Abstract:In this communication, a straighforward asymmetric synthesis of spiro‐indoline‐pyrazolone compounds is described. This methodology consists in a formal [4+1] cycloaddition reaction of 4‐bromopyrazolones and aza‐ortho‐quinone methides generated in situ catalyzed by a bisquinine‐derived squaramide in CHCl3 under basic conditions. A variety of chiral spirocyclic compounds bearing a pyrazolone and an indoline moieties were obtained in moderate to good yields (up to 68%) and moderate to excellent enantioselectivities (up to 93% ee).

Abstract:A palladium-catalyzed C-H functionalization/intramolecular asymmetric allylation cascade of N-sulfonyl benzamides with 1,3-dienes has been developed. In the presence of a chiral pyridine-oxazoline ligand, this protocol enables the synthesis of chiral 3,4-dihydroisoquinolones in yields of up to 83% with enantioselectivities of up to 96%, using environmentally friendly air as the terminal oxidant.

Abstract:An efficacious method in which BINOL-type chiral imidodiphosphoric acid catalyzed the asymmetric [5 + 1] annulation reaction of 2-pyrrolylphenol with 1-methylindoline-2,3-dione was established. The strategy tolerated a broad substrate scope, and 30 examples were obtained. A range of enantioenriched spiro[3,2'-morpholine-oxindole] derivatives which incorporate a tertiary stereocenter, with moderate to excellent yields (up to 96%) and enantioselectivities (up to 99%) under mild conditions, was delivered.

Abstract:An unprecedented chiral secondary amine-catalyzed [3 + 3] annulation of isatin N,N'-cyclic azomethine imines with α,β-unsaturated aldehydes was developed. Abstract:In this work, we present an organocatalytic spiroannulation method that provides access to spirocyclic benzofuran‐2‐ones. The reaction proceeds via the generation of an azolium dienolate from an α‐bromo‐α,β‐unsaturated aldehyde using a chiral N heterocyclic carbene, followed by annulation with benzofuran‐2‐one‐derived alkenes. The resulting chiral spirobenzofuranone‐fused cyclohexenones were obtained with enantioselectivity ranging from 77% to 99% ee and diastereomeric ratios (dr) from 1.2/1 to 20/1. The utility of the protocol was underscored by gram‐scale reaction and follow‐up transformations.

Abstract:A diastereoselective and enantioselective formal [4+1] ylide annulation of chiral sulfonium salts with various substituted Morita-Baylis-Hillman (MBH) adducts leading to optically active isoxazoline N-oxides with three chiral carbon centers has been explored. The salient features of this methodology include a high diastereo- and enantioselective transformation, easily accessible starting materials, and mild reaction conditions. In addition, the isoxazoline N-oxides products can be conveniently transformed into functionalized compounds.

Abstract:A chiral NHC-catalyzed [3 + 3] cycloaddition reaction of 3-aminobenzofurans with isatin-derived enals has been documented, furnishing 3,4'-piperidinoyl spirooxindoles bearing a quaternary stereocenter with good yields and excellent enantioselectivities. Further gram-scale preparation and synthetic transformation of the cycloadducts to δ-amino acid derivative demonstrated good practicality and applicability of this reaction.

Abstract:Exploration of the diastereodivergent synthesis of spirocyclic oxindoles has been challenging. Herein we report asymmetric [3 + 2] annulations of isatin-derived Morita–Baylis–Hillman (MBH) carbonates and 5-alkenylthiazol-4(5H)-ones. Interestingly, two different chiral catalysts, amide-phosphine and 4-dimethylaminopyridine (DMAP)-thiourea, could lead to two kinds of diastereomeric dispiro oxindoles with three contiguous stereogenic centers. The hexafluoroisopropanol (HFIP) additive plays a vital role in accelerating the reaction and tuning the diastereoselectivity.
