tension through finite element simulations. At large deformation, a correction factor, depending on the geometric ratio of indenter radius to cell radius, is introduced to modify the force-indent depth relation of classical Hertzian model. Moreover, when the indent depth is comparable with an intrinsic length defined as the ratio of surface tension to elastic modulus, the surface tension evidently affects the indentation response, indicating an overestimation of elastic modulus by the Hertzian model. The dimensionless-analysis-based theoretical predictions, which include both large deformation and surface tension, are in good agreement with our finite element simulation data. This study provides a novel method to more accurately measure the mechanical properties of biological cells and soft materials in AFM indentation experiments.
AFM Indentation and Material Property Identification of Soft Hydrogels

Abstract:There is growing evidence of the importance of mechanical deformations on various facets of cell functioning. This asks for a proper understanding of the cell’s characteristics as a mechanical system in different physiological and mechanical loading conditions. Many researchers use atomic force microscopy (AFM) indentation and the Hertz contact model for elastic material property identification under shallow indentation. For larger indentations, many of the Hertz assumptions are not inherently satisfied and the Hertz model is not directly useful for characterizing nonlinear elastic or inelastic material properties. We have used exponential hyperelastic material in FE simulations of the AFM indentation tests. A parameter identification approach is developed for hyperelastic material property determination from the simulated data. We collected AFM indentation data on agarose gel and developed a simple algorithm for contact point detection. The contact point correction improves the prediction of elastic modulus over the case of visual contact point identification. The modulus of 1% agarose gel was found to be about 15 kPa using the proposed correction, with mild but non-trival hardening with deeper indentation. The experimental data is compared with the results from the FE simulations and shows that over the hardening portion of the indentation response, our proposed parameter identification approach successfully captures the experimental data.

Abstract:The finite element method (FEM) for elastoplastic large strain was employed to study the indentation experiment of Al film on Si substrate by a rigid spherical indenter. Two relationships between the mechanical properties of the film and the specific values of the load–displacement curve have been obtained. Using a hybrid method that involves the combination of these two relationships with nanoindentation test, the hardening index and yield strength of the metal film can be determined.

Abstract:We hypothesize that both compression and elongation stress-strain data should be considered for modeling and simulation of soft tissue indentation. Uniaxial stress-strain data were obtained from in vitro loading experiments of porcine liver tissue. An axisymmetric finite element model was used to simulate liver tissue indentation with tissue material represented by hyperelastic models. The material parameters were derived from uniaxial stress-strain data of compressions, elongations, and combined compression and elongation of porcine liver samples. in vitro indentation tests were used to validate the finite element simulation. Stress-strain data from the simulation with material parameters derived from the combined compression and elongation data match the experimental data best. This is due to its better ability in modeling 3D deformation since the behavior of biological soft tissue under indentation is affected by both its compressive and tensile characteristics. The combined logarithmic and polynomial model is somewhat better than the 5-constant Mooney-Rivlin model as the constitutive model for this indentation simulation.
Spherical indentation on biological films with surface energy

Abstract:Micro- and nano-indentations have been widely used to measure the mechanical properties of biological cells and tissues, but the direct application of the classical Hertzian contact model would lead to the overestimation of elastic modulus due to the influence of finite thickness and surface energy. In this work, we analyze spherical indentation of biological films, considering both large deformation and surface energy. The hyperelastic behavior of biological films is characterized by the neo-Hookean model, and the influence of surface energy is addressed through finite element simulation. Based on dimensional analysis, the explicit expressions of load-depth relation accounting for film thickness, large deformation and surface energy are achieved for bonded or non-bonded films. Under a specific load, the consideration of large deformation increases the indent depth, while the finite thickness of films tends to decrease the indent depth, compared to the linear elastic Hertzian solution. More importantly, surface energy evidently alters the load-depth relation for micro-/nano-indentations, which reduces the indent depth and makes the films seemingly stiffer. These results provide a fundamental relationship to accurately extract the mechanical properties of biological films from indentation tests.

Abstract:As critical components or platforms, stiff film-compliant substrate structures are widely used in micro-electro-mechanical systems (MEMS) and other devices, in which their in-dentation behavior plays an important role. In this work, a multi-scale indentation model (MSI) for stiff film-compliant substrate structures is established based on the strain gradi-ent theory and the surface elastic model. With this model, the indentation size effect of the structure is investigated from three aspects: the load-depth relationship, the distribu-tion of bending moments in the film, and the surface morphology. The results show that the indentation size effect is mainly caused by the characteristic length and the surface residual stress of the film, and it is sensitive to the modulus ratio of the film to the sub-strate but has little relation with Poisson's ratio and the contact radius. Based on the above analysis, two dimensionless numbers are proposed to evaluate the indentation size effect of stiff film-compliant substrate structures. Besides, the predicted bending stiffness by the theoretical model is consistent with the experimental result, which verifies the effective-ness of the MSI model. Our research sheds light on the understanding of the indentation size effect of stiff film-compliant substrate structures and provides theoretical guidance for the design of related devices. Indentation Depth-Dependent Hardness of Metal-Organic Framework Crystals: the Effect of Local Amorphization Induced by Indentation

Abstract:The hardness of metal-organic frameworks (MOFs) is an important mechanical property metric measuring their resistance to the permanent plastic deformation. The hardness of most MOFs measured from nanoindentation experiments usually exhibits the similar unique indentation depth dependence feature, the mechanism of which still remains unclear. In order to explain the effect of the indentation depth on the hardness of MOFs, we conducted nanoindentation simulations on HKUST-1 by using reactive molecular dynamics simulations. Our simulations reveal that the HKUST-1 material near the indenter can transform from the parent crystalline phase to a new amorphous phase due to the high pressure generated, while its counterpart far from the indenter remains in the crystalline phase. By considering the crystalline-amorphous interface in the energy analysis of MOFs, we derived an analytical expression of the hardness at different indentation depths. It is found that the interface effect can greatly increase the hardness of MOFs, as observed in nanoindentation simulations. Moreover, the proposed analytical expression can well explain the indentation depth-dependent hardness of many MOF crystals measured in nanoindentation experiments. Overall, this work can provide a better understanding of the indentation depth dependence of the hardness of MOFs. Based on reactive molecular dynamics simulations, a local crystalline-to-amorphous phase transition occurring in HKUST-1 crystals under indentation is reported. By considering the effect of two-phase interface, an analytical expression of hardness is derived that explains the size dependence of hardness of many metal-organic framework crystals measured in experiments.+image
Surface Effects on the Spherical Indentation of Biological Film/substrate Structures

Abstract:Micro-/nano-indentation has been the most popular technique to extract the mechanical characteristics of biological cells and tissues. However, due to surface effects and the existence of substrates, conventional contact models are unable to determine the accurate elastic modulus of biological samples by analyzing the measured load-indent depth data. In this study, the spherical indentation of the film/substrate structure considering the surface energy and large deformations is investigated. The hyperelasticity of biological films and substrates is considered through neo-Hookean constitutive model, and the surface effect is incorporated using the finite element method. The explicit formulas for the relations between load and indent depth are presented for films with two orders of magnitude modulus mismatch to the substrate. It is found that the modulus mismatch between film and underlying substrate would lead to an overestimation of modulus for the film on a stiffer substrate, but an underestimated modulus for that on a softer substrate if the conventional Hertzian theory is directly adopted in the analysis. Moreover, for indentation at micro-/nano-scale, the surface energy would pronouncedly reduce the indent depth under a given load and lead to a seemingly stiffer film. Our results provide the explicit expressions to accurately predict the spherical indentation response of biological film/substrate structures.

Abstract:A comprehensive study on the spherical indentation of hyperelastic soft materials is carried out through combined theoretical, computational, and experimental efforts. Four widely used hyperelastic constitutive models are studied, including neo-Hookean, Mooney–Rivlin, Fung, and Arruda–Boyce models. Through dimensional analysis and finite element simulations, we establish the explicit relations between the indentation loads at given indentation depths and the constitutive parameters of materials. Based on the obtained results, the applicability of Hertzian solution to the measurement of the initial shear modulus of hyperelastic materials is examined. Furthermore, from the viewpoint of inverse problems, the possibility to measure some other properties of a hyperelastic material using spherical indentation tests, e.g., locking stretch, is addressed by considering the existence, uniqueness, and stability of the solution. Experiments have been performed on polydimethylsiloxane to validate the conclusions drawn from our theoretical analysis. The results reported in this study should help identify the extent to which the mechanical properties of hyperelastic materials could be measured from spherical indentation tests.

Abstract:Abstract Modern developments of biomedical applications demand a better understanding of mechanical behaviour of soft biological tissues. As human soft tissues demonstrate a significant structural and functional diversity, characterisation of their mechanical behaviour still remains a challenge. Limitations related with implementation of mechanical experiments on human participants lead to a use of finite-element models for analysis of mechanical responses of soft tissues to different loads. This study focuses on parameters of numerical simulation considered for modelling of indentation of a human lower limb. Assessment of the effect of boundary conditions on the model size shows that at a ratio of its length to the tissue’s thickness of 1.7 for the 3D model this effect vanishes. The numerical results obtained with models employing various sets of mechanical parameters of the first-order Ogden scheme were compared with original experimental data. Furthermore, high sensitivity of the resulting reaction forces to the indenting direction is demonstrated for cases of both linear and angular misalignments of the indenter. Finally, the effect of changes in material parameters and their domain on their contribution to the reaction forces is discussed with the aim to improve our understanding of mechanical behaviour of soft tissues based on numerical methods. The undertaken research with its results on minimal requirements for finite-element models of indentation of soft tissues can support inverse analysis of their mechanical properties and underpin orthopaedic and medical procedures.

Abstract:Hertzian mechanics is the most frequently used theory for data processing in Atomic Force Microscopy (AFM) indentation experiments on soft biological samples, due to its simplicity and significant scientific results previously published. For instance, using the Hertz model, it has been proven that there are significant differences in the mechanical properties of normal and cancerous tissues and that cancer cells’ invasive properties are correlated with their nanomechanical properties. However, many scientists are skeptical regarding the applicability of the Hertz theory to biological materials, as they are highly heterogeneous. The main critical question to be addressed is “what do we calculate” when fitting the force-indentation data to Hertz equations. Previous studies have shown that when using cylindrical, parabolic, or conical indenters, the fitting parameter is the average Young’s modulus. In this paper, it is demonstrated that it is also valid to fit equations derived from Hertzian mechanics to force-indentation data when testing soft, heterogeneous samples for any indenter geometry. The fitting factor calculated through this approach always represents the average Young’s modulus for a specific indentation depth. Therefore, Hertzian mechanics can be extended to soft heterogeneous materials, regardless of the indenter’s shape.

Abstract:Atomic Force Microscopy (AFM) is a leading nanoscale technique known for its significant advantages in the analysis of soft materials and biological samples. Traditional AFM data analysis is often based on the Hertz model, which assumes perpendicular indentation of a planar sample. However, this assumption is not always valid due to the varying geometries of soft materials, whether natural, synthetic or biological. In this study, we present a new theoretical model that incorporates correction coefficients into Hertz's model to account for cone-like and spherical probes, and to consider local tilt at the probe-sample interface. We validate our model using finite element analysis (FEA) simulations and experimental AFM measurements on tilted polyacrylamide gels. Our results highlight the need to include local tilt at the probe-sample contact to ensure accurate AFM measurements. This represents a step forward in our understanding of the elastic properties at the surface of soft materials in the broadest sense.

Abstract:The indentation process of aluminum film/silicon substrate combination by a rigid spherical indenter has been simulated using finite element method (FEM). It is demonstrated that both the yield strength and hardening index of the film affect the load-displacement curve. An empirical relationship among the strength parameters and hardness of the film has been established from the FEM results. Utilizing the relationship and the hardness measured by nanoindentation test for two different indentation depths, the yield strength and plastic hardening index of the film can be estimated.

Abstract:Atomic force microscopy (AFM) provides a three-dimensional topographic representation of a sample surface, at nanometre resolution. Computational simulations can aid the interpretation of such representations, but have mostly been limited to cases where both the AFM probe and the sample are hard and not compressible. In many applications, however, the sample is soft and therefore deformed due to the force exerted by the AFM tip. Here we use finite element modelling (FEM) to study how the measured AFM topography relates to the surface structures of soft and compressible materials. Consistent with previous analytical studies, the measured elastic modulus in AFM is generally found to deviate from the elastic modulus of the sample material. By the analysis of simple surface geometries, the FEM modelling shows how measured mechanical and topographic features in AFM images depend on a combination of tip-sample geometry and indentation of the tip into the sample. Importantly for the interpretation of AFM data, nanoparticles may appear larger or smaller by a factor of two depending on tip size and indentation force; and a higher spatial resolution in AFM images does not necessarily coincide with a more accurate representation of the sample surface. These observations on simple surface geometries also extend to molecular-resolution AFM, as illustrated by comparing FEM results with experimental data acquired on DNA. Taken together, the FEM results provide a framework that aids the interpretation of surface topography and local mechanics as measured by AFM.

Abstract:Mechanical and toughness properties of the stiff layers on polymeric substrate are studied by AFM indentation and FEM modeling. Analysis of indentation data of heterogeneous material, in particular, layer on an elastic substrate requires information about the contact area that is essential for calculating mechanical properties. The actual shape of the AFM‐tip is not described by simple body of revolution. In this work, the indentation of a stiff layer on a hyperelastic substrate by a truncated conical tip is studied using finite element methods. The size of the tip, elastic modulus, and thickness of the layer are varied. The obtained model dependences of loading and contact area versus the depth of indentation are used in analysis of the experimental data of AFM indentation of stiff inhomogeneous nanolayers of polyurethane surface. Thickness and elastic modulus of the layers, fracture properties of the surface are studied. The obtained results can find application in the study of mechanical properties and fracture toughness of thin flexible films on an elastic substrate.
