Abstract:Photoelectrochemical detection represents a unique signal transducing modality, where the photogenerated charge carriers are modulated by the redox reactions of molecular targets over the electrode surface. In this work, we report a novel glucose sensor based on hydrothermally-grown, single-crystalline TiO2 nanowires with surface-functionalized glucose oxidase (GOx), which can oxidize glucose to gluconic acid. The key feature of this glucose sensor design is that the photogenerated holes over the TiO2 anode are utilized to form O-2, which directly serves as an efficient electron shuttling mediator between the enzymatic redox center of GOx and the sensor surface, thus leading to an increase of the photocurrent. As a proof-of-concept, the GOx-functionalized TiO2 (TiO2-GOx) nanowires exhibit a high sensitivity of similar to 0.9 nM in the detection of glucose in buffer. The capability of detecting glucose in mouse serum is also demonstrated. Our work suggests the potential of developing these nanowire-based photoelectrochemical biosensors for convenient, efficient and low-cost biomarker detection and disease diagnosis.

Abstract:As a type of nanomaterials with enzyme-mimetic catalytic properties, nanozymes have attracted wide concern in biological detection. H2O2 was the characteristic product of diverse biological reactions, and the quantitative analysis for H2O2 was an important way to detect disease biomarkers, such as acetylcholine, cholesterol, uric acid and glucose. Therefore, there is of great significance for developing a simple and sensitive nanozyme to detect H2O2 and disease biomarkers by combining with corresponding enzyme. In this work, Fe-TCPP MOFs were successfully prepared by the coordination between iron ions and porphyrin ligands (TCPP). In addition, the peroxidase (POD) activity of Fe-TCPP was proved, in detail, Fe-TCPP could catalyze H2O2 to produce ·OH. Herein, glucose oxidase (GOx) was chosen as the model to build cascade reaction by combining Fe-TCPP to detect glucose. The results indicated glucose could be detected by this cascade system selectively and sensitively, and the limit of detection of glucose was achieved to 0.12 μM. Furthermore, a portable hydrogel (Fe-TCPP@GEL) was further established, which encapsulated Fe-TCPP MOFs, GOx and TMB in one system. This functional hydrogel could be applied for colorimetric detection of glucose by coupling with a smartphone easily.

Abstract:Nanozyme/natural enzyme hybrid plays a vital role in biosensing, therapy, and catalysis owing to the integrated advantages in the selectivity of natural enzymes and controllable catalytic activity of nanozymes. Herein, Fe-MIL-88B-NH2 [(Fe-metal-organic framework (MOF)] with remarkable peroxidase-like activity, ultrahigh stability, and high biocompatibility was utilized for immobilization of glucose oxidase (GOx) via an amidation coupling reaction. On the basis of the excellent selectivity and catalytic activity of Fe-MOF-GOx, a cascade catalysis was performed for the colorimetric detection of glucose. The integrated Fe-MOF-GOx not only exhibited higher stability and reusability than their mixtures including Fe-MOF and free GOx system but also possessed a wide linear range (1-500 μM), with a low detection limit of 0.487 μM for glucose detection.

Abstract:A novel amperometric glucose biosensor based on self-assembling glucose oxidase (GOx) and dendrimer-encapsulated Pt nanoparticles (Pt-DENs) on nanofibrous polyaniline (PANI) was described. PANI nanofibers were synthesized via an interfacial polymerization method. A sulfonated polyelectrolytes-poly(sodium 4-styrenesulfonate) (PSS) was used to form the negative PANI/sulfonated polyelectrolyte complex, which had good disperse in aqueous solution. GOx was immobilized on the PANI/PSS surface by alternatively assembling a cationic Pt-DENs layer and an anionic GOx layer. The unique sandwich-like layer structure (Pt-DENs/GOx/Pt-DENs/PANI/PSS) formed by self-assembling provides a favorable microenvironment to keep the bioactivity of GOx and to prevent enzyme molecule leakage. The fabricated Pt-DENs/GOx/Pt-DENs/PANI/ PSS electrode exhibited excellent response performance to glucose with a detection limit of 0.5 mu M, wide linear range from 10 mu M to 4.5 mM, short response time within 5 s, improved sensitivity of 39.63 mu A/(mM cm(2)), and good stability (85% remains after 20 days). Abstract:Promoting the electrocatalytic oxidation of glucose is crucial in glucose biosensor design. In this study, nanoporous gold (NPG) was selected for glucose oxidase (GOx) immobilization and glucose biosensor fabrication because of its open, highly conductive, biocompatible, and interconnected porous structure, which also facilitates the electrocatalytic oxidation of glucose. The electrochemical reaction on the surface of the resulting GOx/NPG/GCE bioelectrode was attributed to the co-catalysis effect of GOx and NPG. A surface-confined reaction in a phosphate buffer solution was observed at the bioelectrode during cyclic voltammetry experiments. Linear responses were observed for large glucose concentrations ranging from 50μM to 10mM, with a high sensitivity of 12.1μAmM(-1)cm(-2) and a low detection limit of 1.02μM. Furthermore, the GOx/NPG/GCE bioelectrode presented strong anti-interference capability against cholesterol, urea, tributyrin, ascorbic acid, and uric acid, along with a long shelf-life. For the detection of glucose in human serum, the data generated by the GOx/NPG/GCE bioelectrode were in good agreement with those produced by an automatic biochemical analyzer. These unique properties make the GOx/NPG/GCE bioelectrode an excellent choice for the construction of a glucose biosensor.

Abstract:Considering the application prospects of a cascade catalytic system in biomedical analysis, it remains significant to develop a cascade catalytic system for one-step rapid colorimetric detection. Herein, PVP/NH 2 -MIL-101(Fe) possessing enhanced peroxidase ( POD )-like activity was successfully synthesized by microwave-assisted and polyvinylpyrrolidone ( PVP ) modification strategies. Then, PVP/NH 2 -MIL-101(Fe) and glucose oxidase ( GOx ) were covalently coupled via the bifunctional cross-linker glutaraldehyde, the GOx@PVP/NH 2 -MIL-101(Fe) with dual enzymatic functional activities (POD-like and GOx activities) was subsequently synthesized. GOx@PVP/NH 2 -MIL-101(Fe) had strong affinities for the substrates glucose, 3,3',5,5'-tetramethylbenzidine, and hydrogen peroxide. Based on this specific nanozyme GOx@PVP/NH 2 -MIL-101(Fe), a one-step colorimetric assay for rapid glucose determination was built. This detection method not only took 20 min to analyze but also possessed a linear range of 10–250 μM with a detection limit of 1.05 μM. Furthermore, the cascade catalytic system was applied to determine glucose concentrations in human serum samples. Compared with commercial blood glucose meters, the proposed method was also accurate and effective, making it a candidate method for determining blood glucose.

Abstract:Nanomaterials with enzyme-mimicking characteristics have engaged great awareness in various fields owing to their comparative low cost, high stability, and large-scale preparation. However, the wide application of nanozymes is seriously restricted by the relatively low catalytic activity and poor specificity, primarily because of the inhomogeneous catalytic sites and unclear catalytic mechanisms. Herein, a support-sacrificed strategy is demonstrated to prepare a single iron site nanozyme (Fe SSN) dispersed on the porous N-doped carbon. With well-defined coordination structure and high density of active sites, the Fe SSN performs prominent peroxidase-like activity by efficiently activating H2 O2 into hydroxyl radical (•OH) species. Furthermore, the Fe SSN is applied in colorimetric detection of glucose through a multienzyme biocatalytic cascade platform. Moreover, a low-cost integrated agarose-based hydrogel colorimetric biosensor is designed and successfully achieves the visualization evaluation and quantitative detection of glucose. This work expands the application of single-site catalysts in the fields of nanozyme-based biosensors and personal biomedical diagnosis.

Abstract:A novel amperometric glucose biosensor based on layer-by-layer (LbL) electrostatic adsorption of glucose oxidase (GOx) and dendrimer-encapsulated Pt nanoparticles (Pt-DENs) on multiwalled carbon nanotubes (CNTs) was described. Anionic GOx was immobilized on the negatively charged CNTs surface by alternatively assembling a cationic Pt-DENs layer and an anionic GOx layer. Transmission electron microscopy images and zeta-potentials proved the formation of layer-by-layer nanostructures on carboxyl-functionalized CNTs. LbL technique provided a favorable microenvironment to keep the bioactivity of GOx and prevent enzyme molecule leakage. The excellent electrocatalytic activity of CNTs and Pt-DENs toward H2O2 and special three-dimensional structure of the enzyme electrode resulted in good characteristics such as a low detection limit of 2.5 mu M, a wide linear range of 5 mu M - 0.65 mM, a short response time (within 5 s), and high sensitivity (30.64 mu A mM(-1) cm(-2)) and stability (80% remains after 30 days).

Abstract:A smart sensing platform based on a triboelectric nanogenerator (TENG) possesses various advantages such as self-powering, convenience, real-time and biocompatibility. However, the detection limit of the TENG-based sensor is required to be improved. In this study, a high performance TENG-based glucose sensor was proposed by using the Ti3C2Tx (MXene)/graphene oxide (GO) composite electrode. The MXene and GO nanosheets are popular 2D materials which possessed high conductivity and a rich surface functional group. The MXene/GO thin films were prepared through electrostatic self-assembly technology, which can effectively impede the agglomeration of two nanoflakes. The as-prepared MXene/GO film presented outstanding mechanical property. To figure out the relationship between the nanostructure of MXene/GO film and the TENG, a series of MXene/GO-based TENG with different GO sizes was characterized. As a result, the TENG with 400 nm GO demonstrated the highest output performance. Subsequently, the optimized TENG was used in glucose detection application without the assistance of a glucose enzyme. This simple and flexible TENG shows promising potential in biosensors and non-invasive health monitoring.

Glucose biosensors based on nano-composites of multi-walled carbon nanotubes and zinc oxide nanorods

Abstract:We demonstrate herein a newly developed amperometric glucose biosensor by using multi-walled carbon nanotubes (MWCNTs) and zinc oxide (ZnO) nanorods composites film. The latter is generated by utilizing an electrostatic coordination approach under the assistance of the ultrasonic. The presence of ZnO nanorods in the composites enhances the abilities to electrocatalyze the oxidation of H2O2 and substantially raises the response current, which results in the composites exhibiting more efficiently electrocatalytic activity than those of MWCNTs and ZnO alone. As a result of the cross-linking reactions via glutaraldehyde, a layer of glucose oxidase (GOx) was firmly bound to the MWCNTs-ZnO film and an anti-interferent layer of poly (o-phenylenediamine) (PoPD) film was further electropolymerized on the enzyme film. At an applied potential of +0.8 V, the resulting biosensor performs a sensitive and selective electrochemical response to glucose in the presence of common interferences, such as ascorbic acid (AA) and uric acid (UA), with a linear dependence (R=0.997) on the glucose concentration in the range of 5.0x10(-6)similar to 5.0x10(-3) mol . L-1 and a detection limit of 3.5x10(-6) mol . L-1 at signal/noise=3. The response time was less than 10 s with addition of 5 mmol . L-1 glucose. The MWCNTs-ZnO/GOx/PoPD modified glassy carbon electrode presents stable, high sensitivity and also exhibits fast amperometric response to the detection of glucose, which is promising for the development of glucose sensor.
Enhanced glucose detection using dendrimer encapsulated gold nanoparticles benefiting from their zwitterionic surface

Abstract:The application of ultrasmall gold nanoparticles as enzyme mimics has been drawing great attention. Herein, we developed zwitterionic dendrimer encapsulated gold nanoparticles (Au-G5MC NPs) for highly sensitive and simple colorimetric detection of glucose. Au-G5MC NPs showed peroxidase-like property, which could efficiently catalyze oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of H2O2, producing a blue color product (oxTMB). This peroxidase-like reaction follows a typical Michaelis-Menten kinetics. The Km towards TMB exhibited a lower value (0.194 mM) than that of horseradish peroxidase (HRP, 0.434 mM). Furthermore, the peroxidase-like properties of Au-G5MC NPs enable colorimetric detection of the concentration of glucose with high selectivity. The linear concentration range of this method was from 14 μM to 166 μM with the detection limit down to 3.8 μM. More importantly, the detection was not interfered by proteins due to the single zwitterionic layer on the Au-G5MC NPs surface. These excellent properties are attributed to the ultrasmall size of gold nanoparticles and high stability of Au-G5MC NPs in complex medium. This catalytic system might have great potential applications for glucose detection in medical diagnostics and biochemistry in the future.

Abstract:Novel glucose oxidase (GOX) sensor arrays with nanoelectrode population densities of up to 1×1010/cm2 nanoelectrodes are reported. These nanoelectrode sensors, polypyrrole (PPy) nanofibers containing entrapped GOX, were fabricated via a two-step process, which consisted of an electroadsorption of GOX followed by an electrochemical codeposition of PPy with GOX within the nanopores of an anodic aluminum oxide (AAO) membrane. It was found by SEM observation that GOX could intercalate in PPy nanofibers, especially in the region close to their side surface. The GOX/PPy nanofiber array electrode demonstrates a good biocatalytic activity to glucose with an apparent Michaelis–Menten constant (KMapp) of 5.7 mM and a response time of about 15 s. The PPy/GOX nanofiber array electrode may have potential applications in monitoring and diagnostic tests in practical use in the future.

Abstract:<p>Traditional colorimetric glucose biosensor generally involves complex assay procedures. Free labile enzymes and peroxidase substrates are used separately for triggering a chromogenic reaction. These limits result in inferior enzyme stability and defective enzymatic catalytic efficiency, making it hard to routinely utilize them for the direct and fast test of glucose. In this work, we provide an all-inclusive substrates/enzymes nanoparticle employed 3,3′5,5′-tetramethylbenzidine (TMB) as chromogenic substrates and glucose oxidase (GOx)/horseradish peroxidase (HRP) as signal amplifier enzymes (TMB-GH NPs) by the molecule self-assembly technique. The "all-inclusive" nanoparticles can realize the tandem colorimetric reactions, and the oxidation product of TMB (ox-TMB) exhibits a strong NIR laser-driven photothermal effect, thus allowing quantitative photothermal detection of glucose. Owing to the restriction of the molecular motion of GOx, HRP, and TMB, the distance of mass transfer between substrates was shortened largely, leading to improved catalytic activity for glucose. Overall, our strategy will simplify the analysis procedure, furthermore, these integrated nanoparticles not only display higher stability and activity than that of the free GOx/HRP system and possesses an excellent performance for colorimetric and photothermal bioassay of glucose simultaneously. We believe that this unique technique will give good inspirations to develop simple and precise methods for bioassay.</p>

A New Route to the Considerable Enhancement of Glucose Oxidase (gox) Activity: the Simple Assembly of A Complex from Cdte Quantum Dots and Gox, and Its Glucose Sensing

Abstract:A new complex consisting of CdTe quantum dots (QDs) and glucose oxidase (GOx) has been facilely assembled to achieve considerably enhanced enzymatic activity and a wide active temperature range of GOx; these characteristics are attributed to the conformational changes of GOx during assembly. The obtained complex can be simultaneously used as a nanosensor for the detection of glucose with high sensitivity. A mechanism is put forward based on the fluorescence quenching of CdTe QDs, which is caused by the hydrogen peroxide (H2O2) that is produced from the GOx-catalyzed oxidation of glucose. When H2O2 gets to the surface of the CdTe QDs, the electron-transfer reaction happens immediately and H2O2 is reduced to O2, which lies in electron hole traps on CdTe QDs and can be used as a good acceptor, thus forming the nonfluorescent CdTe QDs anion. The produced O2 can further participate in the catalyzed reaction of GOx, forming a cyclic electron-transfer mechanism of glucose oxidation, which is favorable for the whole reaction system. The value of the Michaelis-Menton constant of GOx is estimated to be 0.45 mM L(-1), which shows the considerably enhanced enzymatic activity measured by far. In addition, the GOx enzyme conjugated on the CdTe QDs possesses better thermal stability at 20-80 degrees C and keeps the maximum activity in the wide range of 40-50 degrees C. Moreover, the simply assembled complex as a nanosensor can sensitively determine glucose in the wide concentration range from micro- to millimolar with the detection limit of 0.10 microM, which could be used for the direct detection of low levels of glucose in biological systems. Therefore, the established method could provide an approach for the assembly of CdTe QDs with other redox enzymes, to realize enhanced enzymatic activity, and to further the design of novel nanosensors applied in biological systems in the future.

Abstract:Development of highly sensitive and accurate biosensors still faces a great challenge. Herein, glucose oxidase (GOx) is efficiently immobilized on the AuCu hydrogels owing to their porous structure and interfacial interaction, demonstrating enhanced catalytic activity, satisfactory stability and recyclability. Besides, by integration of AuCu@GOx and electrochromic material of Prussian blue, a sensitive and stable biosensing platform based on the excellent electrochromic property of Prussian blue and the enhanced enzyme activity of AuCu@GOx is developed, which enables the electrochemical and visual dual-mode detection of glucose. The as-constructed biosensing platform possesses a wide linear range, and good selectivity for glucose detection with a limit of detection of 0.82 μM in visual mode and 0.84 μM in electrochemical mode. This easy-to-operate biosensing platform opens a door for the practical application of the multi-mode strategy for glucose detection.

The electrode modified with the PB/MWNTs-GOx-CS-ICPTES sol–gel composite film showed good electrical conductivity and effective low-potential electron transfer mediation toward H2O2 reduction attributed to the incorporation of PB/MWNTs. The biosensor exhibited a linear response to glucose in the concentration range from 2.5×10−5 to 1.3×10−3M with a correlation coefficient of 0.9998, a detection limit of 7.5×10−6M, a low response time (10s), good sensitivity and high anti-interference ability. Compared with the control biosensor based on the traditional tetraethoxysilane derived sol–gel composite film, the biosensor showed a similarly small apparent Michaelis–Menten constant of 3.67mM but much higher electrochemical and biosensing stability.
Ti3C2Tx MXene/Graphene/AuNPs 3D porous composites for high sensitivity and fast response glucose biosensing

Abstract:Glucose oxidase (GOx) is one of the typical enzymes widely used in glucose sensors. In recent years, novel nanomaterials have been developed to immobilize the enzyme, which is of great importance for the development of biosensors. Here we constructed 3D porous Ti3C2Tx MXene/Graphene/AuNPs (MGA) composites by a simple mixing-freeze-drying and self-reduction process. The abundant hydrophilic groups on the surface of Ti3C2Tx MXene nanosheets endow the MGA composites with highly hydrophilic microenvironment. The 3D porous composites provide a more open structure and promote the entry of GOx into the internal pores, which may enhance the stable immobilization and retention of GOx in the film. The unique electrocatalytic properties and synergistic effects between AuNPs and Ti3C2Tx MXene/Graphene are used to further enhance the electron transport in the 3D porous composites and reduce the redox potential. The MGA/GOx glucose sensor shows linear current in the range of glucose concentration from 2 μM to 0.4 mM and 0.4 mM to 6 mM, with sensitivities of 169.49 μA/(mM·cm2) and 34.13 μA/(mM·cm2) respectively, showing high sensitivity at low concentrations. The detection limit of glucose sensor reaches 2 μM and the response time is less than 3 s. This sensor can be applied to the detection of glucose in human sweat.

Abstract:Catalytic cascades derived from the combination of multiple enzymes with efficient catalytic activity and high selectivity have attracted widespread attention in biosensing and biomedical applications . A key consideration in the development of cascade reaction systems is the combination of multiple enzymes to improve overall catalytic performance. Inspired by natural protein self-assembly, herein, proteinosomes formed by enzyme-surfactant complexes based on an interfacial assembly strategy are developed and they exhibit significant enhanced activity via cascade reaction of glucose oxidase (GOX) and horseradish peroxidase (HRP) that are located in proximity. The proteinosomes not only exhibit significantly elevated stability and reusability , but also possess enhanced cascade catalytic performance. These enhanced activities can be explained on the basis of the proximity effect of multiple enzymes within proteinosomes. Moreover, we employed highly efficient and reusability dual-enzyme-based proteinosomes to demonstrate the ultrasensitive detection of glucose, achieving a detection limit as low as 1.08 μM. Finally, the proteinosomes can be immobilized in a test strip for easy and fast glucose detection. Due to their ultra-sensitive detection limit, they can be used for non-invasive glucose detection. They can accurately assess glucose levels in saliva, yielding results highly congruent with those from commercially available assay kits. We further indicate the rapid capability of our system to detect glucose, as compared to commercial glucose kit. These finds demonstrate the potential of the test strip as a promising tool for daily glucose monitoring, and the facile approach presents an inventive solution for test strip development.
