Abstract:We report a stereoretentive cross-coupling reaction of configurationally stable nucleophiles with disulfide and N-sulfenylsuccinimide donors promoted by Cu(I). We demonstrate the utility of this method in the synthesis of thioglycosides derived from simple alkyl and aryl thiols, thioglycosides, and in the glycodiversification of cysteine residues in peptides. These reactions operate well with carbohydrate substrates containing common protective groups and reagents with free hydroxyl and secondary amide functionalities under standardized conditions. Competition experiments in combination with computational DFT studies established that the putative anomeric intermediate is an organocopper species that is configurationally stable and resistant to epimerization due to its short lifetime. The subsequent reductive elimination from the Cu(III) intermediate is rapid and stereoretentive. Taken together, the glycosyl cross-coupling is ideally suited for late stage glycodiversification and bioconjugation under highly controlled installation of the aliphatic carbon-sulfur bonds.

Abstract:Palladium‐catalyzed desulfitative Hiyama cross‐coupling reactions of various arylsulfonyl chlorides with aryltriethoxysilane have been achieved in good yields. The reported cross‐coupling reactions are tolerant to the common functional groups regardless of electron‐withdrawing or electron‐donating, making these transformations attractive alternatives to the traditional cross‐coupling approaches. Abstract:Ortho-chelation assistance was proved to be crucial for the success of the desulfitative Sonogashira cross-coupling reaction of thioamide-type pyridine derivatives and alkynes. Corresponding alkynylated products were obtained with moderate to excellent yields. Thanks to this reaction, furo[3,2-b]pyridine and 1H-pyrrolo[3,2-b]pyridine derivatives were synthesized through a one-pot cross-coupling reaction/heteroannulation sequence.

Abstract:A concise and efficient cross-coupling synthetic strategy has been developed to construct 2-aryl-4H-thiochromen-4-one derivatives from 2-sulfinyl-thiochromones and arylboronic acids. This reaction proceeds via a catalyst system of Lewis acid and palladium(II) combined with XPhos as an optimal ligand in moderate to good yields. Besides, this flexible methodology provides a wide scope for the synthesis of different functionally substituted thiochromone scaffolds and can be further exploited to construct diverse thioflavone libraries for pharmaceutical research.

Abstract:The present methods deal with the facile synthesis of unsymmetrical thiosulfonates by the reactions of N‐hydroxy aryl sulfonamides and disulfides using the iodine and TBHP system. The oxidative coupling reactions afford a variety of thiosulfonates in good to high yields. Moreover, the large‐scale synthesis and practical application of the protocol have been demonstrated. An efficient and novel method has been developed for the synthesis of structurally diverse unsymmetrical thiosulfonates via the oxidative coupling reactions of N‐hydroxy aryl sulfonamides and disulfides. The iodine and TBHP system enables the transformations to afford a variety of thiosulfonates with good to high yields. Furthermore, the large‐scale synthesis and practical application to access sulfonamide have been investigated. Notably, the present protocol offers easily accessible materials, a wide range of substrates, radical pathways, and metal‐free conditions.

Abstract:A protocol for the copper-catalyzed C–S bond formation between aryl, alkyl, or heteroaryl thiols and aryl or heteroaryl halides is reported. The reaction is catalyzed by Cu2O which shows the highest catalytic activity with ethyl 2-oxocyclohexanecarboxylate ligand in DMSO at 80°C. The corresponding coupling products are obtained with good to excellent yields under relatively mild reaction conditions with good chemoselectivity and functional group tolerance.

Abstract:A palladium catalyzed multicomponent cross-electrophile coupling involving sulfur dioxide under a metal-reductant free condition is presented here. Both aryl-alkyl and alkyl-alkyl sulfones could be successfully achieved. This protocol features mild reaction conditions, wide substrate scope, good functional group tolerance and excellent regioselectivity. Preliminary mechanistic studies reveal that two Pd catalytic cycles are involved and SO2 exclusively coupled with aryl iodides rather than allylic acetates, which are consistent with the DFT studies.

Abstract:A highly efficient and practical procedure for palladium-catalyzed direct thioetherification of quinolone derivatives with diaryl disulfides through decarboxylative C-S coupling has been established. The reaction could proceed smoothly under air in the presence of Pd(OAc)2 and Ag2 CO3 in DMSO. This protocol provides an appealing alternative to existing approaches to construct aryl sulfides of quinolone derivatives, which may be used as key intermediates in the synthesis of drug candidates.

Abstract:Abstract: A highly efficient and simple cross-coupling reaction of N-aryl aluminum reagents with Naryl thiosuccinimides for the synthesis of diarylsulide derivatives using CuCl (20 mol%) as a catalyst is reported. Under the optimum reaction conditions, the coupling reaction between aryl aluminum reagents with different substituents and N-aryl thiosuccinimides with different substituents can be carried out smoothly with 18-94% isolated yields of diarylsulides. The method avoids the use of expensive transition metals, such as Pd, Ir, or Rh, phosphine ligands, and has the advantages of simple operation and high reaction efficiency. The structures of all the target compounds are confirmed by 1 HNMR and 13CNMR.

Notably, the new approach employed active thioureas as coupling partners in the presence of arylsilanes to give amides in good yield. Abstract:[GRAPHICS] A novel thiourea-based C-2-symmetric ligand was synthesized, and its application in the palladium-catalyzed Heck and Suzuki coupling reactions of arenediazonium salts was evaluated, The reactions, which were performed at room temperature, without added base, and under aerobic conditions, produced product in 4 h with good yield. The corresponding arenediazonium salts were easily generated in one step from anilines.

Abstract:Thiuram disulfides undergo a Cu(I)-catalyzed C–S cross-coupling with aryl iodides through Cu(OAc) 2 ·H 2 O-assisted desulfurization to produce the S -thiocarbamate ester compounds efficiently. Various aryl iodides containing diverse substituents underwent a smooth reaction with a series of cyclic and acyclic secondary amine-based thiuram disulfides under an open-air atmosphere. A probable mechanistic pathway has been suggested based on control experiments and reports from the literature.
