Abstract:Aromatic aldehydes are very important organic intermediates,which are widely used in pharmaceuticals,pesticides and other areas.In this paper,an improved synthetic method of aromatic aldehydes containing electrowithdrawing groups in ortho or para(or both) position of the aromatic ring was introduced.4-choloro-2-nitrotoluene,4-nitrotoluene and 4-cyanotoluene were used as raw materials,respectively,via enamination with N,N-dimethylformamide dimethyl acetal(DMFDMA) in the present of N,N-dimethylformamide(DMF) as solvent and under reflux,the corresponding styrylamine compounds were produced;then the intermediates were further oxidized by H2O2 under different reaction conditions.The effects of solvent species,reaction temperature and the used amount of H2O2 on the product yield were studied,and the optimal reaction conditions were determined.Under those optimal conditions,the best yields of different aldehydes are 75.6%(4-chloro-2-nitrobenzaldehyde),58.3%(4-nitrobenzaldehyde) and 68.2%(4-cyanobenzaldehyde),respectively with the advantages of simple process,low cost and being environment-friendly..The main intermediates and objective products were confirmed by HPLC and 1H-NMR.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:Comprehensive Summary The asymmetric reductive arylation and alkenylation of benzylic chloride under photoredox/nickel dual catalysis using chiral biimidazoline (BiIm) ligand is reported to access 1,1-diaryl alkanes and aryl allylic compounds with good yield as well as stereo- and enantioselectivities. This protocol uses more commercially available and less expensive C(sp(2))-Br as the electrophile coupling partner. A primary result using alkenyl chloride and alkyl chloride is also reported. Various functional groups are tolerated and the applications of this method are investigated by late-stage functionalization and gram-scale reaction.

Abstract:During the past decades, N-heterocyclic carbene (NHC)-catalyzed reactions have emerged as a versatile tool in synthetic chemistry. In particular, NHC-catalyzed cross-coupling reaction has been significantly developed in many respects, including new reaction development and mechanistic investigation. This concept article presents recent advances towards direct cross-coupling reactions of aldehydes with alkyl halides enabled by NHC organocatalysis. N-heterocyclic carbene-catalyzed cross-coupling reaction has been recognized as an enabling tool in the synthesis of complex ketones in recent years. Abstract:Abstract Methods that enable site selective acylation of sp 3 C-H bonds in complex organic molecules are not well explored, particularly if compared with analogous transformations of aromatic and vinylic sp 2 C-H bonds. We report herein a direct acylation of benzylic C-H bonds by merging N-heterocyclic carbene (NHC) and photoredox catalysis. The method allows the preparation of a diverse range of benzylic ketones with good functional group tolerance under mild conditions. The reaction can be used to install acyl groups on highly functionalized natural product derived compounds and the C-H functionalization works with excellent site selectivity. The combination of NHC and photoredox catalysis offers options in preparing benzyl aryl ketones.
