Abstract:Palladium‐catalyzed desulfitative Hiyama cross‐coupling reactions of various arylsulfonyl chlorides with aryltriethoxysilane have been achieved in good yields. The reported cross‐coupling reactions are tolerant to the common functional groups regardless of electron‐withdrawing or electron‐donating, making these transformations attractive alternatives to the traditional cross‐coupling approaches. Abstract:The present methods deal with the facile synthesis of unsymmetrical thiosulfonates by the reactions of N‐hydroxy aryl sulfonamides and disulfides using the iodine and TBHP system. The oxidative coupling reactions afford a variety of thiosulfonates in good to high yields. Moreover, the large‐scale synthesis and practical application of the protocol have been demonstrated. An efficient and novel method has been developed for the synthesis of structurally diverse unsymmetrical thiosulfonates via the oxidative coupling reactions of N‐hydroxy aryl sulfonamides and disulfides. The iodine and TBHP system enables the transformations to afford a variety of thiosulfonates with good to high yields. Furthermore, the large‐scale synthesis and practical application to access sulfonamide have been investigated. Notably, the present protocol offers easily accessible materials, a wide range of substrates, radical pathways, and metal‐free conditions.

Abstract:A concise and efficient cross-coupling synthetic strategy has been developed to construct 2-aryl-4H-thiochromen-4-one derivatives from 2-sulfinyl-thiochromones and arylboronic acids. This reaction proceeds via a catalyst system of Lewis acid and palladium(II) combined with XPhos as an optimal ligand in moderate to good yields. Besides, this flexible methodology provides a wide scope for the synthesis of different functionally substituted thiochromone scaffolds and can be further exploited to construct diverse thioflavone libraries for pharmaceutical research.

Abstract:An electrochemical thioetherification of NH-sulfoximines with disulfides is reported. The use of electrochemistry to promote these oxidative coupling reactions not only obviates the requirement for external oxidants, bases, and metal catalysts but also ensures high environmental friendliness and selectivity to afford N -sulfenyl-sulfoximines. Good functional group tolerance, and readily available substrates offer this strategy incomparable advantages in preparing a variety of valuable N -sulfenyl-sulfoximines. Meanwhile, this reaction features simple operation, mild reaction conditions, and convenience of amplification, and has great potential in the subsequent modifications of pharmaceutical N -(2-thiophenyl)benzamide derivatives. Furthermore, the cytotoxic effects of the expected products toward different tumor cells were investigated in vitro and compared with 5-fluorouracil ( 5-Fu ), the most widely used chemotherapeutic drug against malignant tumors.

Abstract:Sulfones are common, readily available reagents that have recently attracted attention as versatile reagents for cross-coupling reactions. This Review summarizes advances in desulfonylative transformations of sulfones affected by molecular catalysis, including transition-metal catalysts, photocatalysts, and organocatalysts. In addition to catalyst choice, the design of the sulfonyl group is a critical factor to control reactivity. The concepts presented herein will provide new strategies to construct diverse molecules with high efficiency and modularity.

Abstract:Ortho-chelation assistance was proved to be crucial for the success of the desulfitative Sonogashira cross-coupling reaction of thioamide-type pyridine derivatives and alkynes. Corresponding alkynylated products were obtained with moderate to excellent yields. Thanks to this reaction, furo[3,2-b]pyridine and 1H-pyrrolo[3,2-b]pyridine derivatives were synthesized through a one-pot cross-coupling reaction/heteroannulation sequence.

Abstract:An expedient and practical route for realizing a novel, substrate-controlled diastereoselective switch strategy has been successfully developed. The [4+1] annulation of thioamides with sulfur ylides proceeded smoothly via a Michael addition triggered sequential process, and trans-dihydrothieno[3,4-c]coumarins were obtained because of steric hindrance effects between the benzoyl group and the unit of 2H-chromen-2-one as shown in the Newman projection. Nucleophilic substitution was observed as the key step when the sulfur ylides were replaced with acetophenones in the [4+1] annulation, and a double pi,pi-stacking donor-acceptor interaction stabilized the transition state that led to the cis-dihydrothieno[3,4-c]coumarins. In addition, a facile chemoselective reaction was also investigated, providing a series of thieno[3,4-c]coumarins.

Abstract:A highly efficient and convenient protocol was developed to access 2-amino-4H-benzothiopyran-4-ones through a process of conjugated addition-elimination. The sulfinyl group was proved to be the optimum leaving group by thorough investigations on the elimination of sulfide, sulfinyl, and sulfonyl groups at the 2-position of benzothiopyranone. Most 2-aminobenzothiopyranones were obtained in good to excellent yields under refluxing in isopropanol within 36 h. This method is base-free and the substrate scope in terms of electronic properties of the substituents of the benzothiopyranone is broad. The ten grams scale-up synthesis of the representative compounds 4a and 4d was implemented to show the practical application of this reaction, which afforded the corresponding compounds in good yields and excellent chemical purity without requiring column chromatographical purification.

Abstract:As "chemical chameleons," organosulfones have been widely applied in various desulfonylative functionalization reactions. However, the desulfonylative functionalization of (hetero)arylsulfones through the cleavage of inert C(sp2)–SO2 bonds remains a challenging and underexplored task. Over the past twenty years, the use of (hetero)arylsulfones as arylation reagents has gradually gained attention in diverse cross-coupling reactions under specific catalytic conditions, especially in transition metal-catalysis and photocatalysis chemistry. In this review, we discuss the representative accomplishments and mechanistic insights achieved in desulfonylative reactions of inactive C(sp2)–SO2 bonds in (hetero)arylsulfones, including: (i) transition-metal-catalyzed desulfonylative cross-coupling reactions and (ii) photo-/electrocatalytic radical desulfonylative coupling reactions. We anticipate that this review will provide an overall perspective in this area to a general audience of researchers and stimulate further innovative strategies for desulfonylative functionalization of inert arylsulfones.

Abstract:Here we present a systematic analysis of the synthesis of unsymmetrical disulphides employing 6‐thioguanosine and its mono‐ and dinucleotides by three of the most common strategies: a nucleophilic substitution of the thiol, a thiol exchange reaction with another disulphide and oxidative dehydrogenation coupling reactions of S‐H bonds. The exchange reaction was found to be the most efficient approach and could be used not only for nucleosides but also for nucleotides.

Abstract:The potentially bioactive 2-aryl-thiazolo[4,5-b]pyridines were synthesized via palladium-catalyzed desulfitative cross-coupling reaction between multisubstituted thiazolo[4,5-b]pyridine thioethers and boronic acids. Yields of [2_TD$IF]48%–94% were obtained with copper(I) thiophene-2-carboxylate in the system.

Abstract:An efficient, diversity-orientated synthesis of 5-sulfone-substituted uracils was established. The use of protecting groups to synthesize sulfones from N-heterocycles was avoided. Various heterocycles were synthesized for the first time from favorable, easily accessible starting materials. Diversity-orientated syntheses are important for the medicinal chemistry of virostatics and chemotherapeutics. This approach provides a broad substrate tolerance and excellent yields of up to 98%.
