Abstract:A mild and efficient method for the synthesis of 2,5-disubstituted chalcogenophenes is reported. This method involves two steps of reactions via one-pot approach by employing commercially available and cost-effective alkynes and element chalcogen. Further, by using LiNH2 as the promoter and DMSO/EtOH as the solvent, Cu-catalyzed Glaser coupling-cycloaddition processes are dramatically enhanced that a diverse range of chalcogenophenes can be obtained in good to excellent yields under mild conditions. Controlled experiments show that trisulfur radical anion (S3 & sdot;-) is the key intermediate and DMSO/EtOH serves as the hydrogen source, while alcohol may facilitate the transition state. The mild and highly efficient constructions of 2,5-disubstitued thiophenes, selenophenes and tellurophenes are achieved by using a one-pot, two-step protocol that combine Cu-catalyzed Glaser coupling with LiNH2-mediated cycloaddition. And the DMSO/EtOH mixture solvent is found to short the reaction time and improve the reaction yield by promoting the production of S3 & sdot;- and stabilizing the transition state.image

Abstract:Tandem Friedel–Crafts acylation and alkylation of arenes with 2-alkenoyl chlorides were investigated under the catalysis of Lewis acids. The cascade reaction affords dihydrochalcones in good yields accompanying 1-indanone derivatives in some cases, in the presence of anhydrous aluminum chloride. The scope, limitation, and mechanism of the tandem reaction were also explored. The intermolecular Friedel–Crafts alkylation for the formation of dihydrochalcones is more favorable than the intramolecular one for the generation of 1-indanones in the tandem reaction due to a stable six-membered ring transition state. The sequent process was further studied by the DFT computations at the M06-2X/6-31G(d) level, which are in great agreement with the experimental observation and support the proposed mechanism. The current method provides a convenient and economic method to synthesis of dihydrochalcones.

Abstract:An efficient RuPHOX‐Ru catalyzed asymmetric cascade hydrogenation of 3‐substituted chromones has been achieved under mild reaction conditions, affording the corresponding chiral 3‐substituted chromanols in high yields with excellent enantio‐ and diastereoselectivities (up to 99% yield, >99% ee and >20:1 dr). Control reactions and deuterium labelling experiments revealed that a dynamic kinetic resolution process occurs during the subsequent hydrogenation of the C=O double bond, which is responsible for the high performance of the asymmetric cascade hydrogenation. The resulting products allow for several transformations and it was shown that the protocol provides a practical and alternative strategy for the synthesis of chiral 3‐substituted chromanols and their derivatives.

Abstract:A general and efficient visible-light-induced CF2 radical-based cascade reaction of o-hydroxyaryl enaminones has been described. This protocol afforded the corresponding functionalized 3-CF2-containing chromones in generally good yields under mild reaction conditions. The use of NaHSO3, which has both basicity and reducibility, efficiently suppressed the formation of two byproducts, 4H-chromen-4-one and 3-bromo-4H-chromen-4-one.

Abstract:A convenient method was developed via an NHC mediated three-component cascade reaction for the construction of the medicinally important spirobenzofuranone derivatives. The reaction features mild reaction conditions and generates three consecutive stereocenters, one of which is an all-carbon substitution. The protocol can accommodate various substituents and substitution patterns, afford the products in moderate to good yields with excellent diastereoselectivities and moderate enantioselectivity.

Abstract:A three-step process for the enantioselective assembly of cis-fused octahydrophenanthrenes with a quaternary stereocenter is reported.
