Abstract:An efficient and mild method is reported for the construction of beta-chlorotetrahydrofuran derivatives by 5-endo chlorocycloetherification of homoallylic alcohols. The system employs sulfuryl chloride as the chlorinating agent under catalyst-free conditions. A variety of homoallylic alcohols with aryl or alkyl substituents were smoothly converted into beta-chlorotetrahydrofurans in yields up to 98%.

Abstract:Enantioselective trifluoromethylation of aromatic ketones promoted by the cinchona alkaloid-derived ammonium bromide and sodium hydride was described. A series of trifluoromethyl-substituted aryl alcohols could be obtained in up to 82% ee with 98% yield under mild conditions. A possible catalytic cycle was also presented.

Abstract:In this paper, an efficient way to synthesize 2-(1'(Z)-iodoalkenyl)tetrahydrofurans from 4,5-alkadienols and 12 was developed. The reaction of the 4,5-allenols with a substituent in the 3-position afforded the trans-2,3-disubstituted tetrahydrofurans with very high diastereoselectivity. However, when the axially optically active 4,5-allenol was treated with 12 in n-hexane, the efficiency for chirality transfer was low. This problem was circumvented by conducting the reaction in CH2Cl2 at room temperature and applying N-iodosuccinimide as the electrophilic reagent; however, the Z/E ratio for the products is much lower. Highly optically active Z-products may be prepared via the kinetic resolution via a Sonogashira coupling reaction with propargyl alcohol.

Abstract:HHerein, we disclosed the asymmetric construction of an oxa‐quaternary stereocenter via an intramolecular oxa‐Michael (IOM) reaction in β‐substituted ortho‐hydroxymethyl chalcone by the formation of 1,1‐disubstituted‐1,3‐dihydroisobenzofuran using cinchona alkaloid‐based chiral amino‐squaramide catalyst. Both the (E‐ and Z)‐β‐substituted ortho‐hydroxymethyl chalcone provide (S)‐ and (R)‐enantiomers of the 1,1‐disubstituted‐1,3‐dihydroisobenzofuran with excellent stereospecificity. In general, excellent yields (up to 95%) and enantioselectivity (up to 98% ee) were obtained. Furthermore, the resulting 1,1‐disubstituted isobenzofuran or phthalan was converted to corresponding chiral 3,3‐disubstituted phthalides without losing the enantioselectivity. This methodology provides the core moiety of the (S)‐citalopram drug.

Abstract:Introduction of the trifluoromethyl group (CF3 ) into organic molecules in an enantioselective manner has attracted significant attention, but still remains a challenging problem. We herein report a catalytic asymmetric trifluoromethylation of cyclic ketones via a ScIII /chiral bisoxazoline-catalyzed homologation reaction by employing 2,2,2-trifluorodiazoethane (CF3 CHN2 ) as the CF3 source. This desymmetrization process is highly efficient and generates two chiral centers with excellent diastereoselectivity and enantioselectivity, affording chiral α-trifluoromethyl cyclic ketones in a straightforward manner.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:Chiral tetrahydrofurans are prevalent structural motifs in biologically active compounds, approved drugs and natural products, but relevant enantiocomplementary synthesis remains underdeveloped. In this study, wild type halohydrin dehalogenase HheC from Agrobacterium radiobacter AD1, was first discovered to enable the intramolecular substitution of δ‐halohydrins for the formation of tetrahydrofurans but with unsatisfied activity and stereoselectivity. Through structure‐guided protein engineering, two completely enantiocomplementary variants T134M and P84L/W139A of HheC were obtained for the synthesis of tetrahydrofurans by the kinetic resolution of δ‐halohydrins (both giving products with up to 99% e.e., E >200). This research unveiled the halohydrin dehalogenases' catalytic ability to form chiral cyclic ethers larger ring size than three‐membered epoxides, which has hitherto been unexplored, and provided an enzymatic synthesis access to valuably enantiocomplementary tetrahydrofurans.

Abstract:Fluorinated motifs are frequently encountered in drugs and agrochemicals. Incorporating fluorine-containing motifs in drug candidates for lead optimization in pharmaceutical research and development has emerged as a powerful tool. The construction of molecules that feature a trifluoromethyl (CF3−) group on a stereogenic carbon has accumulated broad research efforts. Unlike its well-explored, biologically active methyl counterpart, asymmetric construction of β-trifluoromethylated alcohols bearing adjacent stereocenters still remains elusive. Through retrosynthetic analysis, we posited that followed by sequential reduction of carbonyl, the initial construction of chiral α-trifluoromethylated ketones could render the desired product in a facile, one-pot fashion. Herein, we developed the first example of nickel-catalyzed asymmtric reductive cross-coupling trifluoroalkylation of acyl chlorides for enantioselective synthesis of diverse α-trifluoromethylated ketones. The one-pot reduction of these α-trifluoromethylated ketones furnished corresponding alcohols bearing β-CF3-substituted stereogenic carbons with excellent diastereoselectivity and complete enantioselective retention. High yields/enantioselectivity, mild conditions, and good functional group compatibility are shown in the system. Abstract:The catalytic enantioselective desymmetrizing bromoaminocyclization of prochiral cyclohexa-1,4-dienes has been achieved by using chiral anion phase-transfer catalysis, providing a range of enantioenriched cis-3a-arylhydroindoles bearing an all-carbon quaternary stereocenter in good yields (up to 78%) and excellent enantioselectivities (up to 97% ee).
