Abstract:Photoacoustic microscopy (PAM) does not rely on contrast agent to image the optical absorption contrast in biological tissue. It is uniquely suited for measuring several tissue physiological parameters, such as hemoglobin oxygen saturation, that would otherwise remain challenging. Researchers are designing new clinical diagnostic tools and multimodal microscopic systems around PAM to fully unleash its potential. However, the sizeable and opaque piezoelectric ultrasonic detectors commonly used in PAM impose a serious constraint. Our solution is a coverslip-style optically transparent ultrasound detector based on a polymeric optical micro-ring resonator (MRR) with a total thickness of 250 μm. It enables highly-sensitive ultrasound detection over a wide receiving angle with a bandwidth of 140 MHz, which corresponds to a photoacoustic saturation limit of 287 cm −1 , at an estimated noise-equivalent pressure (NEP) of 6.8 Pa. We also established a theoretical framework for designing and optimizing the MRR for PAM.

Abstract:Photoacoustic imaging is a new type of noninvasive, nonradiation imaging modality that combines the deep penetration of ultrasonic imaging and high specificity of optical imaging. Photoacoustic imaging systems employing conventional ultrasonic sensors impose certain constraints such as obstructions in the optical path, bulky sensor size, complex system configurations, difficult optical and acoustic alignment, and degradation of signal-to-noise ratio. To overcome these drawbacks, an ultrasonic sensor in the optically transparent form has been introduced, as it enables direct delivery of excitation light through the sensors. In recent years, various types of optically transparent ultrasonic sensors have been developed for photoacoustic imaging applications, including optics-based ultrasonic sensors, piezoelectric-based ultrasonic sensors, and microelectromechanical system-based capacitive micromachined ultrasonic transducers. In this paper, the authors review representative transparent sensors for photoacoustic imaging applications. In addition, the potential challenges and future directions of the development of transparent sensors are discussed.

Abstract:OBJECTIVE: Photoacoustic (PA) imaging emerges as a unique tool to study biological samples based on optical absorption contrast. In PA imaging, piezoelectric transducers are commonly used to detect laser-induced ultrasonic waves. However, they typically lack adequate broadband sensitivity at ultrasonic frequency higher than 100 MHz, whereas their bulky size and optically opaque nature cause technical difficulties in integrating PA imaging with conventional optical imaging modalities. To overcome these limitations, optical methods of ultrasound detection were developed and shown their unique applications in PA imaging.METHODS: We provide an overview of recent technological advances in optical methods of ultrasound detection and their applications in PA imaging. A general theoretical framework describing sensitivity, bandwidth, and angular responses of optical ultrasound detection is also introduced.RESULTS: Optical methods of ultrasound detection can provide improved detection angle and sensitivity over significantly extended bandwidth. In addition, its versatile variants also offer additional advantages, such as device miniaturization, optical transparency, mechanical flexibility, minimal electrical/mechanical crosstalk, and potential noncontact PA imaging.CONCLUSION: The optical ultrasound detection methods discussed in this review and their future evolution may play an important role in PA imaging for biomedical study and clinical diagnosis.

Abstract:We present in this paper a systematic study of the distance dependent detection characteristics of the newly developed micro-ring resonator (MRR)-based ultrasonic detector for photoacoustic microscopy (PAM). A simple analytic model was first developed to study the steady-state response to the continuous ultrasonic waves. While placing the MRR detector at the acoustic far-field provides longer working distance and broader field of view, the detection at acoustic near-field offers the improved sensitivity and broader bandwidth but at the cost of reduction in the field of view. Furthermore, a numerical model was developed to analyze the transient response to the photoacoustic-induced impulsive waves. Notably, far-field detection exhibits a flat wavefront of its response pattern in the time domain while large distortions are clearly visible in the case of near-field detection. Finally, both analytic and numerical models are validated by experimental studies. This work establishes a theoretical framework for quantitatively analyzing the trade-offs between near-field and far-field detection using MRR detector, creating a guideline for optimizing the PAM for various applications in biomedical imaging and diagnostics. Abstract:Abstract Photoacoustic (PA) imaging enables noninvasive volumetric imaging of biological tissues by capturing the endogenous optical absorption contrast. Conventional ultrasound detectors using piezoelectric materials have been widely used for transducing ultrasound signals into the electrical signals for PA imaging reconstruction. However, their inherent limitations in detection bandwidth and sensitivity per unit area have unfortunately constrained the performance of PA imaging. Optical based ultrasound detection methods emerge to offer very promising solutions. In particular, polymer micro-ring resonators (MRRs) in the form of integrated photonic circuits (IPC) enable significant reduction for the sensing area to 80 μm in diameter, while maintaining highly sensitive ultrasound detection with noise equivalent pressure (NEP) of 0.49 Pa and a broad detection frequency range up to 250 MHz. The continued engineering innovation has further transformed MRRs to be transparent to the light and thus, opens up a wide range of applications, including multi-modality optical microscope with isometric resolution, PA endoscope, photoacoustic computed tomography (PACT), and more. This review article summarizes and discusses the evolution of polymer MRR design and the associated nanofabrication process for improving the performance of ultrasound detection. The resulting novel imaging applications will also be reviewed and discussed.

Abstract:Optical whispering-gallery microresonators have attracted considerable interest for ultrasensitive ultrasound detection and photoacoustic imaging because of the combination of high quality factors and small cavity sizes. In the last decade, ultrasonic sensors with on-chip microcavities have been extensively developed; however, they are unsuitable for the near-field photoacoustic microscopy of micro/nanoscale objects in complex biological environments and endoscopic imaging. In this work, we developed ultrasonic sensors using two types of encapsulated microsphere resonators with different cavity materials. A noise equivalent pressure of as low as 160 Pa at 20 MHz was achieved with the acoustic response up to 70 MHz at-6 d B. Furthermore,the microsensor was used for photoacoustic microscopy in which we successfully performed 3 D imaging of hairs and leaf veins.The microsphere ultrasonic sensor has considerable potential as a probe-type ultrasonic detector for near-field photoacoustic microscopy of micro/nanoscale objects such as subcellular structures and high-resolution endoscopic photoacoustic imaging with its high sensitivity and wide bandwidth.

Abstract:Photoacoustic tomography emerged as a promising tool for noninvasive biomedical imaging and diseases diagnosis. However, most of the current piezoelectric ultrasound transducers suffer optical opacity and tissue-mismatched acoustic impedance, hindering the miniaturization and integration of the system for multiscale and multimodal imaging. Here, a transparent polydimethylsiloxane (PDMS) encapsulated optical microfiber ultrasound sensor was demonstrated for photoacoustic imaging with scalable spatial resolution and penetration depth. The sensor comprised a microfiber loop sandwiched by a pair of in-line Bragg gratings, which formed an ultrasound-sensitive Fabry-Perot cavity allowing free delivery of ultrasound/light beams and unique needle-shaped ultrasound focusing along the penetration depth. The sensor with a detection limit of ∼ 700 Pa and a bandwidth of ∼ 10 MHz was applied for multiscale photoacoustic imaging of mouse ear and brain vasculatures. With advantages of flexibility, optical transparence and focusing capability, the sensor offers new opportunities for developing photoacoustic/ultrasound imaging devices for biomedical and clinic applications.

Abstract:We designed an all-optical photoacoustic (PA) probe for endoscopic applications by employing an optically transparent, coverslip-type, polymeric microring resonator ultrasonic sensor. We experimentally quantified the axial, tangential, and radial resolutions and angular sensitive stability of this probe. Using this probe, we achieved volumetric imaging of several phantoms. Our all-optical probe design offers clear benefit in integrating PA endoscope with other optical endoscopic imaging modalities to facilitate the transformation from bench to bedside.

To avoid these limitations, various optical ultrasound sensors have been developed and shown their great potential in PA imaging.AimOur study aims to summarize recent progress in optical ultrasound sensor technologies and their applications in PA imaging.ApproachThe commonly used optical ultrasound sensing techniques and their applications in PA systems are reviewed. The technical advances of different optical ultrasound sensors are summarized.ResultsOptical ultrasound sensors can provide wide bandwidth and improved sensitivity with miniatured size, which enables their applications in PA imaging.ConclusionsThe optical ultrasound sensors are promising transducers in PA imaging to provide higher-resolution images and can be used in new applications with their unique advantages.

Abstract:Optical ultrasonic probes, exemplified by Fabry–Perot cavities on optical fibers, have small sizes, high sensitivity, and pure optical characteristics, making them highly attractive in high-resolution ultrasonic/photoacoustic imaging, especially in near-field or endoscopic scenarios. Taking a different approach, we demonstrate an ultrasensitive and broadband ultrasound microprobe formed by an optical whispering-gallery-mode polymer microcavity coupled to a U-shaped microfiber. With the high-quality (Q) factors (>106), the noise equivalent pressure of the ultrasound microprobe reaches 1.07 mPa/√Hz with a record broadband response of 150 MHz and a large detection angle of 180°. Our results show that this optical microprobe can overcome the strong decay resulting from ultrasound diverging and medium absorption through short working distances. We further demonstrate high-quality in vivo whole-body photoacoustic imaging of a zebrafish larva. Our implementation provides a new strategy for developing miniature ultrasound detectors and holds great potential for broad applications.

Abstract:Ultrasound detection is one of the major components of photoacoustic imaging systems. Advancement in ultrasound transducer technology has a significant impact on the translation of photoacoustic imaging to the clinic. Here, we present an overview on various ultrasound transducer technologies including conventional piezoelectric and micromachined transducers, as well as optical ultrasound detection technology. We explain the core components of each technology, their working principle, and describe their manufacturing process. We then quantitatively compare their performance when they are used in the receive mode of a photoacoustic imaging system.

Abstract:Photoacoustic mesoscopy is an emerging noninvasive imaging modality, which offers high resolution 3D images with optical absorption contrast at depths beyond the light diffusion limit. Ultrasound sensor based on a Fabry-Perot (FP) polymer cavity has the following advantages: broadband frequency response, wide angular coverage and small footprint. We present a photoacoustic mesoscope based on a tunable Fabry-Perot interferometer, which offers the potential for reducing system cost and making array of such sensor. A cw diode laser working at 650nm was used to heat the sensor, offering an active tune range of 5nm by elongating the cavity. Ex-vivo and in-vivo imaging experiments demonstrated the imaging capability of this PA mesoscope, showing great potential in biological and medical applications.

Abstract:Photoacoustic microscopy (PAM) is an attractive imaging tool that complements established optical microscopic modalities by providing additional molecular specificities through imaging optical absorption contrast. While the development of optical resolution photoacoustic microscopy (ORPAM) offers high lateral resolution, the acoustically determined axial resolution is limited due to the constraint in ultrasonic detection bandwidth. ORPAM with isometric spatial resolution along both axial and lateral directions is yet to be developed. Although recently developed sophisticated optical illumination and reconstruction methods offer improved axial resolution in ORPAM, the image acquisition procedures are rather complicated, limiting their capabilities for high-speed imaging and being easily integrated with established optical microscopic modalities. Here we report an isometric ORPAM based on an optically transparent micro-ring resonator ultrasonic detector and a commercial inverted microscope platform. Owing to the superior spatial resolution and the ease of integrating our ORPAM with established microscopic modalities, single-cell imaging with extrinsic fluorescence staining, intrinsic autofluorescence, and optical absorption can be achieved simultaneously. This technique holds promise to greatly improve the accessibility of PAM to broader biomedical researchers. (C) 2015 Optical Society of America

Abstract:Photoacoustic technology is a promising tool to provide morphological and functional information in biomedical research. To enhance the imaging efficiency, the reported photoacoustic probes have been designed coaxially involving complicated optical/acoustic prisms to bypass the opaque piezoelectric layer of ultrasound transducers, but this has led to bulky probes and has hindered the applications in limited space. Though the emergence of transparent piezoelectric materials helps to save effort on the coaxial design, the reported transparent ultrasound transducers were still bulky. In this work, a miniature photoacoustic probe with an outer diameter of 4 mm was developed, in which an acoustic stack was made with a combination of transparent piezoelectric material and a gradient-index lens as a backing layer. The transparent ultrasound transducer exhibited a high center frequency of ~47 MHz and a −6 dB bandwidth of 29.4%, which could be easily assembled with a pigtailed ferrule of a single-mode fiber. The multi-functional capability of the probe was successfully validated through experiments of fluid flow sensing and photoacoustic imaging.

Abstract:Multi-modality imaging is significant for biomedical applications. We propose a dual-sensitive sensor to simultaneously detect optical and ultrasonic signals. Based upon the classical piezoelectric structure, we attach a photosensitive layer made of carbon nanotubes-polydimethylsiloxane (CNTs-PDMS) composite to the surface. The photosensitive layer absorbs light and converts it into ultrasound, while allowing acoustic energy to transmit through concurrently. After optimizing the ratio of PDMS to CNTs, we increase the sensor's light detection sensitivity and maintain the ultrasound detection sensitivity. Finally, the successful implementation in mouse ear optical attenuation-photoacoustic imaging demonstrates the dual-sensitive sensor's potential application in multi-modality imaging.

Abstract:In recent years, photoacoustics has attracted intensive research for both anatomical and functional biomedical imaging. However, the physical interaction between photoacoustic generated endogenous waves and an exogenous ultrasound wave is a largely unexplored area. Here, we report the initial results about the interaction of photoacoustic and external ultrasound waves leading to a micro-Doppler photoacoustic (mDPA) effect, which is experimentally observed and consistently modelled. It is based on a simultaneous excitation on the target with a pulsed laser and continuous wave (CW) ultrasound. The thermoelastically induced expansion will modulate the CW ultrasound and leads to transient Doppler frequency shift. The reported mDPA effect can be described as frequency modulation of the intense CW ultrasound carrier through photoacoustic vibrations. This technique may open the possibility to sensitively detect the photoacoustic vibration in deep optically and acoustically scattering medium, avoiding acoustic distortion that exists in state-of-the-art pulsed photoacoustic imaging systems.

Abstract:Two kinds of piezoelectric PZT, cylindrical tube and disk, and PVDF transducers are utilized to detect the photoacoustic signals of chemical solutions. The vibration modes of the PZT tube and the acoustic pressure distributions along the axis of the tube, which are important for photoacoustic calorimetry, are analyzed in detail. The acoustic properties of PZT disk and PVDF film transducers are also described.

Abstract:Photoacoustics has been broadly studied in biomedicine, for both human and small animal tissues. Photoacoustics uniquely combines the absorption contrast of light or radio frequency waves with ultrasound resolution. Moreover, it is non-ionizing and non-invasive, and is the fastest growing new biomedical method, with clinical applications on the way. This review provides a brief recap of recent developments in photoacoustics in biomedicine, from basic principles to applications. The emphasized areas include the new imaging modalities, hybrid detection methods, photoacoustic contrast agents and the photoacoustic Doppler effect, as well as translational research topics.

Abstract:Over the past decade, ultraviolet (UV) detection has been a subject of major interest for both research scientists and engineers because of its important applications in both the civil and military fields. The rapid development of interdisciplinary research has enabled the realization of UV detectors based on a variety of principles. Among these devices, UV detectors based on surface acoustic wave (SAW) technology offer unique advantages of remote wireless operation capability and zero power consumption. This article provides a comprehensive review of the working principles, important parameters, and the acoustic wave and materials types used in SAW-based UV detectors. The research and development status of these detectors are discussed and the most commonly used methods to optimize device performance are also summarized. Novel types of acoustic UV detectors based on thin film bulk acoustic resonators (FBARs) and Lamb wave resonators (LMRs) are briefly introduced. Finally, future development challenges are proposed and suggestions for future directions are provided to aid the development of this important research field.
