Abstract:A highly enantioselective Michael addition of 3-substituted benzofuran-2(3H)-ones to chalcones catalyzed by a chiral bifunctional thiourea was developed. Several chiral 3,3'-substituted benzofuran-2(3H)-ones derivatives, bearing adjacent quaternary-tertiary stereocenters, were efficiently synthesized with excellent enantioselectivities.

Abstract:A chiral phosphoric acid‐catalyzed asymmetric 1,4‐addition of benzofuran‐derived azadienes with 3‐substituted indoles has been established, which offered enantioenriched tri(hetero)arylmethane products in generally good yields (up to 98%) and high enantioselectivities (up to 99:1 er). This reaction has not only realized the application of chiral phosphoric acid as a competent catalyst in the asymmetric transformations of benzofuran‐derived azadienes, but also has accomplished the task of developing chiral Brønsted acid‐catalyzed asymmetric 1,4‐additions of benzofuran‐derived azadienes, which will enrich the research contents of chiral phosphoric acid catalysis and the chemistry of benzofuran‐derived azadienes.

Abstract:The derivatives of 1-benzoyl-2-arylindolizine were prepared in moderate yields of 41%-78% by CrO3/Et3N promoted 1,3-dipolar cycloaddition of pyridinium N-ylides and chalcones. Under the same conditions, CrO3/Et3N promoted 1,3-dipolar cycloaddition of isoquinolinium N-ylides and chalcones provided the corresponding 1-benzoyl-2-arylpyffolo[2,1-a]isoquinolines in 45%-61% yields.

Abstract:Rocaglaol, a representative flavagline, has attracted significant attention because of its unique chemical structure and biological activities. This paper reports a mild and scalable copper-catalyzed enantioselective conjugate addition of benzofuran-3(2H)-one to α,β-unsaturated thioamides. This method allows for the concise synthesis of all possible stereoisomers of a key intermediate of rocaglaol and its derivatives in a highly diastereo- and enantioselective manner using different chiral phosphine ligands. Theoretical insights into the reaction mechanism and the origin of ligand-dependent diastereodivergence were obtained using density functional theory calculations.

Abstract:Herein, we report an example of a stereoselective γ-addition reaction of trifluoromethyl ketimines to 1-alkynyl ketones mediated by an isothiourea, BTM, under mild conditions, which afforded tetrasubstituted allenes with central chiralities in high yields (up to 94% yield), good enantioselectivities (up to 91% ee), and excellent diastereoselectivities (all >20 : 1 dr). In addition, the BTM-catalyzed γ-addition reaction was successfully applied to the gram-scale reaction, and an unexpected benzopyrrolothiazine derivative was successfully converted, albeit racemic.

Abstract:The trifluoromethylthio group is significant in drug design for its high stability, electronegativity, and lipophilicity. Herein, a series of indenone structures containing the SCF3 group and an adjacent chiral quaternary stereocenter were synthesized by an asymmetric Michael addition reaction using an ion pair catalyst derived from amino acids. The desired products were obtained in excellent yields (up to 98%) and good to high stereoselectivities (up to 97% ee and >20:1 dr).
