Abstract:A direct, efficient, and practical protocol to install a chiral oxazoline unit onto aryl/heteroaryl rings via palladium-catalyzed C-H functionalization of 2-positions of oxazolines with a variety of halides using dppe as the ligand has been developed. Various chiral oxazoline ligands could be synthesized, even in a 10-g scale process. This protocol is a good supplement to traditional methods and for diverse synthesis of chiral oxazoline ligands.

Abstract:A palladium-catalyzed C-H functionalization/intramolecular asymmetric allylation cascade of N-sulfonyl benzamides with 1,3-dienes has been developed. In the presence of a chiral pyridine-oxazoline ligand, this protocol enables the synthesis of chiral 3,4-dihydroisoquinolones in yields of up to 83% with enantioselectivities of up to 96%, using environmentally friendly air as the terminal oxidant.

Abstract:We report herein the Pd-catalyzed oxazoline-directed C–H olefination of the <i>N</i>-arylindole skeleton, affording two diastereomers of axially chiral olefin–oxazoline ligands in a one-step procedure. Abstract:Oxazolines are a very important class of heterocyclic compounds. However, catalytic enantioselective syntheses are very limited. Here, a highly enantioselective palladium-catalyzed coupling-cyclization of readily available N-(buta-2,3-dienyl) amides with aryl or 1-alkenyl iodides has been developed for the asymmetric construction of oxazoline derivatives. Many synthetically useful functional groups are tolerated in this reaction. The absolute configuration of the chiral center in the products has been established by X-ray diffraction study. A model for prediction of the absolute configuration of the chiral center in the products from this cyclic enantioselective nucleophilic allylation has been proposed. The synthetic potentials based on the unique structure of the products formed have also been demonstrated.

Abstract:An enantioselective palladium-catalyzed C-H arylation of functionalized pyrazoles/triazoles/imidazoles is developed, affording a variety of axially chiral ortho-nitro/formyl-substituted heterobiaryls with excellent enantioselectivities and good yields. The method features a deuterated P-chiral phosphorus ligand CD(3)-AntPhos, a broad substrate scope of functionalized heterobiaryls, mild reaction conditions, and low palladium loadings.

Abstract:A palladium-catalyzed asymmetric chloroenolate allylation with vinyl benzoxazinanones under mild reaction conditions has been developed, affording a series of optically active 3,3-disubstituted oxindoles exhibiting a chloro-group and a linear aryl amino side chain in good yields with up to 96% ee. Versatile functional group tolerance on the benzene ring has been demonstrated, and the utility of this method was probed by a scale-up synthesis and highlighted by product derivatizations.

Abstract:Pyrazolones are a vital class of heterocycles possessing various biological properties and much attention is paid to the diversified synthesis of enantiopure pyrazolone derivatives. We describe here the development of diphenylphosphinoalkanoic acid based chiral bisphosphine ligands, which are successfully applied to the palladium-catalyzed asymmetric allenylation of racemic pyrazol-5-ones. The reaction affords C-allenylation products, optically active pyrazol-5-ones bearing an allene unit, in high chemo- and enantioselectivity, with DACH-ZYC-Phos-C1 as the best ligand. The synthetic potential of the C-allenylation products is demonstrated. Furthermore, the enantioselectivity observed with DACH-ZYC-Phos-C1 is rationalized by density functional theory studies.

Abstract:A general catalytic protocol for the Pd‐catalyzed intramolecular arylations of N‐(2‐halogenoaryl) amides in eco‐friendly medium was successfully developed by using rationally designed acenaphthoimidazolylidene‐ligated chiral oxazoline palladacycles. Under the optimal reaction condition, a broad scope of substrates with various functional groups were well tolerated, leading to desired 3,3'‐disubstituted oxindoles in excellent yields (up to 98% yield).

Abstract:In transition metal-catalyzed asymmetric synthesis, enantioselectivity strongly depends on the structures of chiral ligands, so the development of new chiral ligands is crucial. Here, an efficient and highly enantioselective palladium-catalyzed intramolecular hydroarylation has been developed, and a new kind of N-heterocycles, 1H-pyrazolo[5,1-a]isoindol-2(8H)-ones containing a quaternary stereocenter, was prepared in high yields and excellent enantiomeric excess values. The reaction was effectively catalyzed by palladium-diphosphine complexes with numerous functional group tolerance, in which the newly developed axially chiral cyclic diphosphine ligands played key roles in the reactivity and enantioselectivity of the substrates. We believe that the cyclic diphosphine ligands with adjustable dihedral angles will find wide application in asymmetric synthesis.

Abstract:Reported here is a highly efficient Pd/Xiang-Phos catalyzed enantioselective carboetherification of alkenyl oximes with either aryl or alkenyl halides, delivering various chiral 3,5-disubstituted and 3,5,5-trisubstituted isoxazolines in good yields with up to 97 % ee. The sterically bulky and electron-rich (S,Rs)-NMe-X2 ligand is responsible for the excellent reactivities and enantioselectivities. The salient features of this transformation include mild reaction conditions, general substrate scope, good functional-group tolerance, good yields, high enantioselectivities, easy scale-up, and application in the late-stage modification of bioactive compounds. The obtained products can be readily transformed into useful chiral 1,3-aminoalcohols.

Abstract:Catalytic asymmetric [4+2] cycloadditions via allylic C–H functionalization are of great interest and importance but remain unexplored. We report herein an intermolecular [4+2] cycloaddition involving the direct Pd(0)-catalyzed allylic C–H functionalization with readily accessible olefin substrates. Key to the success of this reaction is a chiral alkylphosphine ligand, which enables the asymmetric allylic C–H alkylation to deliver a broad range of hydroquinolines in high diastereoselectivities (up to >20:1) and enantioselectivities (up to 98% ee). Control experiments show that allylic C–H functionalization takes place possibly via the concerted proton and two-electron transfer process. Abstract:Metal-catalyzed alkene arylalkoxylation is a powerful complexity-building strategy for the synthesis of oxygen heterocycles from simple γ-unsaturated alcohols, but only a few examples of catalytic enantioselective methods exist. Herein, an efficient palladium-catalyzed enantioselective arylalkoxylation of γ-hydroxyalkenes with aryl halides is reported. The salient features of this transformation include a remarkable broad substrate scope, mild reaction conditions, and good functional group tolerance, delivering a series of chiral tetrahydrofurans containing a tertiary or quaternary stereocenter in good yields with up to 95% ee. The Xu10 ligand with a suitable side-arm was responsible for the high reactivity and good enantioselectivity of this transformation.

Abstract:Enantioselective synthesis of tetrahydroisoquinolines bearing an all-carbon quaternary stereogenic center, was achieved via asymmetric C-H activation with high enantioselectivities (up to 93% ee). Fair substrate tolerance was indicated throughout the scope investigation and no evident loss of enantioselectivity was exhibited in late-stage derivatization. This study provides incentives for the construction of diverse chiral isoquinoline derivatives, which are prevalent among pharmaceuticals, natural products, etc.

Abstract:Enantiomerically pure acyclic O,O-acetal compounds (up to 97 % ee) have been accessed through chemo-, regio- and enantioselective palladium-catalyzed addition of oximes to alkoxyallenes. DFT calculations support that a protonative hydropalladation pathway is favourable, in which the hydrogen bonding interaction between the amide group of the diphosphine ligand and the alkoxyallene is critical for the highly stereoselective formation of the dioxygenated stereogenic center.
